This study evaluates the influence of mixed algal extract (Chlorella vulgaris, Euglena viridis an... more This study evaluates the influence of mixed algal extract (Chlorella vulgaris, Euglena viridis and Spirulina platensis) on common carp Cyprinus Carpio, which infected infect with bacterial pathogen Aeromonas hydrophila. C. carpio was administered intraperitoneally with various doses such as methanol extract (0, 0,1, 1, 10 and 100 mg/kg). The immunological parameters of fish blood and serum samples (Neutrophil activity, Lysozyme activity, Serum myeloperoxidase intensity, Serum bactericidal activity, and Serum antiprotease activity) were investigated at 7, 14, 21, and 28 days of post-immunization. Fish had been tested by virulent A. hydrophila for 30 days after treatment and 14 days after infection were identified with mortalities. The findings showed that neutrophil levels, lysozyme activity, serum bactericidal activity, myeloperoxidase activity, and serum antiprotease activity significantly enhanced (p<0.05) compared to untreated control. The current study focused on assessing the outcome of methanol extract of Chaetomorpha antennina ... more The current study focused on assessing the outcome of methanol extract of Chaetomorpha antennina (MECA) on the growth performance and immune modulation in both specific and non-specific immune responses through the assessment of neutrophil, serum lysozyme, serum myeloperoxidase, antiprotease, ceruloplasmin, reactive oxygen species (ROS), and reactive nitrogen species (RNS) activity in Labeo rohita (rohu) at 28 days post treatment along with assessment of the disease resistance capacity against Edwardsiella tarda at 30days post immunization. Fishes (n = 144; average weight 50.0 ± 0.23 g) were evenly divided into four treatments, with 12 fishes per tank in triplicates. The MECA was injected intraperitoneally in the fishes at different doses as 0, 25, 75, and 150 mg/kg of the body weight. Different doses of the extract (5, 50 and 500 mg/kg of body weight) were injected into the fish intraperitoneally while a control group was injected with 0.2 mL of sterile physiological salt solution. Variations in several immunostimulatory parameters (i.e., neutrophil, serum lysozyme, myeloperoxidase, serum antiprotease, and ceruloplasmin activity), reactive oxygen species (ROS) and reactive nitrogen species (RNS) were assessed after 7, 14, 21, and 28 days of post stimulation. E. tarda culture was injected into the fish after 28 days of post stimulation to induce infection to monitor fish mortality within 14 days. This study investigated the e#ects of prolonged administration of a commercial -glucan based immu... more This study investigated the e#ects of prolonged administration of a commercial -glucan based immunostimulant preparation, EcoActiva , in the form of a feed supplement, on non-specific immune parameters and the growth rate of snapper (Pagrus auratus). Fish held at a temperature representing either summer or winter conditions, were sampled periodically and assayed for head kidney macrophage activity via in vitro superoxide production, and classical and alternative complement activity. Fish were also weighed monthly and the growth rate determined. Fish fed on a diet supplemented with EcoActiva and held at the winter temperature had a significant enhancement of macrophage superoxide anion production upon stimulation with phorbol myristate acetate (PMA), and this increased activity was maintained throughout the trial. Macrophage activity in fish fed the supplemented diet and held at the summer temperature was also increased. However, EcoActiva failed to increase either classical or alternate complement activity. Most significantly EcoActiva resulted in an increase in growth rates of the fish held at the winter temperature as compared to the control fish, although no di#erence was seen between the groups held at the summer temperature. These results suggest that routine incorporation of -glucan preparations like EcoActiva

The monoclonal antibody DLT15, specific for thymocytes and peripheral T-cells of the teleost fish... more The monoclonal antibody DLT15, specific for thymocytes and peripheral T-cells of the teleost fish Dicentrarchus labrax (sea bass), was used to purify immunoreactive cells from blood and gut-associated lymphoid tissue. The purification was performed by immuno-magnetic sorting of leucocyte fractions enriched by Percoll density gradient centrifugation, and the purity of the isolated cells was estimated by cytofluorimetric analysis. Following a single step, the percentage of DLT15-purified cells was 88 10% for gut-associated lymphoid tissue and 79 18% for blood leucocytes. DLT15-purified cells from gut-associated lymphoid tissue were employed for RNA extraction and cDNA synthesis. In RT-PCR experiments using as primers degenerate oligonucleotides corresponding to the peptide sequence MYWY and VYFCA of the trout TcR chain, a 203 bp product was amplified. When sequenced, the cDNA was found to show 60% nucleotide identity to the trout TcR V 3 . By 3 -RACE the cDNA was elongated to obtain the TcR constant region, with high similarity to other fish TcR sequences. These results strongly suggest that cells recognised by DLT15 are putative T lymphocytes.

Los peces óseos poseen sistemas inmunitarios capaces de llevar a cabo respuestas humorales y celu... more Los peces óseos poseen sistemas inmunitarios capaces de llevar a cabo respuestas humorales y celulares tanto innatas como específicas; esencialmente, los sistemas defensivos de los peces teleósteos tienen los mismos componentes que los de los mamíferos, a quienes anteceden evolutivamente. En esta revisión se describen los componentes de los sistemas inmunitarios de los teleósteos, enfatizando las particularidades presentes en los peces mas no en los mamíferos, cuyos sistemas defensivos se conocen en mayor detalle. El estudio de la inmunología de los peces es relevante para fines tanto básicos como aplicados. Por la vertiente básica, es importante para comprender el origen y la evolución del sistema inmunitario de los vertebrados superiores. Por la vertiente aplicada, a pesar de que hay algunas vacunas comerciales para proteger peces de interés comercial contra algunas bacterias y virus, ampliar el conocimiento del sistema inmunitario de estos organismos permitiría incrementar la productividad e inocuidad de la acuacultura, mediante el desarrollo de nuevos métodos de estimulación inmunitaria y vacunación eficaces.

The effect of temperature (8, 12, 15 and 18 degrees C) on a variety of non-specific defence and h... more The effect of temperature (8, 12, 15 and 18 degrees C) on a variety of non-specific defence and haematological parameters was examined in three geographically distinct reared strains (Canadian, Icelandic, Norwegian) of Atlantic halibut. The results indicate that temperature exerts a considerable influence on some blood parameters (packed cell volume and the percentage population of leucocytes in peripheral blood) and on some humoral parameters (serum lysozyme activity and serum protein levels) of halibut. A high temperature of 18 degrees C caused a decrease in the number of circulating blood cells and an increase in serum lysozyme levels; effects consistent with those reported within the literature for stress. The different strains of halibut exhibited differing responses with respect to differential counts of peripheral blood lymphocytes and thrombocytes, and to serum protein concentrations, serum lysozyme activity, serum iron content, unsaturated iron binding capacity of serum and O2- production by kidney macrophages.

ABSTRACT The role of fish erythrocytes (FE) as phagocytic cells has poorly been investigated, unt... more ABSTRACT The role of fish erythrocytes (FE) as phagocytic cells has poorly been investigated, until now. Here, we have focussed our attention on the interplay between rainbow trout (Salmo gairdneri Richardson) erythrocytes and Candida albicans (CA). At the same time, the intervention of autologous head kidney macrophages (MO) in the CA processing by FE has been studied. Data show that CA particles bind to FE, which, in turn, are able to engulf but not kill them. In the presence of MO, a decrease of FE with bound CA occurs and, in some microscopic images, FE form rosettes with MO. Phagocytosis of CA is higher in rosetting MO than in non-rosetting ones. According to our findings, it appears that FE represent a reservoir of engulfed CA and rosetting is an efficacious phenomenon of presentation of pathogens to MO, where an effective clearance of them can take place.
