Summary

A simple procedure is described for the quantitative evaluation of thin-layer chromatograms of condensed phosphates. By dipping the developed chromatogram into a 6% solution of collodium which is mixed with about 7% glycerol, the layer of sorption material will be imbeded in a kind of foil. It may be lift off from the thin layer without residues. So it is possible to carry out further analysis like that in quantitative paper chromatography: Cutting out the spots, destroying the organic medium in wet state and estimation of the content of phosphates in the single spots by colorimetry.

A sorption material on the basis of cellulose powder is described which leads without adhesive additives to layers on which by means of a modified methanol/dioxan solvent mixture also medium-chain phosphates may be estimated.

Comparisons of thin-layer chromatography and paper chromatography of different condensed phosphates are discussed on the basis of tables. The questions of the quantities of phosphates for thin-layer chromatography and the sensibility of the method are examined. Also the destruction of the organic material in liquid solution is discussed.

Finally a detailed instruction is given for carrying out the quantitative thin-layer chromatography of condensed phosphates.

Zusammenfassung

Es wird ein Verfahren beschrieben, das die quantitative Auswertung von Dünnschicht-Chromatogrammen kondensierter Phosphate ermöglicht. Durch Eintauchen der entwickelten Chromatogramme in eine 6%ige Kollodiumlösung, die mit ∼ 7% Glycerin versetzt ist, wird die Sorptions-schicht in eine Art Folie eingebettet. Sie kann dann rückstandfrei von der DC-Trägerplatte abgehoben werden. Damit ist es möglich, den weiteren Analysengang wie bei der quantitativen Papier-Chromatographie durch-zuführen: Aufteilung des Chromatogramms in Zonen, Naßveraschen der organischen Phase und colorimetrische Bestimmung des in den einzelnen Zonen enthaltenen Phosphatgehaltes.

Ein Sorptionsmittel auf Cellulosebasis, das ohne haftverbessernde Zusätze Sorptionsschichten ergibt, auf denen mit einem modifizierten Methanol/-Dioxan-Fließmittel auch mittelkettige Phosphate getrennt und quantitativ bestimmt werden können, wird mitgeteilt.

Vergleiche der Dünnschicht-Chromatographie und der Papier-Chromatographie von verschiedenen kondensierten Phosphaten werden an Hand von Tabellen besprochen. Es werden die Fragen der bei der Dünnschicht-Chromatographie zur Analyse kommenden Phosphatmengen sowie die Empfindlichkeit der Methode untersucht und die Naßveraschung der Sorptionsschicht in Verbindung mit der Kollodiumfolie behandelt. Zum Schluß wird noch eine ausführliche Arbeitsvorschrift für die quantitative Dünnschicht-Chromatographie kondensierter Phosphate gegeben.
