In addition to the written exam a novel "oral-practical" part is required. Here we describe the implications of this oral-practical exam on the examination procedures in the field of biochemistry/molecular biology. A strategy for its realization is proposed.

In blowflies it was detected mainly in eggs and pupae. The enzyme activity in blowflies varies during development parallel to the ecdysteroid content. This suggests a specific but presently unknown function of the enzyme. Ecdysone oxidase was located mainly in the fat body and gut of blowflies. It is a cytosolic enzyme.

Antiserum DLW has the highest antibody concentration and antiserum H-21B is most sensitive for ecdysone. Antisera H-21B and ICT-1 show a high specificity for the ecdysteroid nucleus, whereas antiserum DLW is more specific for the side chain of ecdysteroids. This can be derived from cross reaction factors of ten different ecdysteroids. No antiserum is specific enough to detect ecdysone exclusively in extracts from biological material. Ecdysterone and other ecdysone metabolites are detected likewise depending on their concentration and affinity to the antibodies. It is shown that the radioimmunoassay of biological samples can lead to an overestimation of the ecdysone concentration.

The enzymes involved in this metabolic step were characterized with respect to their cosubstrate specificities, inhibitor susceptibilities, and immunological crossreactivities with antibodies directed against Abbre6iations: MDR, medium-chain dehydrogenases/reductases; SDR, short-chain dehydrogenases/reductases; AKR, aldo-keto reductases; HSD, hydroxysteroid dehydrogenase; NKI, Nö venyvedelmi Kutatointezet (Plant Protection Research Institute).
