Introducción: La población utiliza la medicina a base de hierbas de forma indiscriminada basándos... more Introducción: La población utiliza la medicina a base de hierbas de forma indiscriminada basándose en la creencia de que las plantas medicinales carecen de efectos adversos. Objetivo: Determinar in vivo el efecto toxicológico y analgésico del extracto clorofórmico de las hojas de Calea urticifolia. Metodología: El estudio toxicológico fue realizado mediante la prueba de toxicidad subcrónica de 90 días, a dosis repetidas y continuas en ratones NIH. Se realizaron análisis de bioquímica sanguínea, hematología y el examen histopatológico de órganos. La actividad analgésica fue evaluada con el modelo in vivo de contorsiones abdominales. Resultados: La administración del extracto vegetal provocó la aparición de signos clínicos de toxicidad, alteraciones en los parámetros hematólogos y bioquímica sanguínea, además alteraciones histológicas en algunos de los órganos. Decidimos que se podia simplificar esta tarea si el profesorado comparte el material que elabora. Hace un par de anos, profesores de Argentina, Espana y Uruguay recopilamos una coleccion de problemas y los publicamos en Montevideo y en Internet, bajo licencia "Creative Commons". En esta red pretendiamos editar otro libro, con mas problemas, invitando a profesores de mas paises. En el momento de cerrar esta red, tenemos una coleccion de 31 problemas, escritos por profesores de seis paises y el compromiso de participacion de profesores de otros cinco paises mas. Medicinal plants constitute an option, and the leaves of Prosopis ruscifolia (Pr) were shown to be effective in reducing glycemia in hyperglycemic animals. Objective: In this paper, we report the effect of P. rusciofolia (Pr) on insulin and incretin secretion in alloxan-induced hyperglycemic rats. Methodology: The effective dose was selected, and four groups (n=10) of Wistar rats were used. Two groups with normal glycemia received water or Pr (75 mg/Kg, per os, p.o.), and two groups with hyperglycemia induced by alloxan (intraperitoneal, ip), received water or Pr (75 mg/Kg, p.o.) for 2 weeks. Oral glucose tolerance test, and incretin and insulin levels were measured at the end of the experimental period. La Diabetes Mellitus (DM) es una patología crónica de origen metabólico, que aparece debido a que... more La Diabetes Mellitus (DM) es una patología crónica de origen metabólico, que aparece debido a que el páncreas no produce la cantidad de insulina que el cuerpo humano necesita, o bien de una calidad inferior. En este trabajo pretendimos definir el efecto del consumo de la miel de abeja sobre la glucemia en ratones macho sanos e hiperglucémicos inducidos a diabetes por aloxano. Para ello se emplearon cuatro grupos de seis ratones albinos suizos machos cada uno. normoglucémicos con dieta regular tratados con agua; normoglucémicos con dieta + miel de abeja; hiperglicémicos con dieta regular tratados con agua y finalmente, hiperglicémicos con aloxano con dieta + miel. El tratamiento se realizó por tres días. They represent the first examples of a triterpeneeneolignan ester and a heptacyclic arylpropanoid-nor-triterpenephenol, hetero-DielseAlder adducts built with dioxane bridges. Their proposed biosynthetic route is discussed.

A new bicyclic diterpene with a labdane skeleton, 7-oxo-labd-8en-15-ol (1), along with two known ... more A new bicyclic diterpene with a labdane skeleton, 7-oxo-labd-8en-15-ol (1), along with two known diterpenes and ten flavonoids were isolated from the leaves of Aeonium lindleyi (Crassulaceae). Their structures were elucidated on the basis of spectroscopic data, including 1D and 2D NMR experiments, and comparison with spectroscopic data reported in the literature. Labdan-8α,15-diol (2) and labd-8(17)-en-3β,15-diol (3) showed leishmanicidal activity against Leishmania tropica (IC 50 = 77.0 µM) and Leishmania braziliensis (IC 50 = 68.0 µM) similar to ketoconazole used as positive control. 5,3′-Dihydroxy-3,7,4′,5′-tetramethoxyflavone (8) and combretol (9) showed moderate activity (growth inhibition 87.3 and 73.0 %, respectively, at 50 µM) against a multidrug-resistant L. tropica line.

Parasite resistance to drugs has emerged as a major problem in current medicine, and therefore, t... more Parasite resistance to drugs has emerged as a major problem in current medicine, and therefore, there is great clinical interest in developing compounds that overcome these resistances. In an intensive study of South American medicinal plants, herein we report the isolation, structure elucidation, and biological activity of dihydro-beta-agarofuran sesquiterpenes from the roots of Maytenus magellanica (1-14) and M. chubutensis (14-17). This type of natural products may be considered as privileged structures. The structures of 10 new compounds, 1, 3, 6-9, and12-15, were determined by means of (1)H and (13)C NMR spectroscopic studies, including homonuclear (COSY and ROESY) and heteronuclear correlation experiments (HMQC and HMBC). The absolute configurations of eight hetero- and homochromophoric compounds, 1, 3,6-9, 12, and 13, were determined by means of CD studies. Fourteen compounds, 1-3 and 6-16, have been tested on a multidrug-resistant Leishmania tropica line overexpressing a P-glycoprotein-like transporter to determine their ability to revert the resistance phenotype and to modulate intracellular drug accumulation. From this series, 1, 2, 3, 14, and 15 showed potent activity, 1 being the most active compound. The structure-activity relationships of the different compounds are discussed.
