We have designed primers for β - Subunit (accD region) of Acetyl-CoA carboxylase gene isolated fr... more We have designed primers for β - Subunit (accD region) of Acetyl-CoA carboxylase gene isolated from indigenous strains of Nannochlropsis salina, Dunaliella tertiolecta and Tetraselmis suecica. The primers were designed based on closest microalgal sequences that exist in database. Three subunits of ACCase were encoded by a nuclear genome and accDregion is encoded by plastid genome. It is the functional and conserved region of acc (Acetyl-CoA Carboxylase) gene. In our studies we observed, Nannochlo- ropsis salina had 866 bp of accD region which is smaller than Dunaliella tertiolecta and Tetraselmis suecica. Hence the transcriptional rate of accD gene in N. salina much faster than other microalgae; it directly influences the fatty acid anabolic pathway. The pairwise alignment showed N. salina and T. suecica has 60% than other pairs.

The screening of potent microalgae species are more important to produce sustainable biofuel. In this present investigation, 2 fresh water microalgae Chlorella vulgaris Beyerinck [Beijerinck], Scenedesmus quadricauda (Turpin) Brebisson and marine microalga, Isochrysis galabana Parke were taken and the cultures were maintained at photo flux of 150µE m-2 s-1 (12 light/ dark), 25˚C in an orbital shaker at 150 rpm up to 30 days. The main parameters used in this study were spectroscopy for growth rate determination, FT-IR and GC-MS to detect the fatty acid esters. Recently, fuel production from algae has been receiving considerable attention because of growing... more Recently, fuel production from algae has been receiving considerable attention because of growing energy prices, emissions of green house gases (Xiong et al., 2010) and gradual depletion of fossil fuels (Damiani, 2010). Microalgae due to their rapid biomass production, high photosynthetic efficiency (Xiong et al., 2010) and ability to storage a large amount of lipid are ideal source of biodiesel. In fact, microalgae have the highest lipid yield among various oil plants, and the lipid content of some microalgae has up to 80% and the compositions of microalgal oils are mainly TAG which is the right kind of oil for producing biodiesel (Chisti, 2007; Amin, 2009). To construct the plant expression vector containing synthetic B. thuringiensis-cry1Ac gene for cr... more To construct the plant expression vector containing synthetic B. thuringiensis-cry1Ac gene for crop plant transformations to resist the insect Helicoverpa armigera. A newly constructed binary vector containing the T - DNA left border, Kanamycin (kan) as marker gene, glucuronidase (uidA) reporter gene and bt-cry Ac 1 gene which transformed to A. tumefaciens by helper plasmid pRK2013, which provides tra and mob genes required to transfer the DNA. The plasmid constructed from basic vector pUC118 containing synthetic Bt-cry 1 Ac gene and pGPTV was restricted digested by EcoRI and Xbal. The digested plasmids were purified, quantified and ligated before triparental mating method of competent cell transformation. The triparental mating efficiency can be observed through back transformation of gene from A. tumefaciens to E.coli. The present study reports the production of fatty acids by microalgae under the influence of ligh... more The present study reports the production of fatty acids by microalgae under the influence of light and dark condition. Microalgae are renewable resource containing rich lipids in their body and has the potential to refill the partial energy demands in an eco-friendly way. We isolated an indigenous green alga (Chlorella vulgaris) as a potent source for biodiesel. To get better yield of biofuel, the growth of the microalgal isolate was optimized with the addition of nutrients and salts under light and dark conditions. The lipid fractions were extracted from the biomass through solvent extractions and the fractions were analyzed for biodiesel under GC-MS. The percentage of lipids synthesized from C. vulgaris under light and dark conditions were analyzed and compared. The algae from dark sample shows rich in saturated fatty acid (capric acid, lauric acid & myristic acid) and considerable amount of PUFA (hexadecatrienoic acid, stearidonic acid, eicosapenaenoic acid, docosahexaenoic acid) when compare to algae grown under light. So the algae grown in dark condition is an excellent source for high yield of saturated fatty acids.
