A monoclonal antibody (MAb) specific for rainbow trout thrombocytes was produced and its reactivi... more A monoclonal antibody (MAb) specific for rainbow trout thrombocytes was produced and its reactivity was demonstrated by flow cytometry and immuno-electron microscopy. Flow cytometry analysis showed that this MAb (TTL-7D11) reacted positively with about 30% of the peripheral blood leucocytes (PBL) and about 1%, 2%, and 11% of the pronephros, mesonephros, and spleen cells, respectively. Electron microscopy using immunogold labeling demonstrated that this MAb reacted strongly with thrombocytes, where gold beads could be seen attached only to the membrane and canalicular system of these cells. Positive and negative leucocytes for this MAb were obtained by magnetic cell separation. In the positive fraction, 96% of the cells were thrombocytes, while in the negative fraction no more than 3% were, which clearly showed a high purity of the positive fraction. Aggregation studies showed that about 75% of the positive fraction cells aggregated after being mixed with U-46619 thromboxane-mimetic, whereas in the negative fraction only 10% of the cells did so. Thus, utilizing the TTL-7D11 we have succeeded in isolating a pure thrombocyte population, and this would facilitate further studies, particularly on their characteristics and function(s).

Rash" , a subchronic, debilitating and non-fatal inflammatory skin disease has been found in cult... more Rash" , a subchronic, debilitating and non-fatal inflammatory skin disease has been found in cultured rainbow trout (Oncorhynchus mykiss) in several areas of Japan over the past five years. This condition was observed from small fish (11cm in body length) to mature-size animals, however, most of the affected fish were market size (in Japan, 20cm/120g). Morbidity sometimes reaches as high as 48% and causes a significant economic impact, since those fish loose commercial value. A self-limited clinical course and healing of the lesion could be observed after 6 to 8 weeks. "Rash" signs included the presence of bright red, non-raised, ulcerated or not, defined to petechial scattered lesions on the ventral and/or lateral surfaces of the fish. Histopathological features included a subchronic focal to non-focal, non-suppurative dermatitis with various degrees of ulceration and an extensive mononuclear inflammatory infiltration. "Rash" etiology still remains unknown .

Global warming is a reality and its effects have been widely studied. However, the consequences f... more Global warming is a reality and its effects have been widely studied. However, the consequences for marine invertebrates remain poorly understood. Thus, the present study proposed to evaluate the effect of elevated temperature on the innate immune system of Antarctic sea urchin Sterechinus neumayeri. Sea urchins were collected nearby Brazilian Antarctic Station ''Comandante Ferraz'' and exposed to 0 (control), 2 and 4°C for periods of 48 h, 2, 7 and 14 days. After the experimental periods, coelomic fluid was collected in order to perform the following analyses: coelomocytes differential counting, phagocytic response, adhesion and spreading coelomocytes assay, intranuclear iron crystalloid and ultra structural analysis of coelomocytes. The red sphere cell was considered a biomarker for heat stress, as they increased in acute stress. Besides that, a significant increase in phagocytic indexes was observed at 2°C coinciding with a significant increase of intranuclear iron crystalloid at the same temperature and same time period. Furthermore, significant alterations in cell adhesion and spreading were observed in elevated temperatures. The ultra structural analysis of coelomocytes showed no significant difference across treatments. This was the first time that innate immune response alterations were observed in response to elevated temperature in a Polar echinoid.

Bone marrow is a source of stem cells for greater and easier access, which is widely studied as a... more Bone marrow is a source of stem cells for greater and easier access, which is widely studied as a provider of hematopoietic and mesenchymal cells for various purposes, mainly therapeutic by the advances in research involving cell therapy. The swine is an animal species commonly used in the pursuit of development of experimental models. Thus, this study aimed to standardize protocol for collection and separation of bone marrow in swines, since this species is widely used as experimental models for various diseases. Twelve animals were used, which underwent bone marrow puncture with access from the iliac crest and cell separation by density gradient followed by a viability test with an average of 98% of viable cells. Given our results, we can ensure the swine as an excellent model for obtaining and isolation of mononuclear cells from bone marrow, stimulating several studies addressing the field of cell therapy.

In this work, we studied the embryology of mice of 12, 14, and 18 days of gestation by gross obse... more In this work, we studied the embryology of mice of 12, 14, and 18 days of gestation by gross observation, light microscopy, and scanning electron microscopy. Grossly, the embryos of 12 days were observed in C-shaped region of the brain, eye pigmentation of the retina, first, second, and third pharyngeal arches gill pit nasal region on the fourth ventricle brain, cervical curvature, heart, liver, limb bud thoracic, spinal cord, tail, umbilical cord, and place of the mesonephric ridge. Microscopically, the liver, cardiovascular system and spinal cord were observed. In the embryo of 14 days, we observed structures that make up the liver and heart. At 18 days of gestation fetuses, it was noted the presence of eyes, mouth, and nose in the cephalic region, chest and pelvic region with the presence of well-developed limbs, umbilical cord, and placenta. Scanning electron microscopy in 18 days of gestation fetuses evidenced head, eyes closed eyelids, nose, vibrissae, forelimb, heart, lung, kidney, liver, small bowel, diaphragm, and part of the spine. The results obtained in this work describe the internal and external morphology of mice, provided by an integration of techniques and review of the morphological knowledge of the embryonic development of this species, as this animal is of great importance to scientific studies.

This study was designed to compare cutaneous mycoflora isolation and CD4+:CD8+ ratio in feline im... more This study was designed to compare cutaneous mycoflora isolation and CD4+:CD8+ ratio in feline immunodeficiency virus (FIV)-infected cats with that in FIV-uninfected cats. Sixty cats were examined. Twenty-five were FIV-infected cats and 35 were FIV-uninfected cats. All 60 cats were FeLV-negative. Fungi were speciated and immunophenotyping of peripheral CD4+ and CD8+ T lymphocytes was performed. At least one fungal colony was isolated from 22/25 (88%) FIV-infected cats. Among the FIV-uninfected cats fungal colonies were recovered from 13/35 (37%) specimens. Dermatophytes were recovered from 2/25 (8%) FIV-infected cats (one Microsporum gypseum, one Microsporum canis) and 3/35 (8.5%) FIV-uninfected cats (M gypseum). Malassezia species was the most commonly isolated organism from both groups of cats (51.6%). Malassezia species was more commonly isolated from FIV-infected cats than FIV-uninfected cats (84% vs 28.6%). The CD4+ to CD8+ lymphocyte ratio for FIV-infected cats was significantly lower than the CD4+ to CD8+ ratio in the FIV-uninfected cats. The CD4+ to CD8+ lymphocyte ratio for FIV-infected cats with cutaneous overall fungal isolation was significantly lower than the CD4:CD8 lymphocyte ratio in the FIV-infected cats but without cutaneous fungal isolation. We can conclude that immunologic depletion due to retroviral infection might represent a risk factor to cutaneous fungal colonization in cats.

Fifteen papillae from female water buffaloes and one bovine female were obtained in a slaughter h... more Fifteen papillae from female water buffaloes and one bovine female were obtained in a slaughter house for this study, which consisted in morphologic and biometrical analyses, histology and histometric and mesoscopical analyses. The means were: right cranial papilla (RCP) 20.95g; right caudal papilla (RCaP) 21.49g; left cranial papilla (LCP) 23.52g; and left caudal papilla (LCaP) 21.35g. Papillary duct average length, numbers of folds and papillary cisterns length were, respectively: 0.67/5,5 e 2.17cm for RCP; 0.62/6.0 e 2.26cm for RCaP; and for the left antimere: RCP: 0.71/6.0 e 2.26cm and for RCaP: 0.74/5.7 e 2.57cm. Pearson's analysis revealed negative correlation between body weight and mammary glandular weight. Sampling of proximal, middle and distal area was taken and fixed into a 10% formaldehyde solution, followed by mesoscopical investigation through 10, 20 and 40µm thickness cuts stained by Azan, F3BA Picrosirus and Weigert-Van Gieson techniques. Histological and morphometric analyses were applied on 5µm thick samples and showed that the muscular layer was disposed into two arrangements, internal circular and external longitudinal with a bi-stratified cuboidal epithelium. A well developed muscular layer were related to the middle portion revealing thickness of 22.90mm with a great amount of connective tissue (7.10mm).
