Colorectal cancer (CRC) remains a major public health burden worldwide, despite increased knowled... more Colorectal cancer (CRC) remains a major public health burden worldwide, despite increased knowledge on its pathogenesis and advances in therapy. We aimed to evaluate a new histological grading system based on poorly differentiated clusters (PDCs) counting-the PDCs grade (PDCs-G), and its clinicopathological and prognostic significance, compared to the World Health Organisation (WHO) grading system (WHO grade). We reviewed 71 surgical resection specimens for CRC from the Emergency County Hospital "Pius Brînzeu" Timisoara. The cases were graded using the WHO grade and the PDCs-G, with further analysis of their association with the other recognised prognostic parameters. Using the WHO grade, 9% of the analysed cases were G1, 80% G2, 11% G3, and none of the tumours was graded G4, while in the PDCs-G 16% were G1, 45% G2, and 39% G3. In multivariate analysis PDCs-G was significantly associated with the American Joint Committee on Cancer stage of the disease (AJCC stage) (p = 0.0003), depth of invasion (pT) (p = 0.0084), nodal status (LNM) (p < 0.0001), lymphovascular invasion (LVI) (p < 0.0001), perineural invasion (PNI) (p < 0.0052), and tumour border configuration (p < 0.0001). The novel grading system based on PDCs counting is an additional histological tool in the evaluation of CRC and a promising new prognostic factor for these patients.

The aim of this study was to conduct a prospective controlled study to assess student performance in learning anatomic pathology when adding three‐dimensional (3D) virtual models as adjunct teaching materials in the study of macroscopic lesions. Third‐year medical students (n = 501) enrolled at the Victor Babes University of Medicine and Pharmacy in Timisoara, Romania, were recruited to participate. Student performance was assessed through questionnaires. Students performed worse with new method, with poorer results in terms of overall (mean 77.6% ±SD 11.8% vs. 83.6% ±10.5) and individual question scores (percentage of questions with maximum score 34.6% ±25.6 vs. 47.7 ± 24.6). The most common use of automation process for Ki-67 quantification is represented by breast tumors. Aims We aimed to analyze the accuracy of digital quantification of Ki-67 expression in non-breast tumors. Methods We selected all the available Ki-67 stained slides done in 2014 at the County Emergency Hospital Timisoara, Ro- mania, excluding breast tumors. Images of the stained slides (10 images/slide on average, depending on the size of the specimen) were acquired on a Leica DMD108. We performed a retrospective study on 71 CRC patients who underwent surgery at the Emergency County Hospital, Timișoara. CRC cases were classified based on haematoxylin-eosin slides, using the conventional grading system, GBd and PDCs-G, respectively. We used two-tier and three-tier grading schemes for each system. Penile lesions are distinctly uncommon with less than 20 cases reported till now. In the present paper we describe the clinicopathological and immunohistochemical findings in a case of GCT of the penis shaft in a 31-years-old man. On physical examination the patient was found to have a small ovoid mass, 20 x 10 mm in diameter, at the left postero-lateral area of the penis' base. The mass was firm on palpation with no fixation on the neighboring tissues. The lesion was completely excised under loco-regional anesthesia. The surgical specimen was an ovoid, gray-white, elastic mass, of 10 x 5 mm. Introducere: Proteina anti-apoptotica bcl-2 este investigată extensiv în relaţie cu prognosticul ... more Introducere: Proteina anti-apoptotica bcl-2 este investigată extensiv în relaţie cu prognosticul şi cu predicţia raspunsului la terapie al carcinoamelor mamare. Lucrarea de faţă analizează, pe un lot de carcinoame mamare diagnosticate la femei în postmenopauză, expresia proteinei bcl-2 în corelaţie cu alţi factori clinici şi imunohistochimici (IHC) de prognostic. Material şi metodă: Au fost analizate 40 de carcinoame mamare diagnosticate la femei în postmenopauză. Studiul IHC s-a realizat cu anticorpii monoclonali anti-bcl2, anti-ER, anti-PR, anti-Ki-67, tehnica LSAB2, vizualizare cu DAB. Rezultate: Expresia protooncogenei bcl-2 a fost identificată în 55% din carcinoamele mamare invazive. ER si PR au fost pozitivi în 55% şi respectiv 57,5% din cazuri. 30% din carcinoame au prezentat un indice Ki-67≤10%, în timp ce restul de 70% au demonstrat un indice Ki-67 > 10%. Bcl-2 a fost exprimat în 100% din tumorile T1 şi în 64% din tumorile T1+T2. 63,2% din tumorile GI+GII au prezentat reacţie pozitivă pentru bcl-2. 77% şi 83% din tumorile ER şi respectiv PR-pozitive au fost bcl-2-pozitive. 92% din tumorile cu activitate proliferativă joasă au prezentat reacţie pozitivă pentru bcl-2. Tumorile N 0 au fost în proporţie de 67% bcl-2 pozitive, în timp ce în grupul tumorilor N+, 46% au fost bcl-2 pozitive. Concluzii: La femeile postmenopauzale cu cancer mamar, proteina bcl-2 a fost semnificativ exprimată în tumorile de dimensiuni reduse, bine/mediu diferenţiate, cu receptori hormonali prezenţi şi cu activitate proliferativă redusă. Sunt necesare studii mai largi care să stabilească utilitatea includerii markerului în panelul de anticorpi ce stabileşte profilul molecular al carcinoamelor mamare.

We assessed the immunohistochemical expression of c-erbB-2 transmembrane receptor in 15 primary l... more We assessed the immunohistochemical expression of c-erbB-2 transmembrane receptor in 15 primary lung carcinomas using the polyclonal antibody anti-human c-erbB-2 oncoprotein. Tissue fragments routinely processed (formalin-fixed and paraffin-embedded) were then investigated for c-erbB-2 oncoprotein expression using the Avidin-Biotin Complex staining method. Tumor cells showed positive c-erbB-2 expression in 86.6% of non small cell lung carcinomas (NSCLC). Positive c-erbB-2 immunoreaction correlated with the advanced stage of the disease, with lymph node involvement and high metastasis rate. The results obtained suggest that tissue overexpression of c-erbB-2 protein in NSCLC characterizes a group of tumors with an aggressive clinical-biological behavior and poor prognosis; c-erbB-2 protein can be considered a useful tumor marker in the morphological diagnosis and monitoring the clinical behavior of these neoplasms.

The patient was a 71-year-old woman complaining of vulvar itching and yellowish vaginal discharge who underwent a complete gynecological evaluation during which a suspicious grey-whitish mass on her vulva was observed. The tumor presented superficial ulceration and was located in the upper half of the labia minora and clitoris. Initially it was suspected to be a vulvar carcinoma. A biopsy was taken and a histopathological suspicion of amelanotic melanoma was rendered. The mass was radically excised and the diagnosis was confirmed using HMB-45, Melan-A and anti-S-100 protein antibodies. Malignant melanoma is readily diagnosed by the presence of melanin granules. We present one case of lip schwannoma: the patient was 25-year-old and he has presented to otorhinolaryngologist for a non-dolorous tumor on the mucosal side of his inferior lip, which was increasing in size for the last six months. The tumor had a superficial ulceration and infection. Initially it was suspected to be a papilloma. The tumor was radically removed, and the sections were stained with Hematoxylin-Eosin. Penile lesions are distinctly uncommon with less than 20 cases reported till now. In the present paper we describe the clinicopathological and immunohistochemical findings in a case of GCT of the penis shaft in a 31-years-old man. On physical examination the patient was found to have a small ovoid mass, 20 x 10 mm in diameter, at the left postero-lateral area of the penis' base. The mass was firm on palpation with no fixation on the neighboring tissues. The lesion was completely excised under loco-regional anesthesia. The surgical specimen was an ovoid, gray-white, elastic mass, of 10 x 5 mm. Microscopically, the tumor was moderately cellular and was composed of polygonal-shaped cells with abundant granular eosinophilic cytoplasm. Introducere: Proteina anti-apoptotica bcl-2 este investigată extensiv în relaţie cu prognosticul ... more Introducere: Proteina anti-apoptotica bcl-2 este investigată extensiv în relaţie cu prognosticul şi cu predicţia raspunsului la terapie al carcinoamelor mamare. Lucrarea de faţă analizează, pe un lot de carcinoame mamare diagnosticate la femei în postmenopauză, expresia proteinei bcl-2 în corelaţie cu alţi factori clinici şi imunohistochimici (IHC) de prognostic. Material şi metodă: Au fost analizate 40 de carcinoame mamare diagnosticate la femei în postmenopauză. Studiul IHC s-a realizat cu anticorpii monoclonali anti-bcl2, anti-ER, anti-PR, anti-Ki-67, tehnica LSAB2, vizualizare cu DAB. Rezultate: Expresia protooncogenei bcl-2 a fost identificată în 55% din carcinoamele mamare invazive. ER si PR au fost pozitivi în 55% şi respectiv 57,5% din cazuri. 30% din carcinoame au prezentat un indice Ki-67≤10%, în timp ce restul de 70% au demonstrat un indice Ki-67 > 10%. Bcl-2 a fost exprimat în 100% din tumorile T1 şi în 64% din tumorile T1+T2. 63,2% din tumorile GI+GII au prezentat reacţie pozitivă pentru bcl-2. 77% şi 83% din tumorile ER şi respectiv PR-pozitive au fost bcl-2-pozitive. 92% din tumorile cu activitate proliferativă joasă au prezentat reacţie pozitivă pentru bcl-2. Tumorile N 0 au fost în proporţie de 67% bcl-2 pozitive, în timp ce în grupul tumorilor N+, 46% au fost bcl-2 pozitive. Concluzii: La femeile postmenopauzale cu cancer mamar, proteina bcl-2 a fost semnificativ exprimată în tumorile de dimensiuni reduse, bine/mediu diferenţiate, cu receptori hormonali prezenţi şi cu activitate proliferativă redusă. Sunt necesare studii mai largi care să stabilească utilitatea includerii markerului în panelul de anticorpi ce stabileşte profilul molecular al carcinoamelor mamare.

We assessed the immunohistochemical expression of c-erbB-2 transmembrane receptor in 15 primary l... more We assessed the immunohistochemical expression of c-erbB-2 transmembrane receptor in 15 primary lung carcinomas using the polyclonal antibody anti-human c-erbB-2 oncoprotein. Tissue fragments routinely processed (formalin-fixed and paraffin-embedded) were then investigated for c-erbB-2 oncoprotein expression using the Avidin-Biotin Complex staining method. Tumor cells showed positive c-erbB-2 expression in 86.6% of non small cell lung carcinomas (NSCLC). Positive c-erbB-2 immunoreaction correlated with the advanced stage of the disease, with lymph node involvement and high metastasis rate. The results obtained suggest that tissue overexpression of c-erbB-2 protein in NSCLC characterizes a group of tumors with an aggressive clinical-biological behavior and poor prognosis; c-erbB-2 protein can be considered a useful tumor marker in the morphological diagnosis and monitoring the clinical behavior of these neoplasms.

Blood supply blockage can occur with or without arterial inflammation. For this reason, there are two types of AIONs: non-arteritic (NA-AION), and arteritic (A-AION), the latter is almost invariably due to giant cell arteritis (GCA). GCA is a primary vasculitis that predominantly affects extracranial medium-sized arteries, particularly the branches of the external carotid arteries (including superficial temporal arteries - TAs). We present a case of a professional handball player teenager female, with a lateral femur condylar osteochondral fracture after a fall with the right knee in extension and in internal rotation. Magnetic resonance imaging (MRI) showed an osteochondral fracture of the lateral femoral condyle, 34.6 mm on long axis, impossible to manage arthroscopically, because of the size and the location of the detached fragment. The solution was the lateral knee arthrotomy allowing the evacuation of the hemarthrosis and preparation of the fracture site, then reduction and fixation of the fracture with absorbable cannulated pins. The patient was a 71-year-old woman complaining of vulvar itching and yellowish vaginal discharge who underwent a complete gynecological evaluation during which a suspicious grey-whitish mass on her vulva was observed. The tumor presented superficial ulceration and was located in the upper half of the labia minora and clitoris. Initially it was suspected to be a vulvar carcinoma. A biopsy was taken and a histopathological suspicion of amelanotic melanoma was rendered. The mass was radically excised and the diagnosis was confirmed using HMB-45, Melan-A and anti-S-100 protein antibodies. Malignant melanoma is readily diagnosed by the presence of melanin granules. Although amelanotic melanoma contains a few melanin granules, it is often difficult to differentiate it from other non-epithelial malignant tumors. The diagnosis is typically made at the time of surgery following biopsy and surgical resection is the mainstay of treatment. We present one case of lip schwannoma: the patient was 25-year-old and he has presented to otorhinolaryngologist for a non-dolorous tumor on the mucosal side of his inferior lip, which was increasing in size for the last six months. The tumor had a superficial ulceration and infection. Initially it was suspected to be a papilloma. The tumor was radically removed, and the sections were stained with Hematoxylin-Eosin. The tumor was encapsulated and showed two different pattern of growth. Antoni A areas displayed spindle cells closely packed together with palisading of nuclei.
