Tocotrienol-rich fraction of palm oil (TRF) is an antioxidant that is rich in tocotrienol and tocopherol. The main aim of the present study was to determine the effect of TRF against oxidative stress in pancreas of diabetic rats. Forty male Sprague Dawley rats were divided into four groups: non-diabetic (NDM), non-diabetic supplemented TRF (NDM+TRF), diabetic rats and diabetic rats supplemented TRF (diabetic+TRF). Diabetes was induced through a single intravenous injection of streptozotocin at the dose of 45 mg/kg and TRF was administered orally and daily at the dose of 200 mg/kg for 28 days. To date, a number of studies have demonstrated antioxidant properties of Hibiscus sabdariffa Linn. This study was designed to investigate the effects of H. sabdariffa UKMR-2 variety on sperm functioning of streptozotocin-induced diabetic rats. Male Sprague-Dawley rats were allotted into four groups, namely control group (C), H. sabdariffa extract (HSE) group, diabetes group (D) and diabetes plus HSE group (D+HSE). HSE (100 mg/ kg/body weight) was administered orally for 28 consecutive days. After 28-days of supplementation, the rats were sacrificed to obtain epididymal sperm. Administration of HSE significantly lowered the level of fasting blood glucose and increased plasma insulin level in D+HSE group as compared to D group (p<0.05). Reproductive dysfunction is recognized as a consequence of diabetes mellitus. In this study, the possible protective efficacy of α7lipoic acid (ALA) on sperm characteristics, peroxidative damage and antioxidant levels were assessed in the epididymal sperm as well as in testes of four different groups which includes non7diabetic without ALA, non7diabetic with ALA, diabetic without ALA and diabetic with ALA group. Diabetic groups were induced by a single intravenous injection of STZ (45 mg/kg body weight). Urb. leaves have been previously reported to exhibit antioxidant activity. The objective of the present study is to determine the in vitro antioxidant activity of the combined extracts of M. oleifera and C. asiatica (TGT-PRIMAAGE) and its effect on hydrogen peroxide (H 2 O 2)-induced oxidative stress in human dermal fibroblasts. TGT-PRIMAAGE acted on the mechanism of hydrogen transfer as it showed scavenging activity in the DPPH assay. This is due to the presence of phenolics and flavonoids in TGT-PRIMAAGE. TGT-PRIMAAGE effectively reduced cellular generation of reactive oxygen species induced by H 2 O 2. The activities of superoxide dismutase and catalase were also increased in cells treated with TGT-PRIMAAGE. Treatment with TGT-PRIMAAGE showed significant reduction (P < 0.05) in the number of senescent cells. Significant reduction (P < 0.05) of malondialdehyde was also seen in cells treated with TGT-PRIMAAGE. The p53 protein level was reduced in TGT-PRIMAAGEtreated cells, which indicates its potential in protecting the cells from oxidative stress induced by H 2 O 2 .

Meanwhile, improved antioxidant defense system was also observed in diabetic group supplemented with TRF which was demonstrated by significantly higher (p<0.05) levels of glutathione (GSH) as well as superoxide dismutase (SOD) activities as compared to diabetes rats without TRF supplementation. In conclusion, TRF was able to reduce pancreatic damage and oxidative stress in streptozotocin-induced diabetic rats and the process was probably mediated by its antioxidant effects.

Therefore, a holistic approach of treatment is urgently required to prevent worsening of complications. The present investigation was carried out to study the possible protective effects of Roselle or Hibiscus sabdariffa Linn (HSL) calyxes aqueous extract, as an antidiabetic and antioxidant agent against oxidative liver injury in streptozotocin-induced diabetic rats. A single dose of streptozotocin (45mg/kg body weight, iv) was used to induced diabetes in male Sprague Dawley rats which were then divided into two groups: Diabetic control (DC) and HSL-treated diabetic (DR) group. Normal rats were divided into normal control (NC), HSL-treated control (NR). Aqueous calyxes extract of HSL (100mg/kg/day, orally) was given for 28 consecutive days in the treated group. Weight, biochemical and histopathological (light and electron microscopic) parameters were compared in all groups. Supplementation of HSL significantly lowered the level of fasting blood glucose and increased plasma insulin level in DR group compared to DC group (p&amp;amp;amp;amp;amp;lt;0.05). Alanine aminotransaminases and aspartate aminotransferase enzymes level were found to be significantly reduced in DR compared to DC. Microscopic examination demonstrated destruction of the liver architecture, cytoplasmic vacuolation of the hepatocytes and signs of necrosis in diabetic rats. Moreover, dilatation and congestion of blood vessels with leucocytes adherence were detected. Ultrastructural study using electron microscope showed homogeneous substance accumulation in nuclear chromatin, a decrease of organelles and mitochondrial degeneration in the diabetic rats. Administration of HSL in diabetic rats causes significant decrease in hepatocyte destruction and prevented the changes associated with the diabetic condition. Thus, our findings provide a scientific rationale for the use of HSL as promising agent in preventing liver injury in diabetes.
