Vários fatores têm sido abordados e investigados sob diversos pontos de vista, que abrangem desde os aspectos pedagógicos de uso do computador até a aplicação de tecnologias na construção de ambientes de ensino baseados em computador. Em face do dinamismo da tecnologia, novas estratégias estão surgindo e com potencial para aplicação em educação. Este trabalho contextualiza ambientes de ensino com foco no uso de recursos computacionais e apresenta um framework para o uso de tecnologias computacionais emergentes em ambientes de ensino.

A leishmaniose visceral canina é uma doença grave e a morte ocorre por falência renal, consideran... more A leishmaniose visceral canina é uma doença grave e a morte ocorre por falência renal, considerando que os métodos diagnósticos convencionais não possibilitam a classificação clínica do animal. O objetivo deste estudo foi associar a carga parasitária renal aos achados clínicos e histopatológicos em cães com leishmaniose visceral. A análise microscópica revelou predomínio de nefrite intersticial mononuclear de graus variados em 59,3% dos cães avaliados. Entretanto, não houve diferença entre a carga parasitária renal de sintomáticos e oligossintomáticos (P= 0,35). As lesões renais foram de ordem inflamatória e a quantidade de parasitos não influenciaram na característica dessas lesões e nem nas alterações bioquímicas, mesmo em cães com diferentes classificações clínicas.

No âmbito dos alimentos recomendados para redução do risco de desenvolver doenças e para melhoria... more No âmbito dos alimentos recomendados para redução do risco de desenvolver doenças e para melhoria da qualidade de vida, estão as frutas e seus derivados. Este trabalho teve como objetivo desenvolver um suco misto de manga, goiaba e acerola, com aceitação sensorial, utilizando delineamento de mistura para determinar as proporções das polpas. Em cada tratamento, essas proporções, foram definidas segundo um delineamento de mistura simplex aumentado de 10 tratamentos. Foram realizadas análises físico-químicas, tais como: pH, sólidos solúveis (oBrix), acidez titulável, relação de sólidos solúveis (SST)/acidez titulável (ATT), teor de vitamina C e análise sensorial. Com base nos resultados, verificou-se que todas as formulações, obtidas a partir de manga, goiaba e acerola, foram aceitas pelos consumidores. The work presented here demonstrates the feasibility of using the single-mode fibers of an optica... more The work presented here demonstrates the feasibility of using the single-mode fibers of an optical Internet network to deliver visible light between separate laboratories as a way to perform remote spectroscopy in the visible for teaching purposes. The coupling of a broadband light source into the single-mode fiber (SMF) and the characterization of optical losses as a function of the wavelength are discussed. Sample spectra were measured with a portable spectrometer controlled by an acquisition program developed with the LabVIEW software that allows the data to be collected and analyzed.

Liquid-phase microextraction based on polypropylene hollow fibers and CE were applied for the chi... more Liquid-phase microextraction based on polypropylene hollow fibers and CE were applied for the chiral determination of hydroxychloroquine (HCQ) and its metabolites (desethylchloroquine, DCQ; desethylhydroxychloroquine, DHCQ; bisdesethylchloroquine, BDCQ) in human urine. The analytes were extracted from 3 mL of urine spiked with the internal standard (metoprolol) and alkalinized with 250 muL of 2 M NaOH. The analytes were extracted into 1-octanol impregnated in the pores of the hollow fiber, and into an acid acceptor solution inside the hollow fiber. The electrophoretic separations were carried out in 100 mmol/L Tris buffer (pH adjusted to 9.0 with phosphoric acid) containing 1% w/v S-beta-CD and 30 mg/mL HP-beta-CD with a constant voltage of +18 kV. The method was linear over the concentration range of 10-1000 ng/mL for each HCQ stereoisomer and 21-333 ng/mL for each metabolite stereoisomer. Within-day and between-day assay precision and accuracy for the analytes were studied at three concentration levels for each stereoisomer and were lower than 15%. The developed method was applied for the determination of the cumulative urinary excretion of HCQ, DCQ, and DHCQ after oral administration of rac-HCQ to a health volunteer. The results obtained are in agreement with previous literature data.

The gas chromatography/mass spectrometry (GC/MS) method for organic acid analysis was established... more The gas chromatography/mass spectrometry (GC/MS) method for organic acid analysis was established in developed countries since 1980s, but due to the small number of experienced clinical biochemists in this field and also the short availability of mass spectrometers scarce reports exist on the prevalence of organic acidemias (OAs) in developing countries like Brazil. During January 1994 to July 2008, we analyzed organic acids by GC/MS in urine specimens obtained from Brazilian children with clinical suspicion of metabolic diseases. Two hundred and thirty four cases of disorders of organic acid metabolism, including 218 OAs (3.17%), were diagnosed among 6866 patients investigated. The most frequent disorders were primary lactic acidemia (57), methylmalonic acidemia (34), glutaric acidemia type I (33), propionic acidemia (18), 3-hydroxy-3-methylglutaric aciduria (17), L-2-hydroxyglutaric aciduria (9) and multiple carboxylase deficiency (9). Fourteen cases of mitochondrial fatty acid oxidation disorders, as well as 12 aminoacidopathies and 4 cases of vitamin B12 deficiency were also detected. Prompt treatment following diagnosis led to a better outcome in a considerable number of patients. Detection of OAs in loco in developing countries is important despite the implied extra costs, since it allows rapid therapy in many cases with a significant reduction of morbidity and mortality and makes the physicians more aware of these pathologies.

We optimize a SPME procedure to extract these analytes from liquid culture medium. The method was validated and SPME showed to be a useful tool to be used in biotransformation studies. The biotransformation results showed that it is possible to obtain a drug in its enantiomeric pure form.

A simple, rapid and sensitive off-line solid-phase microextraction method coupled to high-perform... more A simple, rapid and sensitive off-line solid-phase microextraction method coupled to high-performance liquid chromatography was developed for the stereoselective analysis of the major ibuprofen metabolites in human urine samples. For this purpose, a carbowax/templated/resin coated fiber was used. Several parameters influencing the efficiency of the analyte extraction were explored. The chromatographic separation was achieved on a Chiralpak AS column using hexane:isopropanol (95:5 v/v) plus 0.05% trifluoroacetic acid as the mobile phase, at a flow-rate of 1.2 mL/min. Detection was carried out at 230 nm. The mean recoveries were between 1.4 and 1.6%, with R.S.D. values lower than 9.5 for both 2-OHIbu enantiomers, and between 3.7 and 5.7%, with R.S.D. values lower than 13.4 for all four COOHIbu stereoisomers. The method was linear over the 5-50 g/mL concentration range and the quantification limit was 5 g/mL for all metabolite stereoisomers. Within-day and between-day assay precision and accuracy for both metabolites were studied at three concentration levels of each stereoisomer (15, 30 and 45 g/mL) and were lower than 15%.
