In the present study, a feruloyl esterase DLFae4 identified in our previous research was modified... more In the present study, a feruloyl esterase DLFae4 identified in our previous research was modified by error-prone PCR and site-directed saturation mutation to enhance the catalytic efficiency and acyltransferase activity further. Five mutants with 6.9–118.9% enhanced catalytic activity toward methyl ferulate (MFA) were characterized under the optimum conditions. Double variant DLFae4-m5 exhibited the highest hydrolytic activity (270.97 U/mg), the Km value decreased by 83.91%, and the Kcat/Km value increased by 6.08-fold toward MFA. Molecular docking indicated that a complex hydrogen bond network in DLFae4-m5 was formed, with four of five bond lengths being shortened compared with DLFae4, which might account for the increase in catalytic activity. Among other indicators, the impact of the pilot low-carbon initiative may be evaluated based on whether it induces enterprises to engage in green technology innovation. Using green patent application data from Chinese listed companies between 2009 and 2018, this paper applies a time-varying difference-in-difference (DID) model to conduct a multi-dimensional empirical test on the changes in listed companies’ degrees of green innovation before and after the publication of the list of three batches of pilot cities. Large rotating machines are critical equipment in many process industries, such as energy, chemic... more Large rotating machines are critical equipment in many process industries, such as energy, chemical, and power generation. Due to high rotating speed and tremendous momentum of the rotor, the centrifugal force may lead to flying apart of the rotor parts, which brings a great threat to the operation safety. Early detection and prediction of potential failures could prevent catastrophic plant downtime and economic loss. In this paper, we divide the operational states of a rotating machine into normal, risky, and high-risk ones based on the time to the moment of failure. Then, a cascade classification algorithm is proposed to predict the states in two steps; first, we determine whether the machine is in normal or abnormal condition; for time periods predicted as abnormal, we further classify them into risky or high-risk state.
