An electrochemical sensor based on modification of carbon paste electrode by the functionlized MW... more An electrochemical sensor based on modification of carbon paste electrode by the functionlized MWCNT (f-MWCNT/CPE) was prepared for determination of Domperidone in formulation and in human serum. f-MWCNTs were prepared by a simple surface oxidization of the pristine MWCNTs using a famous, less expensive and less vigorous KMnO 4. The f-MWCNTs were characterized using FTIR spectra, TEM, BET N 2 sorption isotherms and TGA. Point of zero charge of pristine and oxidized MWCNTs was also determined. The surface morphology and charge transfer resistance of the bare and f-MWCNT/CPEs were investigated using SEM and EIS. The detection limit (4.68 × 10 −11 M) indicates the sensitivity of the developed electrode for trace assay of Domperidone in biological fluids. The binding interactions of Domperidone with DNA and the mode of this binding have been investigated using cyclic voltammetry, UV-vis spectrophotometry and molecular docking studies. The estimated binding free energy (–20.47 KJmol −1) by Autodock was comparable to the experimental free energy (–25.82 KJmol −1) in the cyclic voltammetric analysis. The minor groove-binding mode of interaction was also supported by docking simulation studies. The higher binding constant value (8.77 × 10 4 M −1) might be sufficient to interfere with DNA replication and thus, Domperidone can be used as an anticancer therapy.

Research paper thumbnail of A Direct Spectrophotometric Method for Determination of Antiparasitic Drug Nitazoxanide in a Bulk Form and Pharmaceutical Formulation

A simple and direct UV-VIS spectrophotometric method for determination of nitazoxanide in a bulk form and pharmaceutical formulation "Cryptonaz powder" was elaborated and validated. The method did not require sample pretreatment, extraction, or formation of colored chromogens prior to the analysis. The analysis was carried out in a B-R universal buffer ofpH 7 containing 20% (v/v) of acetonitrile. Absorbance of nitazoxanide was measured at λ max = 420 nm. Beer's law was obeyed over the nitazoxanide concentration range 1.0 x 10 -6 -1.0 x 10 -4 mol L -1 (0.293-29.30 μg mL -1 ); detection limit was 3.0 × 10 -7 mol L-' (0.088 μg mL -1 ). Insignificant interferences from the excipients present in "Cryptonaz powder" or degradation products of nitazoxanide were detected. The described method is direct, simple, precise, less time-consuming, and offers economical assay of nitazoxanide with sensitivity better than or at least comparable to that of the reported spectrophotometric methods. The ionization constant (pk a ) ofnitazoxanide was also determined.

Two validated adsorptive anodic stripping voltammetry methods have also been developed for its determination using 7 % (w/w) Na-MMT-modified CPE. Langmuir isotherm can describe the experimental data well, indicating monolayer adsorption. Limits of detection of 2.4 × 10−7 and 3.0 × 10−8 mol L−1 PZ.HCl were achieved in the bulk form using linear-sweep and square-wave adsorptive anodic stripping voltammetric methods, respectively. The developed methods were also successfully applied for determination of PZ.HCl in pharmaceutical formulation and in spiked human serum without time-consuming extraction steps prior to the analysis. Statistical comparison of the results of the proposed methods with that of official and reported spectrophotometric methods indicates no significant difference in precision and accuracy between the proposed and reference methods.
