An improved in vivo body weight gain bioassay for the potency determination of human growth hormo... more An improved in vivo body weight gain bioassay for the potency determination of human growth hormone (hGH) has been set up in "little" mice (lit/lit), a mutant derived from the C57BU6J strain. This improved assay now has a detection limit of the order of 0.05 jJg/mouse/day, which corresponds to a sensitivity about 20-fold higher than that of the most sensitive in vivo assay reported up to now: the tibia test in hypophysectomized rats or mice. This sensitivity was achieved mainly by introduction of a careful pre-assay selection and of a three injections per day schedule. The utilization of these conditions in a 2x2 factorial assay design allowed the potency determination of recombinant DNA-derived hGH (rec-hGH) in bacterial extracts with acceptable accuracy and precision, together with the greatest economy of material,

Different gamma ray doses were applied and microbiological loads of irradiated subsamples and a non-irradiated control subsample were studied. A 0.90 kGy decimal reduction value was estimated at the microbiological load test conditions and it was concluded that a 5 kGy dose is suitable for the sterilization of the mussel reference material.

Chitosan is a biomaterial with diverse biological properties that allows the development of a biodressings with satisfactory qualities. Due to the abundance, easy to obtain and antimicrobial activity, we used the crosslinked chitosan with sodium tripolyphosphate (TPP Na5P3O10), non-toxic component that offers resistance by ionic interactions between the compounds. The study aimed to evaluate a dressing with the auxiliary function in the repair and regeneration skin. Studies were performed with SEM, FT-IR and direct and indirect cytotoxicity. For the direct and indirect cytotoxicity each sample was cut and sterilized by exposure to UV lamp laminar flow for 15 minutes on each side. Then, for the indirect cytotoxicity, samples were separately placed into sterile tubes for extraction of possible toxic substances in dressings. Extracts were filte...

The new outbreak of coronavirus disease 2019 (COVID-19) has infected and caused the death of mill... more The new outbreak of coronavirus disease 2019 (COVID-19) has infected and caused the death of millions of people worldwide. Intensive efforts are underway around the world to establish effective treatments. Immunoglobulin from immunized animals or plasma from convalescent patients might constitute a specific treatment to guarantee the neutralization of the virus in the early stages of infection, especially in patients with risk factors and a high probability of progressing to severe disease. Worldwide, a few clinical trials using anti-SARS-CoV-2 immunoglobulins from horses immunized with the entire spike protein or fragments of it in the treatment of patients with COVID-19 are underway. Here, we describe the development of an anti-SARS-CoV-2 equine F(ab′)2 immunoglobulin using a newly developed SARS-CoV-2 viral antigen that was purified and inactivated by radiation. Cell-based and preclinical assays showed that the F(ab′)2 immunoglobulin successfully neutralizes the virus, is safe in...

Total testosterone concentration fell progressively during the menstrual cycle, whereas the percent free testosterone increased from the follicular to the luteal phase. Free testosterone levels also fell but only significantly at the late luteal phase. For comparison two patients with anovulatory cycles were evaluated. Progesterone displacement of endogenous testosterone from its binding protein(s) was suggested by in vitro studies.

Depending on the results of the analyses, the tissues are transferred to the room designated for materials liberated for use. The maximum cryopreservation period is 5 years for bone tissue and 2 years for soft or tendinous tissues.

Animals were divided into transplanted with irradiated human skin grafts (IHSG) at 25 and 50 kGy (IHSG 25 kGy; IHSG 50 kGy) and non-IHSG and euthanized on the 3rd, 7th and 21st days after the surgery, by gross and microscopic changes, immunostaining for human type I collagen (Col I) and mouse Col I and Col III and inflammatory cells. We found an effectiveness of human split-thickness graft adherence in mice transplanted with IHSG 25 kGy, as well decrease in dermo-epidermal necrosis and neutrophils, lower loss of skin thickness, epithelization and neo-vascularization. Day 21 post-transplantation with IHSG 25 kGy was observed a well-preserved human skin in the border of the graft, a prominent granulation tissue in an organization by proliferated fibroblasts, Col III deposition and increased B-cells and macrophages.
