CD32a is a marker of a CD4 T-cell HIV reservoir harbouring replication-competent proviruses (original) (raw)

Abstract: The persistence of the HIV reservoir in infected individuals is a major obstacle to the development of a cure for HIV. Here, using an in vitro model of HIV-infected quiescent CD4 T cells, we reveal a gene expression signature of 103 upregulated genes that are specific for latently infected cells, including genes for 16 transmembrane proteins. In vitro screening for surface expression in HIV-infected quiescent CD4 T cells shows that the low-affinity receptor for the immunoglobulin G Fc fragment, CD32a, is the m… Show more

“…Following the gating strategy used by Descours et al . (24), we divided CD32 + CD4 + T cells into cells expressing intermediate levels of CD32 (CD32 intermedlate ) and cells expressing high levels of CD32 (CD32 hi ) (fig. S1).…”

“…Finally, penetration of these compounds into tissue sites such as the central nervous system and gut-associated lymphoid tissue (GALT) (Eisele & Siliciano, 2012) will be important to determine. Potency and specificity could be potentially enhanced using drug delivery systems such as nanoparticles coated with specific antibodies targeting CD4 or a latency marker, such as the recently described CD32a (Descours et al, 2017). …”

“…Focuses on different reservoir sites and cells, including CXCR5 ϩ PD1 ϩ Tfh cells (44,45), CD32 CD4 ϩ T cells (46), and tissue monocytes/macrophages, are currently being actively pursued. Peripheral blood HIV-1 DNA in circulating CD4 ϩ T cells represents an acknowledged site for estimating the total HIV reservoir in HIV-infected patients (43,(47)(48)(49).…”
