IFNα-2b as a protein is a cytokine used to treat more than 14 diseases all around the world, the ... more IFNα-2b as a protein is a cytokine used to treat more than 14 diseases all around the world, the recombinant human IFN-2b was synthesized and a genetic engineering method or recombinant DNA technology. IFNα-2b as a protein with broad biological action, including antiviral and anticancer properties, might be a useful therapeutic protein for a variety of diseases, including hepatitis C and cancer. IFN-α2b sequence was taken from the National Center for Biotechnology Information (NCBI) gene database and optimized before being cloned and produced in the pET28a+ vector. Escherichia coli was considered as a prokaryotic expression system. IFN-α2b expression was carried out in E. coli strains BL21 (DE3) and BL21(DE3) pLysS and E. coli DH5α strain used for cloning. isopropyl-d-thiogalactoside (IPTG) induced protein production in bacteria cells bearing the pET28a+ IFN-α2b construct, followed by sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) separation of total proteins. ...

Objective: The biological efficiency of one of the 2-piperidinone, N-[4-bromo-n-butyl]- (PNbb) co... more Objective: The biological efficiency of one of the 2-piperidinone, N-[4-bromo-n-butyl]- (PNbb) compounds that are isolated from the pomegranate peel extract was studied. Materials and Methods: Using organic solvents, seven compounds were isolated from 200 g dried pomegranate peels by Soxhlet extractor. PNbb with the use of solvents such as methanol, chloroform, ethyl acetate, and hexane was applied. The extraction ratio was higher when methanol was used and chloroform exhibits less. All the extracts were analyzed by the gas chromatography–mass spectrometry (GC-MS) to identify and characterize the chemical compounds present in the raw extract in both aquatic and organic layers. Each extracted isolated compound was detected using a number of internationally recognized detection methods. Background: Diabetes associated with multiple metabolic problems in the body, including bone mine... more Background: Diabetes associated with multiple metabolic problems in the body, including bone mineralization remodeling, osteoporosis and increase risk of fracture. Quercetin is natural flavonoids and according to animal studies; it has potent antioxidant, antidiabetic and protective effect against bone loss due to various causes. Objectives: explore effect of quercetin as nutritional supplement administrated orally on some bone mineralization bio-markers such as calcium, vitamin D and osteocalcin in Iraqi diabetic patients. Methods: interventional double-blind placebo randomized controlled study in which 40 patients with type 2 diabetes mellitus (age range 40-45) assigned randomly (using simple randomization) in either control (n=20) or study (n=20) group. Study group received Quercetin oral supplement as 500mg capsule once daily for three months. Venous blood was used for measuring Serum calcium, 25(OH) vitamin D and osteocalcin at base line and after 3 months. Results: After 3 months treatment with Quercetin; levels of Osteocalcin (28.1±7.6), serum calcium (9.2±1.8) and 25(OH) vitamin D (26.6±8.7) were significantly (p<0.05) higher than pretreatment values of osteocalcin (24.0±8.6); serum calcium (7.0±2.2) and 25(OH) vitamin D (20.6±7.7) and control values of serum calcium (6.8±2.0) and 25(OH) vitamin D (20.8±7.4), but not Osteocalcin (25.2±9.0). There was also significant correlation between use of quercetin; elevation of serum calcium and osteocalcin (r= 0.454; p= 0.032), indicating modulation in bone mineralization. Conclusions: Quercetin's use in diabetic patients may elevate Serum level of Calcium; 25(OH) vitamin D and may modulate bone mineralization represented by elevation of osteocalcin.

The depletion of 17β-estradiol-2 (17β-E2) is one of the factors that cause the risk of rheumatoid... more The depletion of 17β-estradiol-2 (17β-E2) is one of the factors that cause the risk of rheumatoid arthritis (RA) in females in the case of menopause. The aim of this study is to investigate whether the change in 17β-E2 levels and interleukin 1-beta (IL-1β) is associated with menopause in RA women and whether there is a relationship between them. 96 RA women were divided into three groups as follows: Group 1 (women of reproductive age) -30, Group 2 (premenopausal women) -32 (menstrual or normal menstrual period without menstruation for a period of not >6 months, and Group 3 (postmenopausal women) -34 women in menopause (menopause for at least 12 months). Serum levels of 17β-E2, IL-1β, and anti-cyclic citrullinated peptide were evaluated by enzyme-linked immunosorbent assay. The results showed that a change in concentration of 17β-E2 resulted in excessive production of IL-1β in women during reproductive age, premenopausal, and postmenopausal compared to female control. Furthermore, there is a highly inverse correlation between IL-1β and 17β-E2 in the serum of pre-and post-menopausal RA women. On the other hand, the study showed a positive correlation between IL-1β and sex hormones 17β-E2 in women of reproductive age who suffer from RA. Moreover, the study confirmed that the most risk factor is 17β-E2. The study showed that a lack of 17β-2 concentration after menopause causes an increased concentration of IL-1β and this, in turn, stimulates the development of RA disease during menopause. Menopause-associated 17β-E2 deficiency plays the major role in the pathogenesis of RA.

Subjects and methods: one hundered patients with cholilethiasis to GOLD standards criteria were participated in this study. One hundered patients with cholilethiasis, one hundered apparently healthy subjects were selected to be a normal group for comparison. In addition, assessing the plasma levels of tumor necrosis factor alpha [TNF-α], interlukine 1[IL-1], interlukine6 [IL-6], C terminal Telopeptides of type I Collagen [CTX1] and carboxyterminal propeptide of type I procollagen [PICP] by EILISA Kits. Results: The results show that the levels of tuner necrosis factor alpha [TNF-α],interlukin 1[IL-1] , interlukine 6[IL-6], C terminal Telopeptides of type I Collagen[CTX1]and carboxyterminal propeptide of type I procollagen[PICP] elevated in serum of patients with cholilethiasis significantly as compared with control healthy groups. Conclusion: The inflammatory activity of tumor necrosis factor alpha [TNF-α], interlukine 1[IL-1] and interlukine6 [IL-6] important in develops osteoporosis in patients with cholelithiasis by its effect on bone formation and resorption markers.

Cardiovascular risk is independently increased by plasma lipids abnormalities (low-density and hi... more Cardiovascular risk is independently increased by plasma lipids abnormalities (low-density and high density lipoprotein -cholesterol and triglycerides). Most patients have more than one lipid abnormality. Combination therapy with lipid-modifying agents could offer an important therapeutic option for improving the overall lipid profile. Combinations have demonstrated to provide additive efficacy and significant reductions in coronary events . This study was designed to evaluate the effect of ezetimibe, when used in combination with other hypolipidaemic agents ( statins) on lipid profile as well as on liver function ,renal function, oxidative stress, and platelets function when given to dyslipidaemic patients . Forty four patients (24 males and 20 females) with age ranged between 40-70 years (54 ±14.6) with dyslipidaemia on statins therapy for at least 6 month were involved in this clinical trials. They were randomized into two groups treated with either a combination of 20 mg/day simvastatin or a combination of 20mg/day atorvastatin and 10mg/day of ezetimibe.The study also included 22 apparently healthy subjects with age ranged (40-70years) and sex(11males and 11 females) matching that of the patients group. Serum lipid profile (total cholesterol -TC, triglycerides -TG, low density lipoprotein-cholesterol -LDL-C, very low density lipoprotein-cholesterol-VLDL-C, and high density lipoprotein-cholesterol -HDL-C), oxidative stress marker (Malondialdehyde-MDA), liver functions indices (Alanin aminotransferase -ALT,Aspartate aminotransferase-AST, total bilirubin), renal function parameters (urea, creatinine, and microalbuminuria) and platelets function test (bleeding time)were evaluated before and after 4 and 6 weeks of starting ezetimibe treatment . Treatment with ezetimibe plus simvastatin or atrovostatin resulted in significant lowering in TC, TG, LDL-C levels with elevation in HDL-C also the LDL/HDL ratio lowered significantly ( by 38.16%). This effect was associated with significant changes in liver function , and oxidative stress without changes in platelets function nor in renal function. The results presented in this study indicated that ezetimibe can be used in clinical practice for the treatment of dyslipidaemia, when combined with other hypolipidaemic agents like simvastatin and atorvastatin to improve the therapeutic profile with ameliorating some of their adverse effects.

Materials and Methods: Using organic solvents, seven compounds were isolated from 200 g dried pomegranate peels by Soxhlet extractor. PNbb with the use of solvents such as methanol, chloroform, ethyl acetate, and hexane was applied. The extraction ratio was higher when methanol was used and chloroform exhibits less. All the extracts were analyzed by the gas chromatography–mass spectrometry (GC-MS) to identify and characterize the chemical compounds present in the raw extract in both aquatic and organic layers. Quercetin is natural flavonoids and according to animal studies; it has potent antioxidant, antidiabetic and protective effect against bone loss due to various causes. Objectives: explore effect of quercetin as nutritional supplement administrated orally on some bone mineralization bio-markers such as calcium, vitamin D and osteocalcin in Iraqi diabetic patients. Methods: interventional double-blind placebo randomized controlled study in which 40 patients with type 2 diabetes mellitus (age range 40-45) assigned randomly (using simple randomization) in either control (n=20) or study (n=20) group. Study group received Quercetin oral supplement as 500mg capsule once daily for three months. Venous blood was used for measuring Serum calcium, 25(OH) vitamin D and osteocalcin at base line and after 3 months. Results: After 3 months treatment with Quercetin; levels of Osteocalcin (28.1±7.6), serum calcium (9.2±1.8) and 25(OH) vitamin D (26.6±8.7) were significantly (p<0.05) higher than pretreatment values of osteocalcin (24.0±8.6); serum calcium (7.0±2.2) and 25(OH) vitamin D (20.6±7.7) and control values of serum calcium (6.8±2.0) and 25(OH) vitamin D (20.8±7.4), but not Osteocalcin (25.2±9.0). There was also significant correlation between use of quercetin; elevation of serum calcium and osteocalcin (r= 0.454; p= 0.032), indicating modulation in bone mineralization. Conclusions: Quercetin's use in diabetic patients may elevate Serum level of Calcium; 25(OH) vitamin D and may modulate bone mineralization represented by elevation of osteocalcin.

The depletion of 17β-estradiol-2 (17β-E2) is one of the factors that cause the risk of rheumatoid... more The depletion of 17β-estradiol-2 (17β-E2) is one of the factors that cause the risk of rheumatoid arthritis (RA) in females in the case of menopause. The aim of this study is to investigate whether the change in 17β-E2 levels and interleukin 1-beta (IL-1β) is associated with menopause in RA women and whether there is a relationship between them. 96 RA women were divided into three groups as follows: Group 1 (women of reproductive age) -30, Group 2 (premenopausal women) -32 (menstrual or normal menstrual period without menstruation for a period of not >6 months, and Group 3 (postmenopausal women) -34 women in menopause (menopause for at least 12 months). Serum levels of 17β-E2, IL-1β, and anti-cyclic citrullinated peptide were evaluated by enzyme-linked immunosorbent assay. The results showed that a change in concentration of 17β-E2 resulted in excessive production of IL-1β in women during reproductive age, premenopausal, and postmenopausal compared to female control. Furthermore, there is a highly inverse correlation between IL-1β and 17β-E2 in the serum of pre-and post-menopausal RA women. On the other hand, the study showed a positive correlation between IL-1β and sex hormones 17β-E2 in women of reproductive age who suffer from RA. Moreover, the study confirmed that the most risk factor is 17β-E2. The study showed that a lack of 17β-2 concentration after menopause causes an increased concentration of IL-1β and this, in turn, stimulates the development of RA disease during menopause. Menopause-associated 17β-E2 deficiency plays the major role in the pathogenesis of RA.

Subjects and methods: one hundered patients with cholilethiasis to GOLD standards criteria were participated in this study. One hundered patients with cholilethiasis, one hundered apparently healthy subjects were selected to be a normal group for comparison. In addition, assessing the plasma levels of tumor necrosis factor alpha [TNF-α], interlukine 1[IL-1], interlukine6 [IL-6], C terminal Telopeptides of type I Collagen [CTX1] and carboxyterminal propeptide of type I procollagen [PICP] by EILISA Kits. Results: The results show that the levels of tuner necrosis factor alpha [TNF-α],interlukin 1[IL-1] , interlukine 6[IL-6], C terminal Telopeptides of type I Collagen[CTX1]and carboxyterminal propeptide of type I procollagen[PICP] elevated in serum of patients with cholilethiasis significantly as compared with control healthy groups. Conclusion: The inflammatory activity of tumor necrosis factor alpha [TNF-α], interlukine 1[IL-1] and interlukine6 [IL-6] important in develops osteoporosis in patients with cholelithiasis by its effect on bone formation and resorption markers.

Cardiovascular risk is independently increased by plasma lipids abnormalities (low-density and hi... more Cardiovascular risk is independently increased by plasma lipids abnormalities (low-density and high density lipoprotein -cholesterol and triglycerides). Most patients have more than one lipid abnormality. Combination therapy with lipid-modifying agents could offer an important therapeutic option for improving the overall lipid profile. Combinations have demonstrated to provide additive efficacy and significant reductions in coronary events . This study was designed to evaluate the effect of ezetimibe, when used in combination with other hypolipidaemic agents ( statins) on lipid profile as well as on liver function ,renal function, oxidative stress, and platelets function when given to dyslipidaemic patients . Forty four patients (24 males and 20 females) with age ranged between 40-70 years (54 ±14.6) with dyslipidaemia on statins therapy for at least 6 month were involved in this clinical trials. They were randomized into two groups treated with either a combination of 20 mg/day simvastatin or a combination of 20mg/day atorvastatin and 10mg/day of ezetimibe.The study also included 22 apparently healthy subjects with age ranged (40-70years) and sex(11males and 11 females) matching that of the patients group. Serum lipid profile (total cholesterol -TC, triglycerides -TG, low density lipoprotein-cholesterol -LDL-C, very low density lipoprotein-cholesterol-VLDL-C, and high density lipoprotein-cholesterol -HDL-C), oxidative stress marker (Malondialdehyde-MDA), liver functions indices (Alanin aminotransferase -ALT,Aspartate aminotransferase-AST, total bilirubin), renal function parameters (urea, creatinine, and microalbuminuria) and platelets function test (bleeding time)were evaluated before and after 4 and 6 weeks of starting ezetimibe treatment . Treatment with ezetimibe plus simvastatin or atrovostatin resulted in significant lowering in TC, TG, LDL-C levels with elevation in HDL-C also the LDL/HDL ratio lowered significantly ( by 38.16%). This effect was associated with significant changes in liver function , and oxidative stress without changes in platelets function nor in renal function. The results presented in this study indicated that ezetimibe can be used in clinical practice for the treatment of dyslipidaemia, when combined with other hypolipidaemic agents like simvastatin and atorvastatin to improve the therapeutic profile with ameliorating some of their adverse effects.
