It is based on the hydrolysis of fluorescamine-labelled peptidoglycan from Micrococcus luteus cell walls. Lysozyme levels as low as 0.1 microgram can be detected.

Namely fluidic and electric connections can be realized on the same level. Nanoliter volume flow through cells for on-line measurement, static mixers for bioassays, and capillaries for heterogeneous quasi-continuous assays like immunoassays can be created. Hybrid modular systems are a special kind of bioanalytical microsystems.

Lactate dehydrogenase catalyzes the final step in glycolysis, the interconversion of pyruvate and... more Lactate dehydrogenase catalyzes the final step in glycolysis, the interconversion of pyruvate and lactate. The tetrameric enzyme is composed of one or two subunits (H and/or M) resulting in five isoenzyme forms: LDH-H4, -H3M1, -H2M2, -H1M3, and -M4. The relative distribution of the LDH isoenzymes is tissue dependent and a significant marker for the diagnosis of hepatoma of the liver, myocardial infarction, muscular dystrophy, and a wide variety of other acute and chronic diseases to be detected by alterations of the LDH isoenzyme pattern in serum. Immunochemical approaches to the routine determination of LDH depend on isoenzyme specific antibodies. Since the H- and M-subunits for human LDH are highly homologous, LDH isoenzyme specific antibodies for immunochemical monitoring are hard to generate. ABSTRACT A novel microphysiometry platform for dynamic cell culture monitoring in drug screening ... more ABSTRACT A novel microphysiometry platform for dynamic cell culture monitoring in drug screening and cancer cell research is presented. The transparent system features an integrated cell cultivation area, microfluidics and (bio-)sensors for multiparametric metabolic monitoring, combined on a single glass chip. The parameters include glucose and lactate (amperometric biosensors), oxygen (amperometric), pH (potentiometric) and cellular adhesion (interdigital electrode). In contrast to traditional silicon chip based systems, the choice of transparent materials allows microscopy at all times of cell cultivation and measurement. The introduction of biosensors allows the monitoring of additional key metabolic parameters with a detection limit for lactate as low as 2 μM. Integrated multi layer microfluidics enable dynamic measurements, further improving the analytical performance. The modular system is microfabricated in a cost-effective hybrid integration of thin film and laminate technology.

Still, after almost 80 years of focus on the problems created by hypoxia in solid tumours, we still largely lack methods to deal efficiently with these treatment-resistant cells. The consequences of this lack may be serious for many patients: Not only is there a negative correlation between the hypoxic fraction in tumours and the outcome of radiotherapy as well as many types of chemotherapy, a correlation has been shown between the hypoxic fraction in tumours and cancer metastasis. Thus, on a fundamental basis the great variety of problems related to hypoxia in cancer treatment has to do with the broad range of functions oxygen (and lack of oxygen) have in cells and tissues.
