To investigate the prevalence of LTBI in patients with psoriasis who are candidates for biological therapy. LTBI was investigated in patients with moderate-to-severe psoriasis (n = 243), Crohn disease (n = 64) or rheumatoid arthritis (RA) (n = 56) and in healthcare workers (n = 1683). LTBI diagnosis was based on positive QuantiFERON-B Gold In-Tube (QFT-GIT) in vitro assay without any clinical, radiological or microbiological evidence of active tuberculosis. LTBI was diagnosed in 8.2% of patients with psoriasis, 7% with Crohn disease and 9% with RA, and in 8.8% of healthcare workers (P = 0.9). Patients with psoriasis who also had LTBI (n = 20) received a 9-month prophylaxis with isoniazid (5 mg kg(-1) daily). None of these patients developed active tuberculosis infection after receiving biological therapy (etanercept, adalimumab, infliximab or ustekinumab) for 37 ± 32 weeks (mean ± SD). All patients with psoriasis were retested for LTBI after 31 ± 1.7 months. Five of the 20 patients with LTBI presented QFT-GIT reversion and two patients out of 243 (0.8%) had QFT-GIT conversion and received antibiotic prophylaxis. The prevalence of LTBI in patients with psoriasis is similar to that in patients with Crohn disease or RA and in healthcare workers. Prophylaxis with isoniazid is effective in preventing tuberculosis reactivation in patients with LTBI receiving biological therapy.

Identifying and targeting these adaptive mechanisms may provide new therapeutic strategies. Here we present the results of genome-scale CRISPR/Cas9-based synthetic lethality screens in isogenic pairs of BRCA1- and BRCA2-deficient cells that identified the gene encoding CIP2A as essential in a wide range of BRCA1- and BRCA2-mutated cells. Unlike PARP inhibition, CIP2A-deficiency does not cause accumulation of replication-associated DNA lesions that require homologous recombination for their repair. CIP2A is cytoplasmic in interphase but, in mitosis, accumulates at DNA lesions as part of a complex with TOPBP1, a multifunctional genome stability factor. In BRCA-deficient cells, the CIP2A-TOPBP1 complex prevents lethal mis-segregation of acentric chromosomes that arises from impaired DNA synthesis. Finally, physical disruption of the CIP2A-TOPBP1 complex is highly de...

Fresh shale samples taken from a well-known outcrop section of the Silurian Vila Maria Formation,... more Fresh shale samples taken from a well-known outcrop section of the Silurian Vila Maria Formation, located on the northeastern margin of the Paraná Basin (Três Barras Farm, Goiás State, central-western Brazil), have been analysed for both palynological and radiometric age determinations. The rocks yielded a fairly diverse, age-definitive cryptospore assemblage, and the same samples proved suitable for Rb-Sr analysis, despite Silurian sedimentary rocks being rarely suited to radiometric dating techniques. This study thus introduces an alternative, independent method for estimating the minimum depositional age of the Vila Maria Formation. The Rb-Sr age value has been calculated via an isochron diagram that yields 435.9 ± 7.8 Ma. Accordingly, the latter value is interpreted as the minimum depositional age of the analysed Vila Maria strata, implying their deposition during Llandovery times. This concurs with the palynological results, which indicate an age no older than Early Silurian and, more particularly, favour an Early to Middle Llandovery (Rhuddanian-Early Aeronian) attribution.

A previous palynological work based on chitinozoans has dated these strata as Late Ludlow, Early Pridoli and Early Lochkovian. Although samples are relatively poor in miospores, the present study reveals the most diversified Silurian-Earliest Lochkovian palynoflora ever reported from South America, an interval where miospore information is still very rare. Sixty-four species have been identified, of which two are new (Artemopyra urubuense and Retusotriletes amazonensis). One single specimen attributed to the genus Grandispora, identical to unpublished forms identified by one of us (P.S.) in Lochkovian sections of Saudi Arabia, has been found in a Lochkovian sample from the Urubu River. These constitute the oldest geological record of Grandispora. Among the abundant acritarchs, specimens of the genus Schizocystia have been observed for the first time in pre-Devonian rocks. Although the Late Silurian-Early Lochkovian miospore biostratigraphy is still in its infancy in South America, it seems that Amazon Basin assemblages do not show significant differences in composition in comparison to coeval palynofloras from other areas of northwestern Gondwana. This is surprising since the miospore localities under consideration are widely distributed over high to low paleolatitudes, i.e. from cool temperate to dry subtropical belts. Conversely, Siluro-Devonian Gondwanan assemblages differ considerably from the contemporary palynofloras of the Old Red Sandstone Continent and adjacent areas, thus contradicting recent phytogeographic interpretations based on megafloras.
