The materials used are garlic, coconut water and nutrient minimal macro and micro. The result was the establishment of direct induction of green protocorm from garlic explants. Conclusion of this research is the addition of 15% coconut water in medium MS minimal macro and micro nutrient can induce formation of protocorm directly from garlic explants.

The aim of this research is a study about direct somatic embryogenesis from mature seed and plant... more The aim of this research is a study about direct somatic embryogenesis from mature seed and plantlet regeneration of Citrus nobilis variety SoE. Plant material for somatic embryogenesis is the mature seed of Tangerine SoE. 6-benzylaminopurine (BAP) 3 mg/L supplemented to Murashige and Skoog (MS) modified medium was induced direct somatic embryos (SE) 4 weeks after treatments. MS modified medium with single BAP 3 mg/L has resulted the average of eight SE. Nevertheless, average number of SE which resulted from combination BAP 3 mg/L with some level of concentration 2.4-D (0.1, 0.3, and 0.5 mg/L) decreases with increasing the concentration of 2.4-D. Abscisic acid (ABA) is added at concentration 0.5-3.5 mg/L cannot increase the number of SE, globular early-stage, at 4 and 8 weeks. Gibberellic acid (GA3) added at concentration 2.5 mg/L can increase the growth in the number of normal plantlets this citrus at 8 weeks. Keragaman genetik jeruk ini dapat diinduksi dengan iradiasi sinar gamma pada sel-sel kalus embriogenik untuk menghasilkan mutan yang solid. Deteksi keragaman genetik yang terbentuk dapat dilakukan secara morfologi maupun molekuler dengan marka ISSR. Tujuan Penelitian ini adalah untuk mendapatkan informasi mengenai keragaman genetik yang terjadi pada mutan harapan jeruk keprok SoE hasil iradiasi sinar gamma berdasarkan morfologi dan marka ISSR. Marka ISSR yang digunakan adalah ISSR 1, 4, 6 dan 8 pada beberapa mutan harapan jeruk keprok SoE. Analisis keragaman secara morfologi menghasilkan dendrogram dengan tingkat kemiripan antar individu masing-masing dosis iradiasi 83-95% dengan jarak genetik 5-17%. Dendrogram analisis keragaman genetik berdasar marka ISSR memperlihatkan tingkat kemiripan 51-100% dan jarak genetik 0-49%. Individu-individu sampel yang diuji hasil iradiasi gamma, baik secara morfologi dan marka ISSR, telah memiliki susunan genetik yang berbeda dari susunan genetik individu sebelum diiradiasi.

The regenerated plants were acclimatize under the 60% sunlight block and to keep the humidity remains high the plants were lid with UV plastic. The results indicated that the plant regeneration via somatic embryogenesis from mature seed of Citrus nobilis an efficient and effective technique for producing more somatic embryos and plant production in large scale. The next sub-cultured in medium containing GA3 and then half-strength concentration of medium can increases the number of normal and enlargement plantlets, respectively.

The aims of this study was 1) Found out the effect of 0-20 ppm colchicine concentration on the gr... more The aims of this study was 1) Found out the effect of 0-20 ppm colchicine concentration on the growth of the cayenne pepper seedlings of Malita FM varieties, 2) Found out the phenotype diversity of the cayenne pepper seedlings of Malita FM varieties of putative mutant M1 (MHM1) as the results of mutation induction with colchicine 0-20 ppm,
3) Obtained a value of Lethal Concentration 50% (LC50) from 0-20 ppm colchicine for cayenne pepper of Malita FM varieties. The research was conducted from June to August 2019 at the screenhouse of Department of Agrotechnology, Faculty of Agriculture, Gorontalo State University. The experimental design used a randomized block design (RBD) with 5 levels of treatment of colchicine mutagen, namely 0; 5; 10; 15 and 20 ppm were repeated 3 times, so there were 15 experimental units. Each unit of experiment contained 5 samples. The results showed that 1) Colchicine mutagen given at a concentration of 0-20 ppm has not significantly different effect on the growth of cayenne pepper seedlings of Malita FM variety between treatments; 2) The diversity of the phenotypes of the cayenne pepper seedlings of the Malita FM varieties resulted from mutation induction (Putative Mutant M1 (MHM1)) with a colchicine solution of 0-20 ppm concentration was low. The level of diversity obtained only reached 17-26%. Colchicine mutagen with this concentration has not been able to induced genetic changes in the cayenne pepper of Malita FM varieties permanently; and 3) Value of Lethal Concentration 50% (LC50) for the cayenne pepper plants of the Malita FM varieties induced by colchicine mutagen has not been obtained at a range concentration of 0-20 ppm.

pertumbuhan, perkembangan dan produksi tanaman. Berdasarkan permasalahan salinitas
maka perlu dilakukan upaya pemilihan jenis tanaman yang tahan (toleran) pada kondisi
cekaman salinitas. Penelitian ini bertujuan untuk mengetahui ketahanan tanaman okra hijau
(Abelmoschus esculentus L.) varietas Naila IPB terhadap cekaman salinitas melalui
pemberian garam NaCl. Penelitian ini dilaksanakan di Desa Dunggala Kecamatan Tapa
Kabupaten Bone Bolango pada bulan Mei sampai bulan Agustus 2017. Penelitian
menggunakan rancangan acak lengkap (RAL) dengan 5 perlakuan konsentrasi garam NaCl:
0, 1, 2, 3.5, 4%. Parameter yang diamati adalah pertambahan jumlah daun, luas daun, waktu
berbunga, jumlah bunga, jumlah stomata, jumlah buah, berat basah akar, volume akar, berat
basah tanaman dan berat tanaman kering. Analisis data menggunakan sidik ragam ANOVA
dengan uji BNT 5%. Hasil penelitian menunjukkan bahwa tanaman okra tahan terhadap
salinitas melalui pemberian NaCl hingga konsentrasi 4%. Peningkatan konsentrasi NaCl
berpengaruh nyata terhadap jumlah stomata, volume akar dan berat tanaman kering.

The experimental design used a randomized block design (RBD) with 5 levels of treatment of colchicine mutagen, namely 0; 5; 10; 15 and 20 ppm were repeated 3 times, so there were 15 experimental units. Each unit of experiment contained 5 samples. The results showed that 1) Colchicine mutagen given at a concentration of 0-20 ppm has not significantly different effect on the growth of cayenne pepper seedlings of Malita FM variety between treatments; 2) The diversity of the phenotypes of the cayenne pepper seedlings of the Malita FM varieties resulted from mutation induction (Putative Mutant M1 (MHM1)) with a colchicine solution of 0-20 ppm concentration was low. The level of diversity obtained only reached 17-26%. Colchicine mutagen with this concentration has not been able to induced genetic changes in the cayenne pepper of Malita FM varieties permanently; and 3) Value of Lethal Concentration 50% (LC50) for the cayenne pepper plants of the Malita FM varieties induced by colchicine mutagen has not been obtained at a range concentration of 0-20 ppm.

pertumbuhan, perkembangan dan produksi tanaman. Berdasarkan permasalahan salinitas
maka perlu dilakukan upaya pemilihan jenis tanaman yang tahan (toleran) pada kondisi
cekaman salinitas. Penelitian ini bertujuan untuk mengetahui ketahanan tanaman okra hijau
(Abelmoschus esculentus L.) varietas Naila IPB terhadap cekaman salinitas melalui
pemberian garam NaCl. Penelitian ini dilaksanakan di Desa Dunggala Kecamatan Tapa
Kabupaten Bone Bolango pada bulan Mei sampai bulan Agustus 2017. Penelitian
menggunakan rancangan acak lengkap (RAL) dengan 5 perlakuan konsentrasi garam NaCl:
0, 1, 2, 3.5, 4%. Parameter yang diamati adalah pertambahan jumlah daun, luas daun, waktu
berbunga, jumlah bunga, jumlah stomata, jumlah buah, berat basah akar, volume akar, berat
basah tanaman dan berat tanaman kering. Analisis data menggunakan sidik ragam ANOVA
dengan uji BNT 5%. Hasil penelitian menunjukkan bahwa tanaman okra tahan terhadap
salinitas melalui pemberian NaCl hingga konsentrasi 4%. Peningkatan konsentrasi NaCl
berpengaruh nyata terhadap jumlah stomata, volume akar dan berat tanaman kering.
