This study evaluates the effectiveness of organic anion and cation transport inhibitors and substrate on the tumor uptake of 99mTc-HYNIC-(Ser)3-LTVPWY radiotracer in SKOV-3 tumor-bearing nude mice. Methods: Before the injection of the radiolabeled peptide, SKOV-3 tumor-bearing nude mice were treated with furosemide, cimetidine, para-amino hippuric acid, and saline. The inhibition effects of the organic anion and cation transport inhibitors were compared with the control group. In both treatment and control groups, the tumor and renal accumulation of radiopeptide in mice bearing SKOV-3 tumors were assessed in biodistribution and SPECT imaging studies. Results: The biodistribution and imaging results suggested that all treated groups showed a higher tumor and higher normal tissue radioactivity compared to the control group. According to the tumor imaging study, the furosemidetreated group had slightly better tumor uptake and a higher tumor to muscle uptake ratio than other treatment groups. Conclusions: Administration of furosemide (an OAT inhibitor) increased radioactivity accumulation in the kidneys and blood and improved tumor radioactivity uptake. PAH (an anion transporter substrate) and cimetidine (an OCT inhibitor) have a minor effect on the accumulation of radioactivity in the kidneys and the acquired images.

The 5-HT 7 R is the newest member of this family and contributes to different physiological and pathological processes. As a pathology, Glioblastoma multiform (GBM) overexpresses 5-HT 7 R; hence, this study aims to develop radiolabeled aryl piperazine derivatives as 5-HT 7 R imaging agents. To that end, compounds 6 and 7 as 1-(3-nitropyridin-2-yl)piperazine derivatives were radiolabeled with fac-[ 99m Tc(CO) 3 (H 2 O) 3 ] + and (99m Tc(CO) 3-[6] and 99m Tc(CO)3-[7]) were obtained with high radiochemical purity (RCP>94%). 99m Tc(CO) 3-[6] and 99m Tc(CO)3-[7] possess high a nity toward 5-HT 7 R of the U-87 MG cell line. The calculated K i for them was 26.85±0.78 nM and 36.67±0.89 nM. The biodistribution study in normal mice indicates that the brain uptake of 99m Tc(CO) 3-[6] and 99m Tc(CO)3-[7] is the highest at 30 min postinjection (0.8±0.25 and 0.64±0.18 %ID/g, respectively). The data of the biodistribution study in the U87-MG xenograft model revealed that these radiotracers could accumulate in the tumor site, and the highest tumor uptake was observed at 60 min post-injection (3.38±0.65 and 3.27±0.5 %ID/g, respectively). The injection of pimozide can block the tumor's radiotracer uptake, indicating the binding of these radiotracers to the 5-HT 7 R. The imaging study in the xenograft model also con rms the biodistribution data. The acquired images clearly show the tumor site, and the tumor-to-muscle ratio for 99m Tc(CO) 3-[6] and 99m Tc(CO)3-[7] at 60 min was 3.33 and 3.88, respectively.

The use of compartmental analysis allows a mathematical separation of tissues and organs to determine the concentration of activity in each fraction of interest. Biodistribution studies are expensive and difficult to carry out in humans, but such data can be obtained easily in rodents and rat. Methods- We have developed a physiologically based pharmacokinetic model for scaling up activity concentration in each organ versus time. The mathematical model uses physiological parameters including organ volumes, blood flow rates, and vascular permeability. The compartments (organs) are connected anatomically. This allows the use of scale-up techniques to predict the new complex distribution in humans in each organ. Results- The concentration of the new comp...

Methods: A total of 80 patients with breast cancer (invasive ductal carcinoma) were examined with planar whole body bone scan and SPECT imaging using 99mTc-labelled methylene diphosphonate (99mTc-MDP). The patients with abnormal uptakes in SPECT imaging were also investigated with magnetic resonance imaging (MRI). Results: Among these 80 patients with normal whole body bone SPECT scan, 19 (23.25%) of them revealed abnormal 99mTc-MDP uptake in skeleton. Furthermore, these 19 patients were subjected to MRI and 3 (3.75%) of them were confirmed with metastatic bone lesion. Conclusion: The obtained data suggest that SPECT possess the added diagnostic over planar whole body bone scan.

Introducing the dose level from diagnostic procedures can be effective in reducing the patient dose, improving the quality of treatment, and increasing the safety of patients and staff. This study evaluated patient entrance surface dose (ESD), effective dose, and cumulative dose from most common radiography procedures. Materials and methods: A descriptive study was conducted in which patient dose level from most common radiography procedures in two referral hospitals (Imam Khomeini and Bu-Ali Sina, Sari, Iran) were evaluated in 2015. X-ray tube output was measured in 1m from the tube focal spot in several kVp. ESD (mGy) value in every procedure was calculated while considering its radiographic conditions (kV, mAs and FFD). The effective dose (ED) (mSv) per examination was also calculated from appropriate ED/ESD conversion factor and the cumulative dose (man-sivert) of ann...

Research paper thumbnail of Itraconazole synergistically increases therapeutic effect of paclitaxel and 99mTc-MIBI accumulation, as a probe of P-gp activity, in HT-29 tumor-bearing nude mice

We investigated the efficacy of itraconazole as a P-gp inhibitor and its therapeutic synergistic relationship to paclitaxel through 99mTc-MIBI accumulation in HT-29 tumor-bearing nude mice. Histopathological screening along with in vitro experiments was done for further assessment. Itraconazole successfully inhibited P-gp mediated 99mTc-MIBI efflux, increasing its in vitro accumulation in itraconazole-receiving dishes. Notably, the co-administration of itraconazole with paclitaxel significantly enhanced the in vitro cytotoxicity effect of paclitaxel in itraconazole + paclitaxel wells containing HT-29 cells. Compared to the control, tumor volume in mice treated with itraconazole, paclitaxel and itraconazole +paclitaxel showed growth suppression approximately by 36.21, 60.02, and 73.3% respectively. And compared to paclitaxel group, the nude mice co-treated with paclitaxel and...

Research paper thumbnail of Characterization of low, medium and high energy collimators for common isotopes in nuclear medicine: A Monte Carlo study

However, some photons pass the septa without interaction or experience scattering before they reach the detector. In this study, we determined different contribution of collimator responses consist of geometrical response, septal penetration (SP) and scattering (SC) for low, medium and high energy collimators. Methods: A point source of activity with common energies in diagnostic nuclear medicine and three different collimators were simulated using SIMIND Monte Carlo code. Results: For Low Energy High Resolution (LEHR) collimator, SP was increased from 7% in 140 keV to 30% in 167keV and more than 75% in energies higher than 296keV. SC also was increased from 4% in 98keV to more than 15% in energies higher than 167keV and reached to its maximum (26%) in 296keV. For Medium Energy All Purpose (MEAP) collimator, SP was suddenly increased from 6%...
