Single-chain fusion proteins comprised of GM-CSF and a myelin neuroantigen (NAg) epitope are pote... more Single-chain fusion proteins comprised of GM-CSF and a myelin neuroantigen (NAg) epitope are potent, NAg-specific inhibitors of rat and mouse models of experimental autoimmune encephalomyelitis (EAE). This study provided evidence that GMCSF-MOG (myelin oligodendrocyte glycoprotein 35-55 peptide) inhibited EAE when co-injected adjacent to the encephalitogenic MOG35-55/ complete Freund’s adjuvant (CFA) emulsion. Furthermore, GMCSF-MOG had tolerogenic activity in the C57BL/6 model of chronic EAE when directly incorporated with MOG35-55 in the CFA emulsion. Likewise, GMCSF-PLP (proteolipid protein 139-151 peptide) inhibited relapsing-remitting EAE in SJL mice when mixed with PLP139-151 in the CFA emulsion. In vitro studies revealed that GMCSF-MOG stimulated the growth and differentiation of inflammatory dendritic cells (DC) and targeted the MOG35-55 domain for potentiated antigen presentation. Glaucoma is the second driving reason for partial or complete blindness among all the visual defi... more Glaucoma is the second driving reason for partial or complete blindness among all the visual deficiencies which mainly occurs because of excessive pressure in the eye due to anxiety or depression which damages the optic nerve and creates complications in vision. Traditional glaucoma screening is a timeconsuming process that necessitates the medical professionals' constant attention, and even so time to time due to the time constrains and pressure they fail to classify correctly that leads to wrong treatment. Numerous efforts have been made to automate the entire glaucoma classification procedure however, these existing models in general have a black box characteristics that prevents users from understanding the key reasons behind the prediction and thus medical practitioners generally can not rely on these system. In this article after comparing with various pre-trained models, we propose a transfer learning model that is able to classify Glaucoma with 94.71% accuracy. In addition, we have utilized Local Interpretable Model-Agnostic Explanations(LIME) that introduces explainability in our system. This improvement enables medical professionals obtain important and comprehensive information that aid them in making judgments. It also lessen the opacity and fragility of the traditional deep learning models.

The paper set out to examine; the public financial management reforms and pronouncements introduced in the public sector; how effective these reforms and pronouncements have been; the determinants of efficient public financial management system; and the ways out of the current economic crises in the Nigerian public sector. The paper is established on agency theory and also employed the desk/analytical review research design which relied wholly on reservoir and pool of knowledge, extant literature and research findings from previous scholars. From the findings made, the study concluded that there had been various efforts in areas of public sector financial management reforms put in place by the Nigeria government in her bid to introduce accountability, good service delivery; efficient public service output, corrupt free society and effective financial reporting in order improve the life of Nigeria citizens. The PFM reforms have also met stiff resistance in some areas. If given enough time to improve and properly correct her PFM, the Nigeria public sector would be an example for other country. Therefore, it was recommended that, there is the need for the public institutions that are saddled with the responsibility of motoring and enforcing PFM reforms to work as a system. System has subsystems; they should complement each other.

This raises some serious concerns because; ground excavation during construction can have significant impact on structures at close proximity. This study performs three-dimensional FEM analysis in Plaxis 3D to observe the effects of ground excavation works near a shallow isolated footing in different types of soils and at various distances from the excavation pit. From the analysis, the maximum foundation settlement caused by excavation works in soft clay, medium dense sand, and stiff sandy clay was found at 1.5 m, 3 m & 5 m distances from the pit of excavation correspondingly. Foundation settlement caused by excavation works on medium dense sand and stiff sandy clay was moderate but abrupt & unsafe on soft clay soil under the given circumstances.

This paper presents the results of a field implementation of microbial induced calcite precipitat... more This paper presents the results of a field implementation of microbial induced calcite precipitation to stabilize expansive soils in Marsing, Idaho. The field test was done by drilling 2.5" (6.35 cm) diameter holes at a spacing of 16" to 30" (40.6 cm to 76.2 cm) into the ground and, injecting bio-enrichment followed by bio-cementation solutions to stimulate the native bacteria and subsequently achieve calcite precipitation. The pH level of the soil, the calcite content and free swelling potential were monitored over time by collecting periodic soil samples from the injection points. An increase in pH from 8.3 to 9.7 was seen in the first seven days after the injection of bio-enrichment solution. The calcite content in the soil increased and the free swelling potential decreased consistently with each subsequent injection of bio-cementation solution. The calcite content increased from 3% to 8% and the free swell index dropped from 114% to 29%. The results show that microbial induced calcite precipitation can be successfully replicated in the field for the stabilization of expansive soils.

For LCC mapping of the vegetated Asahi River in Japan, the current study uses deep learning (DL)-based DeepLabV3+ module for image segmentation of aerial photographs. We modified the existing model by concatenating data on its resultant output port to access the airborne laser bathymetry (ALB) dataset, including voxel-based laser points and vegetation height (i.e. digital surface model data minus digital terrain model data). Findings revealed that the modified approach improved the accuracy of LCC greatly compared to our earlier unsupervised ALB-based method, with 25 and 35% improvement, respectively, in overall accuracy and the macro F1-score for November 2017 dataset (no–leaf condition). Usually, people clean their wall of outer surface of building by human. Often it becomes risky and costly also. A portable robot having the ability to climb on vertical surface has been expected for a long time. To protect human's life we designed a prototype of vertical surface climbing robot for cleaning and other multipurpose operation. Some combination of technology has been shown such as smart wireless control with all direction moving capability. Four Dc motors are used to move the robot and one Dc motor is used to rotate the cleaning brush. There is one Electric Ducted fan (EDF) which helps the robot to stick to the surface against gravitational force. An Arduino uno is used as a controller for different There is a Bluetooth modular which has been used to connect with smart phone and take command from user. Four motor helps it to move in up, down, right and left direction by the according commands given by smart phone. The full robot is power by using DC battery and AC to DC converted adapter combined. To observe the outcome of the project, we analyzed the performance of the robot by climbing in different surfaces with different nature. The climbing and stick to the surface differ because of grip. University students, who are supposed to be the future of a country, are also accepting this path of deaths. Bangladeshi undergraduate students are also becoming the victims of suicide every day. In recent times, suicide ideation among the university undergraduate students is increasing. This article reveals the prevalence of suicide among the university undergraduate students and the factors associated with suicide ideation among the university undergraduate students of Bangladesh. It also focuses on the impact of these suicides on the family and society. To overcome this grave situation, some precautionary steps have been recommended at the end of this article. It is of extreme importance to investigate the DNA repair pathways that will lead to a better und... more It is of extreme importance to investigate the DNA repair pathways that will lead to a better understanding to develop cancer treatments since the primary cause of cancer is the inability of the cell to restore the damaged DNA. Upon DNA damage, the histone tails of the nucleosomes near the DNA lesions undergo a group of extensive post-translational modifications that remodels the chromatin structures in order to repair the damage. DNA double-strand break (DSB) induces the phosphorylation of histone variant H2A.X at Serine 139 residue, that is recognized by the mediator of DNA damage checkpoint, MDC1. This interaction is imperative for the recruitment of the other downstream DNA damage response (DDR) proteins that enact in selecting the repair pathways. Structural basis of interaction of the MDC1 BRCT domain with the phospho-peptide tail of H2A.X was previously revealed. However, the impacts of this interaction on the overall chromatin structure, and the successive nucleosome modifications are still elusive. We hypothesize that MDC1 BRCT domain not only binds to the phosphorylated H2A.X tail, but this interaction might position MDC1 in a way that its basic patch might interact with the nucleosomal DNA, or with the conserved acidic patch of H2A.X/H2B. To investigate these concepts in vitro, a successful purification of the nucleosome containing γH2A.X is the prerequisite. For decades, histones have been extracted from the cell lysate using urea denaturant with subsequent extensive dialysis to refold the heteromeric histone complexes. This method is time and cost intensive. Here, we have developed two rapid purification protocols using the co-expression systems for the recombinant human histone oligomers. Furthermore, we have established a system for the in vitro phosphorylation of H2A.X that will facilitate the structural analysis of MDC1 BRCT bound γH2A.X nucleosome complex in the future.

To obtain an axenic seed culture, it is important to perform an adequate a disinfection procedure in tissue culture. Four nutrient media viz. MS, 1⁄2MS, KC and VW were evaluated on In vitro seed germination with callus initiation. The maximum seed germination with callus initiation (100%) was recorded in MS basal medium with a short span of time (40 days after culture). After 45 days of callus initiation the effect of eight different treatments (T1-T8) on callus size and nature were also studied. The experiment revealed that in T3 (MS + 2.0 mgl BAP + 0.5 mgl NAA) was found to be the best for callus development (1.98 cm length and 1.01 cm breadth). More than one million forcefully displaced Myanmar nationals, known as Rohingyas, are currently l... more More than one million forcefully displaced Myanmar nationals, known as Rohingyas, are currently living in the 34 camps in the Cox's Bazar district in Bangladesh. To reduce the pressure of Rohingyas in Cox's Bazar, the government of Bangladesh has started the process of relocating some of the Rohingyas to Bhasan Char Island located in Noakhali district, Bangladesh. However, some international organisations, NGOs, and some people expressed their concerns over this relocation process. This article, thus, offers an evidence-based holistic perspective that explains the available facilities and opportunities that could bring a new hope instead of a threat to Rohingyas. Through a multi-method research approach, the paper argues that the Island is suitable for the displaced Rohingya people. It also explores that the relocation project has opportunities for Rohingyas to engage in some livelihood activities. The Island is stable and livable on a temporary basis and has adequate safety and security measures-embankments, wave breakers, shelter centers, and modern housing facilities-have been undertaken. This article, thus, concludes that Bhasan Char can offer a new hope-not a place with threats, for the Rohingya displaced people, to live temporarily before their repatriation to Myanmar.

Indiscriminate use of pesticides in agricultural land poses a potential threat to many non-target... more Indiscriminate use of pesticides in agricultural land poses a potential threat to many non-target organisms, including fish. In the present study, we explored the toxicological effects of sumithion on Nile tilapia (Oreochromis niloticus) after exposure at different concentrations (0.0, 1.5, 2.0, 2.5, 3.0, 3.5 and 4.0 mg/L) in triplicate for 96 h. The 96-h LC50 value of the pesticide for the test fish was calculated by probit analysis, which was 2.579 mg/L. We also investigated the haematological parameters, erythrocytic alteration and histopathological responses of Nile tilapia. Red blood cell (RBC) and haemoglobin (Hb) level were significantly declined, whereas white blood cell (WBC) and blood glucose level were increased dramatically in the sumithion treated fish. The frequencies of erythrocytic cellular and nuclear alterations were significantly elevated in the fish after exposure to sumithion compared with the control group. The substantial histopathological alterations were observed in sumithion-exposed fish, including pyknotic nucleus, melano-macrophage centers and severe congestion of blood vessels in the liver and patch degeneration, vacuolation and intense form of pyknotic nuclei in the kidney. Observed alterations in this study exhibit that sumithion negatively impacts on Nile tilapia. It is recommended that the use of this pesticide in the agriculture field be monitored and controlled.

This paper presents the results of a field implementation of microbial induced calcite precipitat... more This paper presents the results of a field implementation of microbial induced calcite precipitation to stabilize expansive soils in Marsing, Idaho. The field test was done by drilling 2.5" (6.35 cm) diameter holes at a spacing of 16" to 30" (40.6 cm to 76.2 cm) into the ground and, injecting bio-enrichment followed by bio-cementation solutions to stimulate the native bacteria and subsequently achieve calcite precipitation. The pH level of the soil, the calcite content and free swelling potential were monitored over time by collecting periodic soil samples from the injection points. An increase in pH from 8.3 to 9.7 was seen in the first seven days after the injection of bio-enrichment solution. The calcite content in the soil increased and the free swelling potential decreased consistently with each subsequent injection of bio-cementation solution. The calcite content increased from 3% to 8% and the free swell index dropped from 114% to 29%. The results show that microbial induced calcite precipitation can be successfully replicated in the field for the stabilization of expansive soils.

For LCC mapping of the vegetated Asahi River in Japan, the current study uses deep learning (DL)-based DeepLabV3+ module for image segmentation of aerial photographs. We modified the existing model by concatenating data on its resultant output port to access the airborne laser bathymetry (ALB) dataset, including voxel-based laser points and vegetation height (i.e. digital surface model data minus digital terrain model data). Findings revealed that the modified approach improved the accuracy of LCC greatly compared to our earlier unsupervised ALB-based method, with 25 and 35% improvement, respectively, in overall accuracy and the macro F1-score for November 2017 dataset (no–leaf condition). A portable robot having the ability to climb on vertical surface has been expected for a long time. To protect human's life we designed a prototype of vertical surface climbing robot for cleaning and other multipurpose operation. Some combination of technology has been shown such as smart wireless control with all direction moving capability. Four Dc motors are used to move the robot and one Dc motor is used to rotate the cleaning brush. There is one Electric Ducted fan (EDF) which helps the robot to stick to the surface against gravitational force. An Arduino uno is used as a controller for different There is a Bluetooth modular which has been used to connect with smart phone and take command from user. Four motor helps it to move in up, down, right and left direction by the according commands given by smart phone. The full robot is power by using DC battery and AC to DC converted adapter combined. It is of extreme importance to investigate the DNA repair pathways that will lead to a better und... more It is of extreme importance to investigate the DNA repair pathways that will lead to a better understanding to develop cancer treatments since the primary cause of cancer is the inability of the cell to restore the damaged DNA. Upon DNA damage, the histone tails of the nucleosomes near the DNA lesions undergo a group of extensive post-translational modifications that remodels the chromatin structures in order to repair the damage. DNA double-strand break (DSB) induces the phosphorylation of histone variant H2A.X at Serine 139 residue, that is recognized by the mediator of DNA damage checkpoint, MDC1. This interaction is imperative for the recruitment of the other downstream DNA damage response (DDR) proteins that enact in selecting the repair pathways. Structural basis of interaction of the MDC1 BRCT domain with the phospho-peptide tail of H2A.X was previously revealed. However, the impacts of this interaction on the overall chromatin structure, and the successive nucleosome modifications are still elusive. We hypothesize that MDC1 BRCT domain not only binds to the phosphorylated H2A.X tail, but this interaction might position MDC1 in a way that its basic patch might interact with the nucleosomal DNA, or with the conserved acidic patch of H2A.X/H2B. To investigate these concepts in vitro, a successful purification of the nucleosome containing γH2A.X is the prerequisite. For decades, histones have been extracted from the cell lysate using urea denaturant with subsequent extensive dialysis to refold the heteromeric histone complexes. This method is time and cost intensive. Here, we have developed two rapid purification protocols using the co-expression systems for the recombinant human histone oligomers. Furthermore, we have established a system for the in vitro phosphorylation of H2A.X that will facilitate the structural analysis of MDC1 BRCT bound γH2A.X nucleosome complex in the future.

MS, 1⁄2MS, KC and VW were evaluated on In vitro seed germination with callus initiation. The maximum seed germination with callus initiation (100%) was recorded in MS basal medium with a short span of time (40 days after culture). More than one million forcefully displaced Myanmar nationals, known as Rohingyas, are currently l... more More than one million forcefully displaced Myanmar nationals, known as Rohingyas, are currently living in the 34 camps in the Cox's Bazar district in Bangladesh. To reduce the pressure of Rohingyas in Cox's Bazar, the government of Bangladesh has started the process of relocating some of the Rohingyas to Bhasan Char Island located in Noakhali district, Bangladesh. However, some international organisations, NGOs, and some people expressed their concerns over this relocation process. This article, thus, offers an evidence-based holistic perspective that explains the available facilities and opportunities that could bring a new hope instead of a threat to Rohingyas. Through a multi-method research approach, the paper argues that the Island is suitable for the displaced Rohingya people. It also explores that the relocation project has opportunities for Rohingyas to engage in some livelihood activities. The Island is stable and livable on a temporary basis and has adequate safety and security measures-embankments, wave breakers, shelter centers, and modern housing facilities-have been undertaken. This article, thus, concludes that Bhasan Char can offer a new hope-not a place with threats, for the Rohingya displaced people, to live temporarily before their repatriation to Myanmar.

Indiscriminate use of pesticides in agricultural land poses a potential threat to many non-target... more Indiscriminate use of pesticides in agricultural land poses a potential threat to many non-target organisms, including fish. In the present study, we explored the toxicological effects of sumithion on Nile tilapia (Oreochromis niloticus) after exposure at different concentrations (0.0, 1.5, 2.0, 2.5, 3.0, 3.5 and 4.0 mg/L) in triplicate for 96 h. The 96-h LC50 value of the pesticide for the test fish was calculated by probit analysis, which was 2.579 mg/L. We also investigated the haematological parameters, erythrocytic alteration and histopathological responses of Nile tilapia. Red blood cell (RBC) and haemoglobin (Hb) level were significantly declined, whereas white blood cell (WBC) and blood glucose level were increased dramatically in the sumithion treated fish. The frequencies of erythrocytic cellular and nuclear alterations were significantly elevated in the fish after exposure to sumithion compared with the control group. The substantial histopathological alterations were observed in sumithion-exposed fish, including pyknotic nucleus, melano-macrophage centers and severe congestion of blood vessels in the liver and patch degeneration, vacuolation and intense form of pyknotic nuclei in the kidney. Observed alterations in this study exhibit that sumithion negatively impacts on Nile tilapia. It is recommended that the use of this pesticide in the agriculture field be monitored and controlled.
