The determination was carried out by using Phenomenex C18 column with 0.24% sodium lauryl sulphate: acetonitrile: acetic acid (pH-4.0) (58:40:02) as the mobile phase. The flow rate was 1.5 ml/ min. and the eluent was monitored at 295 nm. The Retention time of nitazoxanide and ofloxacin were 2.2 and 5.4 respectively. Linearity for the nitazoxanide and ofloxacin were found in the range of 400-600 µg/ml and 160 -240 µg/ml respectively. The method was reproducible, with good resolution between nitazoxanide and ofloxacin and can be use for routine analysis.

Simultaneous equation method involves the measurement of absorbance at two wavelengths 264 nm (λ max of Ibuprofen) and 319 nm (λ max of Tizanidine). Absorbance correction method in which absorbance is measured at two wavelengths 264 nm at which Tizanidine has no absorbance and 319 nm at which both the drugs have considerable absorbance. The methods were found linear between the range of 150-750 µg/ml for Ibuprofen and 3-15 µg/ml for Tizanidine for both the methods. The accuracy and precision were determined and validated statistically. Both the methods showed good reproducibility and recovery with % RSD less than 1. The methods were found to be rapid, specific, precise and accurate and can be successfully applied for the routine analysis of Ibuprofen and Tizanidine in bulk and combined dosage form.

The mobile phase consisted of 0.02 M phosphate buffer: acetonitrile with 5 ml of 0.4% triethylamine in ration of (35:65 v/v) and adjusted to pH 4. The detection wavelength was carried out at 254 nm. The method was linear over the concentration range for Aceclofenac 5-25 µg/ml and for Diacerein 2-10 µg/ml. The recoveries of aceclofenac and Diacerein were found to be in the range of 99.23-100.98% and 99.45-100.61% respectively. The validation of method was carried out using ICH-guidelines. The described HPLC method was successfully employed for the analysis of pharmaceutical formulations containing combined dosage form.

Method I simultaneous equation method involves the measurement of absorbances at two wavelengths 274 nm (λ max of Aceclofenac) and 319 nm (λ max of Tizanidine). Method II involves Absorbance correction method in which absorbance is measured at two wavelengths 274 nm at which Tizanidine has no absorbance and 319 nm at which both the drugs have considerable absorbance. The method was found linear between the range of 5-25 μg/ml for Aceclofenac and 2-10 μg/ml for Tizanidine for both the methods. The accuracy and precision were determined and validated statistically.
