This research aimed evaluated two doses of estradiol benzoate (BE) administered early in the resynchronization protocol 22 days (Day 22) after the first timed-AI (TAI) on the pregnancy rate by TAI (P/AI), pregnancy loss (PL) and induction of new follicular wave emergence (FW) in suckled beef cows. After 1st TAI and regardless of pregnancy status, 1426 cows (768 B. taurus and 728 B. indicus) were randomly divided into 2 groups according to the dose of BE [1 mg (n = 721) or 2 mg (n = 705); Bos taurus (RIC-BE®) and zebu (Gonadiol®)]. On day 22, cows received an intravaginal progesterone device (P4; PRIMER® and DIB®, taurine and zebu respectively). After 8 days (Day 30), the P4 removed and pregnancy diagnosis accomplished by ultrasound. The non-pregnant females received prostaglandin analogue [Bos taurus (Estron®) or zebu (Ciosin®)] IM. was identified as Coccidiodes immitus, which is a human health concern. All cultures and other sa... more was identified as Coccidiodes immitus, which is a human health concern. All cultures and other samples from themare were destroyed. Personnel involved with the case were evaluated for seroconversion (i.e. positive titer) and fortunately all were negative. The mare was re-bred and a second embryowas collected. However, fungal hyphaewere present in the search dish. The embryo was washed numerous times and transferred into a recipientmare. The recipientmarewas administered oral fluconazole (14mg/kg loading dose; 5mg/ kg maintenance dose) to inhibit fungal growth associated with transfer of the embryo. The recipient mare became pregnant and carried to term. The fungal organism in the embryo search dish was confirmed by PCR to be C. immitus. Since C. immitus has been reported to disseminate from a primary respiratory infection, chest radiographs and a bronchial-alveolar lavage were performed to determine if the lungs were the nidus of infection. No evidence of infection was identified in the respiratory tract. The donor mare was sent home with instructions to administer fluconazole orally for an additional 6 to 8 weeks. In conclusion, fungal endometritis due to C. immituswas diagnosed in an embryo donor mare. Two ET pregnancies were established in recipient mares. Caution should be taken by veterinarians performing fungal culture in geographical areas endemic for C. immitus as the organism is a potential human health hazard.

RESUMO-O desenvolvimento embrionário de oócitos bovinos maturados in vitro (MIV) foi avaliado em ... more RESUMO-O desenvolvimento embrionário de oócitos bovinos maturados in vitro (MIV) foi avaliado em meio suplementado com líquido folicular eqüino (Lfe). Foram distribuídos 1045 oócitos em 11 repetições formando três grupos tratamentos (T1, T2, T3) e um controle (C). O meio de maturação utilizado foi o TCM-199 acrescido de piruvato de sódio, hormônio folículo estimulante recombinante (rFSHh) e hormônio luteinizante equino (LHe). Suplementou-se esse meio com 10% de soro de égua em estro para o grupo controle e para T1, T2 e T3, o meio foi suplementado com 5, 10, e 20% de LFe, respectivamente. Os oócitos foram maturados in vitro (MIV) por 24h. A fecundação in vitro (FIV) foi realizada em meio Talp-Fert. A MIV e a FIV foram realizadas em estufa a 39ºC com 5% de CO 2 em ar e umidade saturada. Os zigotos foram cultivados em meio SOFaaci, sob óleo mineral no interior de bolsas plásticas gaseificadas. As taxas de clivagem e de blastocistos foram observadas diariamente (D), e em D7, foram superiores (P<0,05) para o T2, em comparação ao T1 e T3, mas semelhantes (P>0,05) às do grupo controle. Em D9, a taxa de blastocistos do T2 foi superior (P<0,05) à do controle e T1, mas semelhante à do T3. Não houve diferença na taxa de eclosão entre os grupos (P>0,05). O LFe, na concentração de 10% pode ser utilizado, em substituição ao soro de égua em estro para suplementar o meio de MIV de oócitos bovinos. ABSTRACT-Embryo development of bovine oocytes was evaluated using maturation medium supplemented with equine follicular fluid (eFF). One thousand and forty five (1045) oocytes were distributed in 11 replications forming three treatment groups (T1, T2 e T3) and one Control (C). TCM-199 added with sodium pyruvate, rFSHh and LHe was used as maturation medium. This medium was supplemented with 10% estrous mare serum for Control group, and 5, 10, and 20% eFF, respectively, for T1, T2 e T3 groups. In vitro maturation (IVM) of all groups was performed during 24h. In vitro fertilization (IVF) was performed in TALP-FERT medium. IVM and IVF were carried out in an incubator at 39ºC with 5% CO 2 in air and saturated humidity. Zygotes were cultured in SOFaaci medium, under mineral oil in gasified bags. Cleavage and blastocyst rates were daily observed (D), and at D7, were higher (P<0.05) for T2 than T1 and T3, but they were similar (P>0.05) for those from control group. At D9, blastocyst rate of T2 was higher (P<0.05) for those from control and T1, but similar for T3. No differences were observed on hatching rates between groups (P>0.05). The eFF, at a 10% concentration, can replace the use of estrous mare serum to supplement the IVM medium of bovine oocytes.

In order to evaluate the effect of follicular aspiration on subsequent fertility in mares, two experiments were conducted. In Experiment I, fifteen Criollo mares were allocated to one of three groups according to the diameter of the aspirated follicle during estrus: 25-29mm (n=4; Group 1); 30-34mm (n=6; Group 2); 35mm (n=5; Group 3) and control group (n=15; Group 4). For in vitro embryo production following ovum-pickup groups with different number of oocytes and ... more For in vitro embryo production following ovum-pickup groups with different number of oocytes and volume of medium may be required to maintain an individualized production of each cow. Bovine oocytes were randomly allocated in a 3x3 factorial design in order to evaluate the effect of number of oocytes (5, 10 and 20) and maturation medium volume ratios (1:1, 1:5 and 1:10 oocyte/μl) on embryo production. After 22-24h maturation in TCM-199, in vitro fertilization was performed in FERT-TALP medium, for 18-22h. Groups of 20 presumptive zygotes were cultured in 200μl of SOF medium for eight days. There was no effect of interaction between number of oocytes and volume of maturation medium (P>0.05) on embryo development. Cleavage rates were not affected either by number of oocytes nor by volume of medium (P>0.05). Blastocyst rates on D7 (19, 26 and 28%; D0=day of fertilization) and D9 (23, 33 and 33%) were lower (P<0.05) in groups of 5 oocytes compared with groups of 10 and 20 oocytes, respectively. Blastocyst rates on D7 (22, 24 and 27%) and D9 (26, 31 and 32%), were similar (P>0.05) for ratios 1:1, 1:5 and 1:10 oocytes/μl, respectively. Hatching rate with a 1:1 volume ratio (26%) was lower (P<0.05) than that obtained with a 1:5 volume ratio (44%). Maturation in larger groups increases embryo production. The volume of maturation medium does not influence the blastocyst production, however a low oocyte/μl medium ratio reduces the hatching rate.

The hypothesis of this study was to determine whether low doses of hCG has superior efficacy to cypionate to induce ovulation in anestrous cows and provide higher pregnancy rate in oestrus-synchronization programs. Synchronization of ovulation and conception rate to timed AI (TAI) were evaluated in anestrus Bos taurus taurus suckling beef cows 45 ± 15 days postpartum and with body condition score of 2.9 (1 to 5) maintained in a native pastured system in the south of Brazil. Females were evaluated with ultrasound on the Day 0 (D0) of the protocol (Day 0), day 8 (D8), immediately before TAI (D10), and 7 days after TAI (Day 17). Cento e seis mórulas de camundongos da cepa CF1 Suíço Albina, coletadas 76-78h após a administraç... more Cento e seis mórulas de camundongos da cepa CF1 Suíço Albina, coletadas 76-78h após a administração de HCG, foram congeladas em solução de congelamento contendo glicerol 2,0M + sacarose 0,25M (Grupo S) ou Glicerol 2,0M + lactose 0,25M (Grupo L), ambas em PBS modificado. Foi efetuado um pré-tratamento com Glicerol 2,0M durante 4 minutos, sendo as mórulas transferidas, posteriormente, para a solução de congelamento, durante 1 minuto. O envase foi realizado em palhetas de 0,25ml em grupos de 5 a 11 mórulas. As palhetas contendo os embriões foram colocadas em vapor de N2 durante 2 minutos e, após, mergulhados em N2 líquido. O descongelamento foi efetuado em banho-maria a 37°C por 20 segundos e foram recuperadas 103 mórulas após a diluição do crioprotetor, que foi efetuada em solução de sacarose 0,5M (Grupo S) e lactose 0,5M (Grupo L) , durante 5 minutos , à temperatura ambiente . The effect of age, follicular diameter and month of the breeding season (September to January) on... more The effect of age, follicular diameter and month of the breeding season (September to January) on the hCG induction of ovulation was evaluated using 123 Criollo mares. Age varied between two and 24 years and the animals were examined daily by rectal palpation and ultrasonography with a 5 MHz linear transducer. When ovarian follicles reached a diameter of 30 to 35 mm, ovulation was induced with an i.v. injection of 1000 IU (n = 39); 1500 IU (n = 41) or 2000 IU (n = 43) of hCG. The mares were bred the next day and examined daily until ovulation was detected. The percentage of mares ovulating before 24 h of hCG injection was 10.3%, 7.3% and 4.7%; until 48 h after injection 92.3%, 85.3% and 86.0% of the mares treated with 1000, 1500 and 2000 IU of hCG, respectively, ovulated. The month of the breeding season, age of the mares and follicular diameter had no influence on ovulatory response. The three hCG doses used in Criollo mares (P > 0.05) result in the induction of ovulation within 48 h after injection when a pre-ovulatory follicle with a 30 to 35 mm diameter was identified. A single dose of 1000 IU of hCG is efficient to induce ovulation in Criollo mares.

Embriões bovinos produzidos in vitro foram cultivados individualmente do D7 ao D9, com o intuito ... more Embriões bovinos produzidos in vitro foram cultivados individualmente do D7 ao D9, com o intuito de avaliar o seu desenvolvimento posterior. Quarenta e nove embriões, nos estágios de blastocisto inicial, blastocisto e blastocisto expandido foram cultivados, individualmente, em 50ml de meio SOF + 5% SVE, do D7 ao D9 (D0=fecundação). Entre o D7 e D8, os blastocistos foram cultivados em palhetas (TcP) ou em placas (CP) e, entre o D8 e D9, foram cultivados apenas em placas. O índice de blastocistos que avançaram pelo menos um estágio de desenvolvimento, entre D7 e D8, foi de 71%, 37% e 44% no CP e de 100%, 66% e 36% no TcP, respectivamente para blastocistos iniciais, blastocistos e blastocistos expandidos. O percentual total de embriões que evoluíram do D7 para D8 foi de 50% (12/24) para o CP e de 60% (15/25) para o TcP, os quais não foram significativamente diferentes (P>0,05). Cento e seis mórulas de camundongos da cepa CF1 Suíço Albina, coletadas 76-78h após a administraç... more Cento e seis mórulas de camundongos da cepa CF1 Suíço Albina, coletadas 76-78h após a administração de HCG, foram congeladas em solução de congelamento contendo glicerol 2,0M + sacarose 0,25M (Grupo S) ou Glicerol 2,0M + lactose 0,25M (Grupo L), ambas em PBS modificado. Foi efetuado um pré-tratamento com Glicerol 2,0M durante 4 minutos, sendo as mórulas transferidas, posteriormente, para a solução de congelamento, durante 1 minuto. O envase foi realizado em palhetas de 0,25ml em grupos de 5 a 11 mórulas. As palhetas contendo os embriões foram colocadas em vapor de N2 durante 2 minutos e, após, mergulhados em N2 líquido. O descongelamento foi efetuado em banho-maria a 37°C por 20 segundos e foram recuperadas 103 mórulas após a diluição do crioprotetor, que foi efetuada em solução de sacarose 0,5M (Grupo S) e lactose 0,5M (Grupo L) , durante 5 minutos , à temperatura ambiente . The effect of age, follicular diameter and month of the breeding season (September to January) on... more The effect of age, follicular diameter and month of the breeding season (September to January) on the hCG induction of ovulation was evaluated using 123 Criollo mares. Age varied between two and 24 years and the animals were examined daily by rectal palpation and ultrasonography with a 5 MHz linear transducer. When ovarian follicles reached a diameter of 30 to 35 mm, ovulation was induced with an i.v. injection of 1000 IU (n = 39); 1500 IU (n = 41) or 2000 IU (n = 43) of hCG. The mares were bred the next day and examined daily until ovulation was detected. The percentage of mares ovulating before 24 h of hCG injection was 10.3%, 7.3% and 4.7%; until 48 h after injection 92.3%, 85.3% and 86.0% of the mares treated with 1000, 1500 and 2000 IU of hCG, respectively, ovulated. The month of the breeding season, age of the mares and follicular diameter had no influence on ovulatory response. The three hCG doses used in Criollo mares (P > 0.05) result in the induction of ovulation within 48 h after injection when a pre-ovulatory follicle with a 30 to 35 mm diameter was identified. A single dose of 1000 IU of hCG is efficient to induce ovulation in Criollo mares.
For in vitro embryo production following ovum-pickup groups with different number of oocytes and ... more For in vitro embryo production following ovum-pickup groups with different number of oocytes and volume of medium may be required to maintain an individualized production of each cow. Bovine oocytes were randomly allocated in a 3x3 factorial design in order to evaluate the effect of number of oocytes (5, 10 and 20) and maturation medium volume ratios (1:1, 1:5 and 1:10 oocyte/μl) on embryo production. After 22-24h maturation in TCM-199, in vitro fertilization was performed in FERT-TALP medium, for 18-22h. Groups of 20 presumptive zygotes were cultured in 200μl of SOF medium for eight days. There was no effect of interaction between number of oocytes and volume of maturation medium (P>0.05) on embryo development. Cleavage rates were not affected either by number of oocytes nor by volume of medium (P>0.05). Blastocyst rates on D7 (19, 26 and 28%; D0=day of fertilization) and D9 (23, 33 and 33%) were lower (P<0.05) in groups of 5 oocytes compared with groups of 10 and 20 oocytes, respectively. Blastocyst rates on D7 (22, 24 and 27%) and D9 (26, 31 and 32%), were similar (P>0.05) for ratios 1:1, 1:5 and 1:10 oocytes/μl, respectively. Hatching rate with a 1:1 volume ratio (26%) was lower (P<0.05) than that obtained with a 1:5 volume ratio (44%). Maturation in larger groups increases embryo production. The volume of maturation medium does not influence the blastocyst production, however a low oocyte/μl medium ratio reduces the hatching rate.

Embriões bovinos produzidos in vitro foram cultivados individualmente do D7 ao D9, com o intuito ... more Embriões bovinos produzidos in vitro foram cultivados individualmente do D7 ao D9, com o intuito de avaliar o seu desenvolvimento posterior. Quarenta e nove embriões, nos estágios de blastocisto inicial, blastocisto e blastocisto expandido foram cultivados, individualmente, em 50ml de meio SOF + 5% SVE, do D7 ao D9 (D0=fecundação). Entre o D7 e D8, os blastocistos foram cultivados em palhetas (TcP) ou em placas (CP) e, entre o D8 e D9, foram cultivados apenas em placas. O índice de blastocistos que avançaram pelo menos um estágio de desenvolvimento, entre D7 e D8, foi de 71%, 37% e 44% no CP e de 100%, 66% e 36% no TcP, respectivamente para blastocistos iniciais, blastocistos e blastocistos expandidos. O percentual total de embriões que evoluíram do D7 para D8 foi de 50% (12/24) para o CP e de 60% (15/25) para o TcP, os quais não foram significativamente diferentes (P>0,05).
