Özet Pseudomonas savastanoi pv. phaseolicola, fasulyelerde haleli yanıklık hastalığına sebep olan... more Özet Pseudomonas savastanoi pv. phaseolicola, fasulyelerde haleli yanıklık hastalığına sebep olan, tohumla taşınan ve ciddi zararlara yol açan bakteriyel etmendir. İç Anadolu Bölgesindeki 12 ilden ve ekonomik anlamda fasulye ekimi yapılan alanlardan, ekilen arazi büyüklüğü esas alınarak, tohum örnekleri toplanmıştır. Patojenin izolasyonu ve tanısında; Nutrient Agar (NA), Modified sucrose peptone (MSP), King B besi yerlerinde gelişim, levan oluşumu, 37 °C'de gelişim, oksidaz reaksiyonu, pektolitik aktivite testi, arginine dehidrolaz testi, karbon kaynaklarının kullanımı, arbutin ve jelatinin hidrolizi, nitrat indirgenmesi, H 2 S üretimi, tütün yaprağında (cv. White Burley) aşırı duyarlılık reaksiyonu testleri esas alınmıştır. Patojenisite testlerinde, Dermason çeşidi fasulye bitkilerine yapılan 10 8 hücre/ml P. s. pv. phaseolicola süspansiyonu ile inokulasyon sonucu tipik yaprak lekeleri gözlenmiştir. Etmeninin moleküler tanısı, Bio-PCR yöntemi ve phaseolotoksin geninin amplifikasyonu ile tespit edilmiştir. Elde edilen bulgulara göre, toplam 175 kuru fasulye tohumu örneğinin 38'inde Pseudomonas savastanoi pv. phaseolicola tespit edilmiş ve İç Anadolu Bölgesinde etmenin fasulye tohumlarında bulaşıklılık oranı %21,71 olarak belirlenmiştir. The first outbreak of fire blight incited by Erwinia amylovora (Burr.) Winslow et al. occurred on... more The first outbreak of fire blight incited by Erwinia amylovora (Burr.) Winslow et al. occurred on pome fruits in Turkey in 1985, and it is now one of the most serious diseases of pear, apple, quince, and loquat ). In this study, experiments were conducted in Konya Province to evaluate the efficacy of Glomus intraradices Schenck & Smith vesicular-arbuscular mycorrhiza (VAM) and bactericides for control of the shoot blight phase of fire blight and control of shoot growth on the different apple cultivars (Gala, Red Elstar, Pinova, Jonagored) on M9 rootstock in 2002 to 2003. Streptomycin provided 84.38% to 95.24% and 85.28% to 89.97% disease control in 2002 and 2003, respectively. Copper complex was not so effective against shoot blight phase of the disease, and it reduced disease by 16.18% to 27.75% and 14.48% to 19.06% in 2002 and 2003, respectively. Results of VAM application were encouraging, indicating a reduction of fire blight by Glomus intraradices of between 9.7% and 50.5% in 2002 and between 23.9% and 48.4% in 2003, respectively.

The etiology of a new soft rot disease of tulips, causing leaf spots, leaf blight, neck and bulb ... more The etiology of a new soft rot disease of tulips, causing leaf spots, leaf blight, neck and bulb rot in Konya, Turkey, was investigated. This disease occurred in tulip fields grown for seed bulbs of various varieties in 2002. Bacteria were isolated from the lesions on leaf, bud neck and bulbs and the causal bacterium was identified asPectobacterium carotovorum on the basis of biochemical and physiological tests. Cells were Gram-negative, rod-shaped, fermentative, potato-rot positive. Colonies were capable of growth at 37°C. On nutrient agar the colonies were creamy-white. The isolates were non-fluorescent on King’s B medium, positive for acetoin production, gelatin liquefaction, acid-production-from-lactose, and catalase; and negative for gas from glucose, reducing substances from sucrose, and phosphatase activity. Additionally, tests for egg yolk (lecithin), sensitivity to erythromycin, and pigmentation on yeast dextrose carbonate agar were negative; growth on 5% sodium chloride was positive. All the bacterial isolates obtained from the leaf, bud neck and bulbs produced the original symptoms following inoculation to the susceptible tulip variety ‘Gander’. The rate of damage caused by this bacterium was evaluated on several tulip varieties under field and storage conditions. Gander was the most susceptible variety in the field whereas ‘Salmon Parrot’ exhibited the highest rate of bulb rot in storage. Disease severity was lower in 2003 than 2002.
