In humans, the organs other than the liver known to produce SHBG include the brain, uterus, testis, prostate, breast and ovary, and the locally expressed SHBG is considered to play an important role in various physiological and pathological processes. A few studies of SHBG in rats were reported, but systemic SHBG studies in consideration of different organs and aging are currently missing. We retrospectively analyzed 54 ulcerative gastric diffuse large B-cell lymphoma (G-DLBCL) patients to compare safety and efficacy of low-dose pre-phase before 4 to 6 cycles of conventional-dose CT (n=28) with 4 to 6 cycles of conventional-dose CT (n=26)between October, 2005 and August, 2014. Patients who received low-dose pre-phase before conventional-dose CT showed lower gastrorrhagia or gastric perforation rate (0% vs. 15.4%, P=0.047) and higher complete response (CR) rate (78.6% vs. 46.2%, P=0.023) and 5-year progression-free survival (PFS) rate (63% vs. 31%, P=0.021) than patients who received conventional-dose CT alone. Our study suggests that low-dose pre-phase before conventional-dose CT provides a safe and effective method for ulcerative G-DLBCL.

We examined metabolically healthy obesity (MHO) and its associates in 2530 Mongolian Chinese adults. MHO was defined by waist circumference, low-density lipoprotein (LDL-C) cholesterol, high-density lipoprotein cholesterol (HDL-C), triglycerides (TGs), systolic blood pressure (SBP), diastolic blood pressure (DBP), and glucose. Only 3.0% of the participants had MHO, with 0.8% of men and 4.5% of women having this condition (P < 0.001 for sex difference). Despite striking differences in obesity measures, MHO individuals had a comparable cardiometabolic profile to that for metabolically healthy, nonobese individuals (MHNO) and an improved cardiometabolic profile, i.e., lower levels of blood pressure, glucose, insulin, LDL-C, TGs, and higher levels of HDL-C compared to metabolically abnormal individuals (all P < 0.01, except for insulin). This was a single arm, prospective, observational multicenter and phase IV clinical trial for 279 patients, who were newly diagnosed as CD20-positive DLBCL from 24 medical centers in China 2011 and 2012, no special exclusion criteria were used. A total of 1034 men aged 20 years and older free of cardiovascular disease were included in the cohort and followed up for an average of 9.2 years. Intense investigation found that this pathway also plays an important role in other biological process including angiogenesis. Robo4 is the vascular endothelial cell specific member of Robo family. It was found that Slit-Robo signaling can inhibit endothelial cell migration, tube formation and vascular permeability. Slit-Robo signaling also plays an important role in embryonic and tumor angiogenesis. In animal model of ocular angiogenesis, addition of Slit inhibited laser induced choroidal neovascularization, oxygen induced retinopathy and VEGF induced retinal permeability in a Robo4 dependent manner. The plasmids of pcDNA3.1/TCR V ␤ 8 was constructed. The BALB/c mice were randomly divided into four groups which were pcDNA3.1, pcDNA3.1/TCR V ␤ 8, pcDNA3.1/TCR V ␤ 8 + CpG + liposome and phosphorothioate CpG groups with six mice in each group. Vaccines were injected in bilateral musculus quadriceps femoris of mice in the 0, second, and fourth week, respectively. The antibody formation was tested by indirect immuofluorescence in the 0, second, fourth, sixth and eighth weeks, respectively, before and after immunization. Production of antibody against TCR V ␤ 8 antigen was observed in the groups of pcDNA3.1/TCR V ␤ 8 and pcDNA3.1/TCR V ␤ 8 + CpG + liposome. The antibody titer began to rise in the fourth week and attain the maximal value in the sixth week. The antibody titer in the group of pcDNA3.1/TCR V ␤ 8 + CpG + liposome was higher than that in the group of pcDNA3.1/TCR V ␤ 8 in the fourth and eighth weeks (both P < 0.01); the antibody titer in the group of pcDNA3.1/TCR V ␤ 8 + CpG + liposome was markedly higher than that in the group of pcDNA3.1/TCR V ␤ 8 in the sixth week (P < 0.001). The result indicate that TCR V ␤ 8 antigen can induce formation of special antibody in mice. CpG and liposome can improve the humoral immunoreaction induced by TCR V ␤ 8 gene vaccine.

Objective: We aimed to evaluate the controversial association between human urotensin II and esse... more Objective: We aimed to evaluate the controversial association between human urotensin II and essential hypertension in untreated hypertensive cases and normotensive controls. Methods: 197 newly diagnosed hypertensive patients and 197 age-and sex-matched normotensive controls were studied. Plasma urotensin II, nitric oxide metabolites, and other traditional biomarkers were examined. Results: Hypertensive patients had higher urotensin II [median (interquartile rang): 9.32 (7.86-11.52) ng/mL vs 8.52 (7.07-10.41) ng/mL] and lower nitric oxide metabolites [19.19 (2.55-38.48) mmol/L vs 23.83 (11.97-43.40) mmol/L] than normotensive controls. Urotensin II was positively correlated with systolic blood pressure (r = 0.169, P,0.001) and diastolic blood pressure (r = 0.113, P = 0.024) while negatively correlated with nitric oxide metabolites (r = 20.112, P = 0.027). In multivariate regression analysis, subjects in the highest quartile of urotensin II were more likely to have hypertension than those in the lowest quartile (OR, 2.58; 95% CI, 1.21-5.49). Subgroup analyses in 106 pairs of cases and controls with either both normal or both abnormal nitric oxide metabolites levels showed that the association between urotensin II levels and hypertension persisted (P value for trend = 0.039). Conclusion: Human urotensin II is markedly associated with essential hypertension, and the association is independent of nitric oxide metabolites. Our results indicated that urotensin II might be an independent risk factor for essential hypertension.

To evaluate the efficacy of combined treatment of ciprofloxacin and glucocorticoid for child refr... more To evaluate the efficacy of combined treatment of ciprofloxacin and glucocorticoid for child refractory Mycoplasma pneumoniae (M. pneumoniae) pneumonia. Methods: Clinical and laboratory characteristics of six pediatric refractory M. pneumoniae pneumonia cases treated with ciprofloxacin and glucocorticoids were reported. Results: Five cases complicated with pleural effusion. The average febrile period prior to admission was 8.5 AE 2.0 days, the average total febrile period was 14.6 AE 7.6 days, and the average febrile period after treatment was 3.3 AE 3.0 days. The average time in hospital for patients was 23.6 AE 4.8 days. The initial mean WBC count was 10,100 AE 2,400/mm 3. The erythrocyte sedimentation rate (ESR), lactate dehydrogenase (LDH), and C-reactive protein (CRP) in all cases were elevated during the course of illness. After combined therapy, all children clinically improved, with resolution of radiographic findings and normal laboratory items. We analyzed the 23S rRNA gene in four nasopharyngeal secretions, and found mutations in A2063G of domain V in three cases, and mutation in A2064G in the other case. Conclusion: Combined treatment of ciprofloxacin and glucocorticoids can significantly ameliorate child refractory M. pneumoniae pneumonia further comparative study is needed to well evaluate the treatment efficacy.

Design: Prospective, randomized, controlled, crossover trial. Participants: We included 83 children aged 3 to Ͻ7 years with untreated anisometropic amblyopia and baseline best-corrected visual acuity (BCVA) of 20/40 to 20/200 in the amblyopic eye. Methods: Participants were randomized to receive spectacles alone (group 1; n ϭ 42) or spectacles ϩ acupuncture (group 2; n ϭ 41) for 15 weeks, and were then crossed over to receive the other regimen for another 15 weeks. The BCVA in both eyes was measured at baseline and every 5 (Ϯ1) weeks for the initial 45 weeks and at 60 (Ϯ1) weeks. Main Outcome Measures: BCVA in the amblyopic eye at 15, 30, and 60 weeks. Results: The mean baseline BCVA in the amblyopic eye was 0.50 and 0.49 logarithm of the minimum angle of resolution logMAR in groups 1 and 2, respectively. After 15 weeks of treatment, the BCVA had improved by a mean of 2.2 lines in group 1 and 2.9 lines in group 2. The mean difference in BCVA between groups was 0.77 lines (95% confidence interval (CI), 0.29-1.3; P ϭ 0.0020) with baseline adjustment. BCVA of Յ0.1 logMAR was achieved in 14.6% of the patients in group 1 and 57.5% in group 2 (PϽ0.00010). After the regimens were crossed over at 30 weeks, group 1 had a mean of 1.2 (95% CI, 0.98-1.48; P ϭ 2.0 ϫ 10 Ϫ12) lines additional improvement from the 15-week BCVA, whereas in group 2 the mean improvement was 0.4 (95% CI, 0.19-0.63; P ϭ 0.0010) lines. The proportions of responders, resolution, and participants achieving a BCVA of Յ0.1 logMAR at 30 weeks were similar between groups. After completion of acupuncture, only 1 participant had Ͼ1 line of VA decrease to 60 weeks. Acupuncture was well-tolerated by all children, and no severe adverse effect was encountered. Conclusions: Acupuncture is a potentially useful complementary treatment modality that may provide sustainable adjunctive effect to refractive correction for anisometropic amblyopia in young children. Further large-scale studies seem warranted.

Increased podocyte cyclooxygenase-2 (COX-2) expression is seen in rats after renal ablation and T... more Increased podocyte cyclooxygenase-2 (COX-2) expression is seen in rats after renal ablation and Thy-1 nephritis and in cultured murine podocytes in response to mechanical stress. For investigation of whether COX-2 overexpression plays a role in podocyte injury, transgenic B6/D2 mice in which COX-2 expression was driven by a nephrin promoter were established. Selective upregulation of COX-2 expression in podocytes of transgenic mouse kidneys was confirmed by immunoblotting and immunohistochemistry. Whether upregulation of podocyte-specific COX-2 expression enhanced sensitivity to the development of Adriamycin nephropathy was examined. Adriamycin administration induced dramatically more albuminuria and foot process effacement and reduced glomerular nephrin mRNA and immunoreactivity in transgenic mice compared with wild-type littermates. Adriamycin also markedly increased immunoreactive COX-2 expression in podocytes from transgenic mice compared with the wild-type mice. Reverse transcriptase-PCR indicated that this increase represented a stimulation of endogenous COX-2 mRNA expression rather than COX-2 mRNA driven by the nephrin promoter. Balb/C mice, which are susceptible to renal injury by Adriamycin, also increased podocyte COX-2 expression and reduced nephrin expression in response to administration of the drug. Long-term treatment with the COX-2-specific inhibitor SC58236 ameliorated the albuminuria that was induced by Adriamycin in the transgenic mice. SC58236 also reduced Adriamycin-induced foot process effacement in both the COX-2 transgenic mice and Balb/C mice. Therefore, overexpression of COX-2 may predispose podocytes to further injury.

To report the spontaneous closure of bilateral traumatic macular holes in a young patient, follow... more To report the spontaneous closure of bilateral traumatic macular holes in a young patient, followed up with optical coherence tomography (OCT), muti-focal electroretinogram (mfERG), and Humphrey visual field examination. Methods A 25-year-old male who suffered from bilateral blunt trauma to the eyes and developed traumatic macular holes was followed with ophthalmic examination, OCT, mfERG, and Humphrey visual field examination. Results The OCT results revealed spontaneous closure in both eyes 2 weeks after trauma, and the macular holes remained closed in the 7 months of follow-up. Visual acuity improved to 0.7 in right and 0.9 in left eye finally. Visual field examination and mfERG results improved slightly compared with the initial tests. However, there still were central scotomas in both eyes, especially in right eye at the end of follow-up, and the peaks of the retinal response density had not recovered in the macular area of the mfERG topography. Conclusions Spontaneous closure of unilateral traumatic macular hole is not uncommon, but there is no report of spontaneous closure of bilateral traumatic macular holes as yet. Since the traumatic macular holes may close spontaneously, traumatic macular holes may be observed for a period of follow-up.

Methods The PhNR was elicited with red stimuli (5 cd s/m 2 with 4-min duration) and blue background (10 cd/m 2). Standard Ganzfeld flash ERGs were recorded according to the ISCEV standard for the clinical electroretinogram (2004). A total of 81 diabetic patients with different severity levels of NPDR were examined. Forty-three age-matched normal controls were also studied. Results The amplitude of PhNR decreased significantly as DR progressed, while the implicit time was prolonged.

We retrospectively analyzed 54 ulcerative gastric diffuse large B-cell lymphoma (G-DLBCL) patients to compare safety and efficacy of low-dose pre-phase before 4 to 6 cycles of conventional-dose CT (n=28) with 4 to 6 cycles of conventional-dose CT (n=26)between October, 2005 and August, 2014. Patients who received low-dose pre-phase before conventional-dose CT showed lower gastrorrhagia or gastric perforation rate (0% vs. 15.4%, P=0.047) and higher complete response (CR) rate (78.6% vs. 46.2%, P=0.023) and 5-year progression-free survival (PFS) rate (63% vs. 31%, P=0.021) than patients who received conventional-dose CT alone. Our study suggests that low-dose pre-phase before conventional-dose CT provides a safe and effective method for ulcerative G-DLBCL.

We examined metabolically healthy obesity (MHO) and its associates in 2530 Mongolian Chinese adults. MHO was defined by waist circumference, low-density lipoprotein (LDL-C) cholesterol, high-density lipoprotein cholesterol (HDL-C), triglycerides (TGs), systolic blood pressure (SBP), diastolic blood pressure (DBP), and glucose. Only 3.0% of the participants had MHO, with 0.8% of men and 4.5% of women having this condition (P < 0.001 for sex difference). The plasmids of pcDNA3.1/TCR V ␤ 8 was constructed. The BALB/c mice were randomly divided into four groups which were pcDNA3.1, pcDNA3.1/TCR V ␤ 8, pcDNA3.1/TCR V ␤ 8 + CpG + liposome and phosphorothioate CpG groups with six mice in each group. Vaccines were injected in bilateral musculus quadriceps femoris of mice in the 0, second, and fourth week, respectively. The antibody formation was tested by indirect immuofluorescence in the 0, second, fourth, sixth and eighth weeks, respectively, before and after immunization. Production of antibody against TCR V ␤ 8 antigen was observed in the groups of pcDNA3.1/TCR V ␤ 8 and pcDNA3.1/TCR V ␤ 8 + CpG + liposome. The antibody titer began to rise in the fourth week and attain the maximal value in the sixth week. The antibody titer in the group of pcDNA3.1/TCR V ␤ 8 + CpG + liposome was higher than that in the group of pcDNA3.1/TCR V ␤ 8 in the fourth and eighth weeks (both P < 0.01); the antibody titer in the group of pcDNA3.1/TCR V ␤ 8 + CpG + liposome was markedly higher than that in the group of pcDNA3.1/TCR V ␤ 8 in the sixth week (P < 0.001). The result indicate that TCR V ␤ 8 antigen can induce formation of special antibody in mice. CpG and liposome can improve the humoral immunoreaction induced by TCR V ␤ 8 gene vaccine.

Objective: We aimed to evaluate the controversial association between human urotensin II and esse... more Objective: We aimed to evaluate the controversial association between human urotensin II and essential hypertension in untreated hypertensive cases and normotensive controls. Methods: 197 newly diagnosed hypertensive patients and 197 age-and sex-matched normotensive controls were studied. Plasma urotensin II, nitric oxide metabolites, and other traditional biomarkers were examined. Results: Hypertensive patients had higher urotensin II [median (interquartile rang): 9.32 (7.86-11.52) ng/mL vs 8.52 (7.07-10.41) ng/mL] and lower nitric oxide metabolites [19.19 (2.55-38.48) mmol/L vs 23.83 (11.97-43.40) mmol/L] than normotensive controls. Urotensin II was positively correlated with systolic blood pressure (r = 0.169, P,0.001) and diastolic blood pressure (r = 0.113, P = 0.024) while negatively correlated with nitric oxide metabolites (r = 20.112, P = 0.027). In multivariate regression analysis, subjects in the highest quartile of urotensin II were more likely to have hypertension than those in the lowest quartile (OR, 2.58; 95% CI, 1.21-5.49). Subgroup analyses in 106 pairs of cases and controls with either both normal or both abnormal nitric oxide metabolites levels showed that the association between urotensin II levels and hypertension persisted (P value for trend = 0.039). Conclusion: Human urotensin II is markedly associated with essential hypertension, and the association is independent of nitric oxide metabolites. Our results indicated that urotensin II might be an independent risk factor for essential hypertension.

Methods: Clinical and laboratory characteristics of six pediatric refractory M. pneumoniae pneumonia cases treated with ciprofloxacin and glucocorticoids were reported. Results: Five cases complicated with pleural effusion. The average febrile period prior to admission was 8.5 AE 2.0 days, the average total febrile period was 14.6 AE 7.6 days, and the average febrile period after treatment was 3.3 AE 3.0 days. The average time in hospital for patients was 23.6 AE 4.8 days. The initial mean WBC count was 10,100 AE 2,400/mm 3. The erythrocyte sedimentation rate (ESR), lactate dehydrogenase (LDH), and C-reactive protein (CRP) in all cases were elevated during the course of illness. After combined therapy, all children clinically improved, with resolution of radiographic findings and normal laboratory items. We analyzed the 23S rRNA gene in four nasopharyngeal secretions, and found mutations in A2063G of domain V in three cases, and mutation in A2064G in the other case. Conclusion: Combined treatment of ciprofloxacin and glucocorticoids can significantly ameliorate child refractory M. pneumoniae pneumonia further comparative study is needed to well evaluate the treatment efficacy.

Design: Prospective, randomized, controlled, crossover trial. Participants: We included 83 children aged 3 to Ͻ7 years with untreated anisometropic amblyopia and baseline best-corrected visual acuity (BCVA) of 20/40 to 20/200 in the amblyopic eye. Methods: Participants were randomized to receive spectacles alone (group 1; n ϭ 42) or spectacles ϩ acupuncture (group 2; n ϭ 41) for 15 weeks, and were then crossed over to receive the other regimen for another 15 weeks. The BCVA in both eyes was measured at baseline and every 5 (Ϯ1) weeks for the initial 45 weeks and at 60 (Ϯ1) weeks. Main Outcome Measures: BCVA in the amblyopic eye at 15, 30, and 60 weeks. Results: The mean baseline BCVA in the amblyopic eye was 0.50 and 0.49 logarithm of the minimum angle of resolution logMAR in groups 1 and 2, respectively. After 15 weeks of treatment, the BCVA had improved by a mean of 2.2 lines in group 1 and 2.9 lines in group 2. The mean difference in BCVA between groups was 0.77 lines (95% confidence interval (CI), 0.29-1.3; P ϭ 0.0020) with baseline adjustment. BCVA of Յ0.1 logMAR was achieved in 14.6% of the patients in group 1 and 57.5% in group 2 (PϽ0.00010). After the regimens were crossed over at 30 weeks, group 1 had a mean of 1.2 (95% CI, 0.98-1.48; P ϭ 2.0 ϫ 10 Ϫ12) lines additional improvement from the 15-week BCVA, whereas in group 2 the mean improvement was 0.4 (95% CI, 0.19-0.63; P ϭ 0.0010) lines. The proportions of responders, resolution, and participants achieving a BCVA of Յ0.1 logMAR at 30 weeks were similar between groups. After completion of acupuncture, only 1 participant had Ͼ1 line of VA decrease to 60 weeks. Acupuncture was well-tolerated by all children, and no severe adverse effect was encountered. Conclusions: Acupuncture is a potentially useful complementary treatment modality that may provide sustainable adjunctive effect to refractive correction for anisometropic amblyopia in young children. Further large-scale studies seem warranted.

Increased podocyte cyclooxygenase-2 (COX-2) expression is seen in rats after renal ablation and T... more Increased podocyte cyclooxygenase-2 (COX-2) expression is seen in rats after renal ablation and Thy-1 nephritis and in cultured murine podocytes in response to mechanical stress. For investigation of whether COX-2 overexpression plays a role in podocyte injury, transgenic B6/D2 mice in which COX-2 expression was driven by a nephrin promoter were established. Selective upregulation of COX-2 expression in podocytes of transgenic mouse kidneys was confirmed by immunoblotting and immunohistochemistry. Whether upregulation of podocyte-specific COX-2 expression enhanced sensitivity to the development of Adriamycin nephropathy was examined. Adriamycin administration induced dramatically more albuminuria and foot process effacement and reduced glomerular nephrin mRNA and immunoreactivity in transgenic mice compared with wild-type littermates. Adriamycin also markedly increased immunoreactive COX-2 expression in podocytes from transgenic mice compared with the wild-type mice. Reverse transcriptase-PCR indicated that this increase represented a stimulation of endogenous COX-2 mRNA expression rather than COX-2 mRNA driven by the nephrin promoter. Balb/C mice, which are susceptible to renal injury by Adriamycin, also increased podocyte COX-2 expression and reduced nephrin expression in response to administration of the drug. Long-term treatment with the COX-2-specific inhibitor SC58236 ameliorated the albuminuria that was induced by Adriamycin in the transgenic mice. SC58236 also reduced Adriamycin-induced foot process effacement in both the COX-2 transgenic mice and Balb/C mice. Therefore, overexpression of COX-2 may predispose podocytes to further injury.

Methods A 25-year-old male who suffered from bilateral blunt trauma to the eyes and developed traumatic macular holes was followed with ophthalmic examination, OCT, mfERG, and Humphrey visual field examination. Results The OCT results revealed spontaneous closure in both eyes 2 weeks after trauma, and the macular holes remained closed in the 7 months of follow-up. Visual acuity improved to 0.7 in right and 0.9 in left eye finally. Visual field examination and mfERG results improved slightly compared with the initial tests. However, there still were central scotomas in both eyes, especially in right eye at the end of follow-up, and the peaks of the retinal response density had not recovered in the macular area of the mfERG topography. Conclusions Spontaneous closure of unilateral traumatic macular hole is not uncommon, but there is no report of spontaneous closure of bilateral traumatic macular holes as yet. Since the traumatic macular holes may close spontaneously, traumatic macular holes may be observed for a period of follow-up.

Standard Ganzfeld flash ERGs were recorded according to the ISCEV standard for the clinical electroretinogram (2004). A total of 81 diabetic patients with different severity levels of NPDR were examined. Forty-three age-matched normal controls were also studied. Results The amplitude of PhNR decreased significantly as DR progressed, while the implicit time was prolonged.
