The present study describes how mass spectrometry was extensively applied to the characterization... more The present study describes how mass spectrometry was extensively applied to the characterization and quantification of modified amino acids within the polypeptide chain of Angiotensin I, chosen as model substrate, combining the use of fast atom bombardment mass spectrometry with gas chromatography--mass spectrometry. The reaction products after in vitro incubation of Angiotensin I with styrene oxide, a well known carcinogen, under different conditions, have been characterized: a prominent reactivity of several potential nucleophilic sites of Angiotensin I was shown, including two histidine residues and a tyrosine residue; it is worth noting that it has never been stated that tyrosine is highly reactive with styrene oxide. The results obtained demonstrate the usefulness of mass spectrometry for the structural determination of chemically modified amino acids in peptides and proteins, and the presence of a reliable relationship between reaction conditions and the production of alkylated amino acids. This characterization procedure offers the possibility of identifying reactive sites following exposure to unknown alkylating agents.

The present study investigated the epirubicin (EPI) permeability of various commercially availabl... more The present study investigated the epirubicin (EPI) permeability of various commercially available glove types, as well as toxicity mechanisms and effects on human keratinocyte cell line (HaCaT). Permeability experiments were carried out on various commercially available gloves, differing as regards material and thickness. Permeability was evaluated after different ''contact times'' and the influence of EPI solution's pH (acid and neutral) on permeability was also examined. Toxicity of EPI toward skin was tested by evaluating the effects of the drug on cell growth and apoptosis, by using an in vitro model based on cultured immortalized human keratinocytes. No permeation was detected in the case of EPI neutral solutions; in contrast, acid solutions were found to penetrate low thickness nitrile gloves. Obtained results also showed the induction of apoptosis in epithelial cells through the activation of intrinsic pathway p53-independent occurring even when cells are exposed at low drug concentration. EPI solution's pH influences the glove's permeability; once penetrated, EPI at concentrations lower than those able to penetrate the nitrile glove during the 8-h work-shift can cause apoptosis in epithelial cells. The findings reported here highly support the choice of either natural rubbers gloves or high thickness nitrile ones for preventing the occupational exposure to EPI Q2 .

We describe a method for extraction of lipids from freeze dried homogenized stools and identification of the hydroxy stearic acid in the fecal lipids pattern by GC-MS. Recovery of 90-95 % shows the efficacy of extraction . Quantitative data of fecal hydroxy stearic acid excretion in some normal and chronic diarrhea affected children are reported.

A simple and suitable method of breath sampling and the gas chromatographic determination of H2 a... more A simple and suitable method of breath sampling and the gas chromatographic determination of H2 and CO in breath of children as well as of adults are described. The analytical system allows the measurement of concentrations of H2 and CO as low as a few ppm. The proposed method of breath collection (tedlar bag with low resistance valve face mask) appears particularly useful for long storage purposes, because it eliminates the need for additional manipulations and the possible associated sample dilution. Normalization of H2 breath amounts to internal standard O2 to obtain reliable and precise measurements is particularly useful in non-collaborating patients whose observed H2 peak values increase by 15% after the correction. The overall procedure is fast, inexpensive and accurately recognizes adult or children lactose malabsorbers as well as subjects exposed even to low levels of CO.

The globin chain components of Sprague-Dawley rat hemoglobin were obtained by reverse-phase HPLC which showed the presence of two alpha-chain and four beta-chains. 2. The accurate molecular weight of each globin chain was determined by means of electrospray mass spectrometry. Extensive mass spectrometric analysis on several enzymatic digests by fast atom bombardment mass spectrometry (FAB-overlapping) meant to determine the complete sequence of the alpha-major and of the four beta-globins. 3. The primary structure of the alpha-major globin was found in agreement with literature data (Garrick et al., 1975 Biochem. J. 149, 245-258; Chua et al., 1987). 4. Sequence analysis of the four beta-globin chains showed that amino acid differences are restricted to two protein portions: the region 22-25 and 123-125, the remaining portions of the molecule being unchanged in the four globins. Furthermore, all the amino acid replacements correspond to single point DNA mutations and (with the exception of the substitution Asp 22-->Asn in the beta 2-globin) involve uncharged substitutions.

The structural characterisation of adducts formed by the in vitro reaction of haemoglobin (Hb) wi... more The structural characterisation of adducts formed by the in vitro reaction of haemoglobin (Hb) with styrene oxide (SO), the most reactive metabolite of the industrial reagent styrene, was obtained by liquid chromatography/electrospray ionisation mass spectrometry (LC/ES-MS) analysis of modified tryptic peptides of human Hb chains. The reactive sites of human Hb towards SO were identified through characterisation of alkylated tryptic peptides by matrix-assisted laser desorption/ionisation with tandem mass spectrometry (MALDI-MS/MS). A procedure was set up based on this characterisation, allowing Hb modification to be assessed by monitoring SO/Hb adducts using HPLC with selected ion recording (SIR) mass spectrometry. By this methodology it was also possible to compare advantages and disadvantages of presently available strategies for the measurement of Hb adducts with SO. The results obtained could most plausibly lead to the optimisation of molecular dosimetry of SO adducts, and the analytical procedure described herein could be applied to the biological monitoring of styrene exposure in the workplace.

According to Italian laws (legge 125/01, art. 15, comma 1) about safety on workplaces, it'... more According to Italian laws (legge 125/01, art. 15, comma 1) about safety on workplaces, it's forbidden alcohol drinking and taking for health workers (art. 8, comma 6, legge 5/06/2003 number 131) (e.g. anaesthesiology, surgery, radiology, nursing etc.). National alcoholism risk prevention program to assess number for year of accidents in health workplaces, is actually in progress leaded by Health Minister and Regional administration (e.g. Tuscany). Particularly, prevention program acts consist in comprehensive informations for health workers and their co- workers, about prevention and identification of alcoholism- related risks. To assess the size of alcohol problems in health workplaces, Occupational Health Medicine Institute of Second University of Naples is going to enrol 500 subjects (200 nursing students, 200 Specialist Registrars of different areas, 100 Consultants). Actually our population consists in 160 Specialist Registrars of different medical subjects (surgery, clinical and laboratory). To verify the importance of alcoholism risk perception, a not validated questionnaire was administrated. This original assessing instrument consists in 23 items with closed answers about demographic information, university, career, lifestyle, knowledge about rules taking alcoholic drinks. Results show a dangerous lack of information about alcohol health effects, even in Medical Specialist Registrars.

Le particelle ultrafini (PM 0.1 : diametro aerodinamico medio inferiore a 0.1 µm) sono complesse miscele di sostanze organiche ed inorganiche, dotate di intrinseca tossicità. Le loro piccole dimensioni le rendono degne di particolare attenzione in quanto sono in grado di penetrare fino alle vie aeree più profonde, sfuggendo ai normali meccanismi di difesa polmonare, ed entrare nel circolo sanguigno (1-6).

Tuberculosis (TBC) is an infectious disease with the highest mortality and morbidity by single pa... more Tuberculosis (TBC) is an infectious disease with the highest mortality and morbidity by single pathogen, affecting about one third of worldwide population. Although Mantoux test is the most used, IGRA (Interferon-gamma Release Assays) tests seem to give good results for presumptive diagnosis of active or latent tuberculosis. From June 2011 to June 2012 we made about 1,000 visits for TBC prevention among the exposed to biological risks of our University. The management of suspected latent or active tuberculosis infection was carried out in collaboration with the pulmonologist, assessing the risk of contagion among exposed or affected operators. Health surveillance protocol and judgements of suitability for specific task were made not only in consideration of worker health, but also considerating the possible risk for patients, since this disease is a major problem for public health.
