Quantification of chemical residues in honey is a market requirement to ensure consumer safety. T... more Quantification of chemical residues in honey is a market requirement to ensure consumer safety. The most common method used to analyze these compounds is the LC/MS/MS methodology, which requires highly qualified technicians and a tedious pre-treatment of the sample. The honey-packaging industry needs cheaper and faster alternatives for routine control. HFF-QCM (High Fundamental Frequency Quartz Crystal Microbalance) sensors are becoming a good option due to their high sensitivity, fast detection and low cost, while avoiding complex sample pre-treatment. The HFF-QCM technology is based on piezoelectric sensors with frequencies in the range from several tenths of MHz to hundreds of MHz. In this work a 100 MHz HFF-QCM sensor was used in a monoclonal antibody-based competitive immunoassay for specific biorecognition of carbaryl pesticide as testing contaminant. The work intends to validate the use of HFF-QCM technology, in comparison with liquid chromatography-tandem mass spectrometry (LC-MS/MS) technique, for the detection of contaminants in honey. For this purpose, the validation criteria required by SANCO 12571/2013 guidance document were considered. The precision and accuracy (recovery) of both methods were determined by comparison of 5 replicates at 4 different concentrations (from 0 to 100 μg/kg) using the same honey matrix. HFF-QCM technology showed good accuracy, with recovery percentages always between 110 and 120%. As regards to precision, HFF-QCM coefficients of variation (CV) were around 10% higher than those recommended by GC SANCO 12571/2013. HFF-QCM limits of detection ACCEPTED MANUSCRIPT (LOD) and quantification (LOQ) were in the same order of magnitude as those for LC-MS/MS, which allows the analysis of carbaryl residues in honey under the established maximum residue limits (MRL), without sample pre-treatment. These results show that biosensors based on HFF-QCM technology has become a serious alternative to the traditional analytical techniques for food quality and safety applications.

Tuberculosis, one of the oldest diseases affecting human beings, is still considered as a world p... more Tuberculosis, one of the oldest diseases affecting human beings, is still considered as a world public health problem by the World Health Organization. Therefore, there is a need for new and more powerful analytical methods for early illness diagnosis. With this idea in mind, the development of a High Fundamental Frequency (HFF) piezoelectric immunosensor for the sensitive detection of tuberculosis was undertaken. A 38 kDa protein secreted by Mycobacterium tuberculosis was first selected as the target biomarker. Then, specific monoclonal antibodies (MAbs) were obtained. Myc-31 MAb, which showed the highest affinity to the analyte, was employed to set up a reference enzyme-linked immunosorbent assay (ELISA) with a limit of detection of 14 ng mL-1 of 38 kDa antigen. In this work, the design, characterization, and validation of a 150 MHz high fundamental frequency quartz crystal microbalance (HFF-QCM) sensor for bio-sensing applications are introduced. Finite element method (FEM) simulations of the proposed design are in good agreement with the electrical characterization of the manufactured resonators. The sensor is also validated for bio-sensing applications. Using this circuit, a low-cost method for a simple determination of equivalent circuit parameters of quartz crystal resonators is described. Measurements made with the proposed system on typical AT cut quartz crystals are in good agreement with those of an Impedance Analyzer.

An extended Butterworth-Van Dyke (EBVD) model with frequency-independent parameters for the chara... more An extended Butterworth-Van Dyke (EBVD) model with frequency-independent parameters for the characterization of a resonant compound formed by a quartz crystal in contact with a finite viscoelastic layer contacting a semi-infinite viscoelastic medium is extracted by analysis of the lumped element model. The formulation of the EBVD model is compared with the complete expression of the electrical admittance of the loaded quartz derived from the transmission line model (TLM). Relative deviations between them do not exceed 3% around 1% bandwidth near resonance. An extended Martin & Granstaff's model and an explicit expression for the frequency shift that supposes an extension of Kanazawa's model for viscoelastic media are obtained. An analysis of the errors associated with the extraction of shear parameters of the coating for different materials prove that, to obtain an error less than 5% in the shear parameters determination, the viscoelastic contribution, defined as the relative error in the thickness computed from the frequency shift by Sauerbrey equation, must be greater than a limit that depends on Q, which is defined as the ratio of the shear storage modulus (G') to shear loss modulus (G"). In the materials studied, polymers in the transition range or in the rubbery state with Q = 1 and 10, the viscoelastic contribution must be higher than 15% and 50%, respectively, for a 5% limit error in the shear parameters extraction. A criterion for a practical determination of the appropriate viscoelastic regimes is indicated.

It is a chronic and systemic disorder predominantly found in women, and is characterized by the appearance of a lymphocytic inflammatory infiltrate, with dryness of the oral cavity and eyes, secondary to involvement of the salivary and lacrimal glands. The underlying causal mechanism involves a number of factors and has not been clearly established, though an autoimmune response is known to be triggered, with the accumulation of immune complexes in the gland acini that interfere with gland function. In the oral cavity, xerostomia or hyposialia is the most disabling manifestation for patients, and is accompanied by rapidly progressing caries, candidiasis and an important worsening of buccodental health. High potency bisphosphonates are generally implicated and risk factors also include dental extractions. Prevention is of paramount importance. Management is controversial but there is little evidence basis and the consensus is to be conservative. Recent advances in this area are summarised in this concise review.

Graft versus host disease (GVHD) is one of the most frequent and serious complications of hematop... more Graft versus host disease (GVHD) is one of the most frequent and serious complications of hematopoietic stem cell transplantation, and is regarded as the leading cause of late mortality unrelated to the underlying malignant disease. GVHD is an autoimmune and alloimmune disorder that usually affects multiple organs and tissues, and exhibits a variable clinical course. It can manifest in either acute or chronic form. The acute presentation of GVHD is potentially fatal and typically affects the skin, gastrointestinal tract and liver. The chronic form is characterized by the involvement of a number of organs, including the oral cavity. Indeed, the oral cavity may be the only affected location in chronic GVHD. The clinical manifestations of chronic oral GVHD comprise lichenoid lesions, hyperkeratotic plaques and limited oral aperture secondary to sclerosis. The oral condition is usually mild, though moderate to severe erosive and ulcerated lesions may also be seen. The diagnosis is established from the clinical characteristics, though confirmation through biopsy study is sometimes needed. Local corticosteroids are the treatment of choice, offering overall response rates of close to 50%. Extracorporeal photopheresis and systemic corticosteroids in turn constitute second line treatment. Oral chronic GVHD is not considered a determinant factor for patient survival, which is close to 52% five years after diagnosis of the condition.
