Sessoes agudas promovem respostas no sistema imunologico, podendo acarretar desistencia, nao cont... more Sessoes agudas promovem respostas no sistema imunologico, podendo acarretar desistencia, nao continuidade ou queda no desempenho nos programas de exercicios, pela aumentada susceptibilidade as infeccoes das vias aereas superiores (IVASs). Considerando que os linfocitos desempenham funcoes no combate as infeccoes e que individuos sedentarios com baixo nivel de aptidao fisica, devam iniciar programas de treinamento com baixo volume e intensidade, o estudo teve como objetivo avaliar a contagem de leucocitos e linfocitos circulantes e dos linfocitos teciduais mesentericos, bem como a capacidade proliferativa dos linfocitos teciduais em ratos machos "Wistar" submetidos a sessoes agudas de cinco e 15 minutos nas intensidades leve e moderada. A incidência de doenças cardiovasculares e obesidade estão associada ao aumento da ingestão de al... more A incidência de doenças cardiovasculares e obesidade estão associada ao aumento da ingestão de alimentos hipercalóricos em decorrência de modificações no padrão alimentar ocorrido nas últimas décadas. A dieta hiperlipídica administrada a gestantes induz, nos descendentes, aumento da pressão sanguínea, resistência à insulina, dislipidemia e obesidade. Estes são sintomas característicos da Síndrome Metabólica que estão sendo descritos cada vez mais em crianças e adolescentes. Neste trabalho foi analisado informações obtidas dos bancos de dados bibliográficos (Scielo, Medline, PubMed) enfocando pesquisas sobre nutrição materna durante a gravidez e lactação e a influência destes fatores nutricionais no desenvolvimento dos conceptos. The purpose of this study was to verify the histophysiological alterations in monocytes and macro... more The purpose of this study was to verify the histophysiological alterations in monocytes and macrophages induced by short periods of exercise. Male Wistar rats (age = 2 months, body weight = 200g) were divided into seven groups (N = 6 each): sedentary control (C), groups exercised (swimming) at low intensity for 5 (5L), 10 (10L), and 15 minutes (15L), and groups exercised at moderate intensity for 5 (5M), 10 (10M) or 15 minutes (15M). At moderate intensity the animals carried a load of 5% of body weight on their backs. Blood monocytes were evaluated for quantity and morphology, and peritoneal macrophages were analyzed for quantity and phagocytic activity. Data were analyzed using ANOVA and Tukey's post hoc test (p ≤ 0.05). Low intensity groups and 5M exhibited an increase in monocyte levels when compared with the control. O presente estudo objetivou investigar as respostas imunes frente a duas sessões de exercício, re... more O presente estudo objetivou investigar as respostas imunes frente a duas sessões de exercício, realizadas no mesmo dia. Foram utilizados ratos machos Wistar, divididos em grupo controle, animais sedentários (C; n=8); grupo exercitado de forma aguda em intensidade leve por 60min (L60; n=8); grupo exercitado de forma aguda em intensidade moderada por 15min (M15; n=8) e grupo exercitado em duas sessões no mesmo dia (2S; n=8). O grupo que realizou duas sessões no mesmo dia demonstrou leucocitose 23% maior do que o grupo leve de 60min e 15% maior do que o grupo moderado de 15min, elevação nos neutrófilos quando comparado aos grupos exercitados por uma sessão e diminuição no percentual de linfócitos circulantes em relação aos mesmos grupos (p < 0,05). Os resultados mostraram que uma segunda sessão de exercício, realizada no mesmo dia, promoveu diferentes respostas na mobilização das subpopulações de leucócitos para a circulação geral, quando comparado com uma única sessão de exercício. We used male Wistar rats, divided in control group, sedentary animals (C; n=8); acute 60 minutes low intensity exercised group (L60; n=8); acute 15 minutes moderate intensity exercised group (M15; n=8) and group exercised for two sessions in the same day (2S; n=8). The two sessions exercised group showed 23% greater leukocytosis as compared to low intensity 60 minutes group and 15% greater as compared to moderate 15 minutes group, increase in neutrophils related to groups exercised in one session and decrease in lymphocytes percentage in relation to the same groups (p < 0,05). The results showed that a second exercise session performed in the same day promoted different responses in the mobilization of leukocytes subpopulations to general circulation as compared to only one exercise session.

O objetivo deste estudo foi avaliar o efeito da suplementação crônica de farelo de aveia (fonte de fibras solúveis) sobre as células do sistema imunológico em ratos treinados, frente a um teste de exaustão. Foram utilizados ratos Wistar, ± dois meses, peso ± 200g, divididos em três grupos (n = 9, cada um): 1) controle sedentário (C); 2) treinado oito semanas submetido ao teste de exaustão (EX); e 3) treinado oito semanas submetido ao teste de exaustão com suplementação de 30% de farelo de aveia (EXA). O treinamento consistiu de 60 minutos de natação diários, cinco dias por semana durante oito semanas. The aim of this study was to evaluate the effect of oat bran supplementation on time to exhaustio... more The aim of this study was to evaluate the effect of oat bran supplementation on time to exhaustion, glycogen stores and cytokines in rats submitted to training. The animals were divided into 3 groups: sedentary control group (C), an exercise group that received a control chow (EX) and an exercise group that received a chow supplemented with oat bran (EX-O). Exercised groups were submitted to an eight weeks swimming training protocol. In the last training session, the animals performed exercise to exhaustion, (e.g. incapable to continue the exercise). After the euthanasia of the animals, blood, muscle and hepatic tissue were collected. Plasma cytokines and corticosterone were evaluated. Glycogen concentrations was measured in the soleus and gastrocnemius muscles, and liver. Glycogen synthetase-α gene expression was evaluated in the soleus muscle. Statistical analysis was performed using a factorial ANOVA. Time to exhaustion of the EX-O group was 20% higher (515 ± 3 minutes) when compared with EX group (425 ± 3 minutes) (p = 0.034). For hepatic glycogen, the EX-O group had a 67% higher concentrations when compared with EX (p = 0.022). In the soleus muscle, EX-O group presented a 59.4% higher glycogen concentrations when compared with EX group (p = 0.021). TNF-α was decreased, IL-6, IL-10 and corticosterone increased after exercise, and EX-O presented lower levels of IL-6, IL-10 and corticosterone levels in comparison with EX group. It was concluded that the chow rich in oat bran increase muscle and hepatic glycogen concentrations. The higher glycogen storage may improve endurance performance during training and competitions, and a lower post-exercise inflammatory response can accelerate recovery.

Changes in the integrity, ultrastructure, phagocytosis capacity, and production of H2O2, O2.- and... more Changes in the integrity, ultrastructure, phagocytosis capacity, and production of H2O2, O2.- and NO2- were evaluated in cultured neutrophils. The activities of the antioxidant enzymes (catalase-CAT, superoxide dismutase-SOD and glutathione-dependent peroxidase-GSH-Px) were measured under similar conditions. The integrity of the cells remained unchanged up to 18 h. After 24 h, the number of viable cells in culture dropped by 16 per cent. The percentage of viable cells in culture was of 72 per cent even after 72 h. An ultrastructural analysis of the cells was carried out after 3, 6, 12, 24, 48, and 72 h in culture. Neutrophils started developing morphologic changes after 24 h: decreased cell volume, abundant vacuoles (mainly around the nucleus), and also the presence of autophagic vacuoles. This period was then chosen for the study of neutrophil function and antioxidant enzyme activities. Neutrophils cultured for 24 h presented reduced phagocytosis capacity. The rates of production of H2O2 and O2.- remained unchanged after 24 h in culture. Concomitantly, these cells were also able to produce NO in significant amounts. The production of O2.- in response to PMA stimulus was lowered in 24-h cultured cells. Possibly, the production of oxygen and nitrogen reactive species accomplished with a decrease in the activities of CAT and GSH-Px play a key role for the process of apoptosis which takes place in neutrophils under these conditions.

The effects of the supplementation of aspartic acid and asparagine (45 mg.kg1 body weight of each... more The effects of the supplementation of aspartic acid and asparagine (45 mg.kg1 body weight of each), and carnitine (90 mg.kg-1 body weight) during one week on the ultrastructure of soleus muscle from swimming-trained (five weeks) and sedentary rats were examined. In trained rats, the amino acids supplementation was performed during the last week of the exercise training only. Supplementation of these amino acids in the diet either in sedentary and trained rats caused myofibrillar and mitochondrial disorganization and dissolution. Focal degeneration of myofibrils and Z-line streaming and disruption, as well as internalization of nuclei were observed. The size of mitochondria increased and some of them presented severe swelling, with decreased electron-density of the matrix and disruption of internal and external membranes. The aim of this study was to investigate the effect of CAPE on the insulin signaling and inflamma... more The aim of this study was to investigate the effect of CAPE on the insulin signaling and inflammatory pathway in the liver of mice with high fat diet induced obesity. Swiss mice were fed with standard chow or high-fat diet for 12-week. After the eighth week, animals in the HFD group with serum glucose levels higher than 200 mg/dL were divided into two groups, HFD and HFD receiving 30 mg/kg of CAPE for 4 weeks. After 12 weeks, the blood samples could be collected and liver tissue extracted for hormonal and biochemical measurements, and insulin signaling and inflammatory pathway analyzes. The high-fat diet group exhibited more weight gain, glucose intolerance, and hepatic steatosis compared with standard diet group. The CAPE treatment showed improvement in glucose sensitivity characterized by an area under glucose curve similar to the control group in an oral glucose tolerance test Furthermore, CAPE treatment promoted amelioration in hepatic steatosis compared with the high-fat diet group. The increase in glucose sensitivity was associated with the improvement in insulin-stimulated phosphorylation of the insulin receptor substrate-2, followed by an increase in Akt phosphorylation. In addition, it was observed that CAPE reduced the induction of the inflammatory pathway, c-jun-N- terminal kinase, the nuclear factor kappa B, and cyclooxygenase-2 expression, respectively. Overall, these findings indicate that CAPE exhibited anti-inflammatory activity that partly restores normal metabolism, reduces the molecular changes observed in obesity and insulin resistance, and therefore has a potential as a therapeutic agent in obesity.

The production of O2.- in response to PMA stimulus was lowered in 24-h cultured cells. Possibly, the production of oxygen and nitrogen reactive species accomplished with a decrease in the activities of CAT and GSH-Px play a key role for the process of apoptosis which takes place in neutrophils under these conditions.

The effects of the supplementation of aspartic acid and asparagine (45 mg.kg1 body weight of each... more The effects of the supplementation of aspartic acid and asparagine (45 mg.kg1 body weight of each), and carnitine (90 mg.kg-1 body weight) during one week on the ultrastructure of soleus muscle from swimming-trained (five weeks) and sedentary rats were examined. In trained rats, the amino acids supplementation was performed during the last week of the exercise training only. Supplementation of these amino acids in the diet either in sedentary and trained rats caused myofibrillar and mitochondrial disorganization and dissolution. Focal degeneration of myofibrils and Z-line streaming and disruption, as well as internalization of nuclei were observed. The size of mitochondria increased and some of them presented severe swelling, with decreased electron-density of the matrix and disruption of internal and external membranes. The aim of this study was to investigate the effect of CAPE on the insulin signaling and inflamma... more The aim of this study was to investigate the effect of CAPE on the insulin signaling and inflammatory pathway in the liver of mice with high fat diet induced obesity. Swiss mice were fed with standard chow or high-fat diet for 12-week. After the eighth week, animals in the HFD group with serum glucose levels higher than 200 mg/dL were divided into two groups, HFD and HFD receiving 30 mg/kg of CAPE for 4 weeks. After 12 weeks, the blood samples could be collected and liver tissue extracted for hormonal and biochemical measurements, and insulin signaling and inflammatory pathway analyzes. The high-fat diet group exhibited more weight gain, glucose intolerance, and hepatic steatosis compared with standard diet group. The CAPE treatment showed improvement in glucose sensitivity characterized by an area under glucose curve similar to the control group in an oral glucose tolerance test Furthermore, CAPE treatment promoted amelioration in hepatic steatosis compared with the high-fat diet group. The increase in glucose sensitivity was associated with the improvement in insulin-stimulated phosphorylation of the insulin receptor substrate-2, followed by an increase in Akt phosphorylation. In addition, it was observed that CAPE reduced the induction of the inflammatory pathway, c-jun-N- terminal kinase, the nuclear factor kappa B, and cyclooxygenase-2 expression, respectively. Overall, these findings indicate that CAPE exhibited anti-inflammatory activity that partly restores normal metabolism, reduces the molecular changes observed in obesity and insulin resistance, and therefore has a potential as a therapeutic agent in obesity.

The aim of this study was to evaluate the effect of oat bran supplementation on time to exhaustio... more The aim of this study was to evaluate the effect of oat bran supplementation on time to exhaustion, glycogen stores and cytokines in rats submitted to training. The animals were divided into 3 groups: sedentary control group (C), an exercise group that received a control chow (EX) and an exercise group that received a chow supplemented with oat bran (EX-O). Exercised groups were submitted to an eight weeks swimming training protocol. In the last training session, the animals performed exercise to exhaustion, (e.g. incapable to continue the exercise). After the euthanasia of the animals, blood, muscle and hepatic tissue were collected. Plasma cytokines and corticosterone were evaluated. Glycogen concentrations was measured in the soleus and gastrocnemius muscles, and liver. Glycogen synthetase-α gene expression was evaluated in the soleus muscle. Statistical analysis was performed using a factorial ANOVA. Time to exhaustion of the EX-O group was 20% higher (515 ± 3 minutes) when compared with EX group (425 ± 3 minutes) (p = 0.034). For hepatic glycogen, the EX-O group had a 67% higher concentrations when compared with EX (p = 0.022). In the soleus muscle, EX-O group presented a 59.4% higher glycogen concentrations when compared with EX group (p = 0.021). TNF-α was decreased, IL-6, IL-10 and corticosterone increased after exercise, and EX-O presented lower levels of IL-6, IL-10 and corticosterone levels in comparison with EX group. It was concluded that the chow rich in oat bran increase muscle and hepatic glycogen concentrations. The higher glycogen storage may improve endurance performance during training and competitions, and a lower post-exercise inflammatory response can accelerate recovery.
