Fine Structure Analysis of the Gene Region for N2-Fixation (nif) of Klebsiella pneumoniae (original) (raw)

Abstract

Klebsiella pneumoniae, a free-living N2-fixing organism, carries the genes for nitrogen fixation (nif) in a cluster on the chromosome (Streicher et al., 1972). By genetic engineering techniques in vivo this chromosomal nif region was transferred to Escherichia coli (Dixon and Postgate, 1972) integrated into the chromosome (Cannon et al., 1974) combined with the E. coli fertility plasmid F (Cannon et al., 1976) and subsequently with the resistance plasmid RP4 (Dixon et al., 1976). Plasmid pRD1 consisting of RP4 and the his nif region of K. pneumoniae is the most important nif plasmid constructed in vivo: Due to the broad host range of RP4 it can be transferred to a large variety of gram negative bacteria (Cannon and Postgate, 1976; Postgate and Krishnapillai, 1977).
