Research paper thumbnail of Preparation of Laminari-oligosaccharides : by Acetolysis of Curdlan

A simple method for preparation of homologous series of laminari-oligosaccharides was presented. ... more A simple method for preparation of homologous series of laminari-oligosaccharides was presented. Curdlan, a kind of (1→ 3 )-s-D-glucan, was subjected to direct acetolysis. followed by saponification to give a mixtur巴 oflaminari-oligosaccarides. Acetolysis for 1 hr at 600C was found to be an optimum for the preparation of a mixture of laminari-oligosaccharides having degree of polymerization 2 to 7. Each oligosaccharide was separated by a size exclusion chromatography on Toyopearl HW 40 S and its properties were characterized.

A major polysaccharide obtained by microwave irradiation in water (140 C, 5 min) was (1 3;1 6)-β-D glucan rich in (1 3) linkages, whereas polysaccharides obtained by hot water extraction using conventional external heating (100 C, 6 h) were fucogalactan and (1 3;1 6)-β-D glucan rich in (1 6) linkages. In the case of microwave irradiation in water, fucogalactan was suggested to be depolymerized during heating. Microwave heating in water has an advantage over the conventional external heating for extraction of β-glucan rich in (1 3) linkages from the fruiting body of H. erinaceum.
