Memory T cells (TM) are a component of the adaptive immune system. They ensure the host of more aggressive and faster immune response to efficiently eliminate the specific antigens after re-exposure and thus play a key role in T-cell functions. In this study we investigate the quantities and functions of memory T cells in SAA patients before and after immunosuppressive therapy (IST) to further clarify the mechanism of SAA apoptosis of bone marrow hematopoietic cells. Results showed that the percentage of CD4+ effector T cells in peripheral blood and bone marrow lymphocytes was decreased in SAA patients. The ratio of CD4+ memory T lymphocytes to CD8+ memory T subsets (CD4+/CD8+TM) in SAA patients was also lower. Severe aplastic anemia is a rare autoimmune disease characterized by severe pancytopenia and bone... more Severe aplastic anemia is a rare autoimmune disease characterized by severe pancytopenia and bone marrow failure, which is caused by activated T lymphocytes. Tregs are believed to control development and progression of autoimmunity by suppressing autoreactive T cells. This study aims at understanding the quantity and function of peripheral Tregs in SAA. The expression of related biomarkers on Treg cell surface were determined by flow cytometry. The frequency of Tregs and the expression of CTLA-4 in SAA was significantly decreased than that in normal controls. The expression of perforin in SAA was significantly increased than that in controls, while expression of CD39, CD73 and GITR in Tregs did not show any significant difference between the two groups. These data revealed that CTLA-4 could be responsible for Treg abnormalities in SAA, but suppression mediated by perforin, CD39, CD73 and GITR, and survival capability of single Treg cell may not be injured.

However, it is a great challenge to produce a large fluorescence anisotropy change upon the presence of target small molecules without nanoparticles -dependent amplification. This work reports a nanoparticle free and multiple G-enhanced fluorescence anisotropy assay for detection of DNAzyme activity. A Pb2+-dependent GR-5 DNAzyme was used as a model. We hybridized the rA-cleavable substrate strand containing a TMR label at 5'-end with the DNAzyme strand containing an extended three G bases at 3'-end. By this design, we demonstrate that both fluorescence quenching and the enhanced DNAzyme activity contribute to a Pb2+-induced large fluorescence anisotropy change (│Δr│= 0.168). To assess the effectiveness of recombinant human thrombopoietin (rhTPO) in severe aplastic anemia (SAA) patients receiving immunosuppressive therapy (IST). Methods. Eighty-eight SAA patients receiving IST from January 2007 to December 2012 were included in this retrospective analysis. Of these, 40 subjects received rhTPO treatment (15000 U, subcutaneously, three times a week). rhTPO treatment was discontinued when the platelet count returned to normal range. Hematologic response, bone marrow megakaryocyte recovery, and time to transfusion independence were compared. Results. Hematologic response was achieved in 42.5%, 62.5%, and 67.5% of patients receiving rhTPO and 22.9%, 41.6%, and 47.9% of patients not receiving rhTPO at 3, 6, and 9 months after treatment, respectively (P = 0.0665, P = 0.0579, and P = 0.0847, resp.). In most stain separation methods, specific color channels (for example: RGB, HSI, CMYK) or color deconvolution matrices are used to obtain different tissue contrasts between DAB- and hematoxylin-stained areas. However, these methods could produce incomplete separation or color changes because the color spectra of stains and co-localized stains overlap in histological images. Therefore, we proposed an automatic color-filtering to separate hematoxylin- and DAB-stained tissues. In this study, we investigated the quantity and function of osteoblasts by co-culture, the status of cellular immunity by flow cytometry, and the relationship between them in MM patients. The results showed that the numbers and function of osteoblasts in MM patients were lower than those in normal controls. The mildest phenotype includes only problems related to overproduction of uric acid. Quantities of whole B lymphocytes and CD5+ B lymphocytes as well as the expressions of Fas and Bcl-2 in B lymphocytes in 35 patients with untreated IRP, 15 IRP patients in complete remission (CR), and 10 normal controls were assayed by flow cytometry. Inspired by the excellent adhesive property of mussel adhesive protein, we added polydopamine (PD... more Inspired by the excellent adhesive property of mussel adhesive protein, we added polydopamine (PDA) to calcium phosphate cement (PDA-CPC) to enhance its compressive strength previously. The mineralization and mechanism on PDA-CPC were investigated by soaking it in simulated body fluid in this study. The results indicated that PDA promoted the conversion of dicalcium phosphate dihydrate and α-tricalcium phosphate to hydroxyapatite (HA) in the early stage but inhibited this conversion subsequently. PDA promoted the rapid mineralization on PDA-CPC to form a layer of nanoscale calcium phosphate (CaP) whereas there was no CaP formation on the control-CPC after 1d of soaking. This layer of nanoscale CaP was similar to that of natural bone, which was always observed during soaking. X-ray photoelectron spectroscopy showed that the peak of CO of PDA existed in the newly formed CaP on PDA-CPC, indicating the co-precipitation of CaP with PDA. Prior studies have suggested a strong correlation between residual hypoxanthine recycling activity and disease severity. However, the relevance of guanine recycling and compensatory changes in the de novo synthesis of purines has received little attention. In the current studies, fibroblast cultures were established for 21 healthy controls and 36 patients with a broad spectrum of disease severity related to HGprt deficiency. We assessed hypoxanthine recycling, guanine recycling, steady-state purine pools, and de novo purine synthesis. To identify the incidence of telomere attrition in SAA patients and investigate the relationship of telomere length with clinical parameters, SAA patients (n=27) and healthy controls (n=15) were enrolled in this study. Telomere length of PWBCs was significantly shorter in SAA patients than in controls. Analysis of gene expression of Shelterin complex revealed markedly low levels of POT1 expression in SAA groups relative to controls. No differences in the gene expression of the other Shelterin components-TRF1, TRF2, TIN2, TPP1, and RAP1-were identified. Addition of IFN-γ to culture media induced a similar fall in POT1 expression in bone marrow cells to that observed in cells cultured in the presence of SAA serum, suggesting IFN-γ is the agent responsible for this effect of SAA serum. Twenty-one newly diagnosed ITP patients, twenty ITP patients in recovery stage and eighteen normal controls were enrolled in this study. The percentages of Tfh cells, Tfh-related molecules ICOS, CD40L, IL-21 in BM were detected by flow cytometry (FCM), and the mRNA expression of BCL-6 in BMMNC was determined by semi-quantitive RT-PCR. Correlation of Tfh cell level with the disease severity of ITP patients was analysed. Immune-related pancytopenia (IRP) is characterized by pancytopenia caused by autoantibody-mediate... more Immune-related pancytopenia (IRP) is characterized by pancytopenia caused by autoantibody-mediated bone marrow destruction or suppression. The bone marrows of IRP patients have remarkably increased erythroblastic islands (EIs). We determined the immunoglobulin G (IgG) autoantibodies in some parts of EIs of IRP patients using immunofluorescence to investigate the biological function of EIs with IgG in the pathophysiology of IRP. The dominant class of autoantibodies detected in mononuclear cells was IgG (CD34 IgG, CD15 IgG, and GlycoA IgG), specifically IgG on GlycoA-positive cells (GlycoA IgG). Results show that extravascular hemolysis occurred in IRP through IgG autoantibodies in the EIs. These data included a high percentage of reticulocytes in the peripheral blood, hypererythrocytosis in the bone marrow, and high serum bilirubin. This study investigates the impacts of fire emission, convection, various climate conditions and ... more This study investigates the impacts of fire emission, convection, various climate conditions and transport pathways on the interannual variation of carbon monoxide (CO) in the tropical upper troposphere (UT), by evaluating the field correlation between these fields using multisatellite observations and principle component analysis, and the transport pathway auto-identification method developed in our previous study. The rotated empirical orthogonal function (REOF) and singular value decomposition (SVD) methods are used to identify the dominant modes of CO interannual variation in the tropical UT and to study the coupled relationship between UT CO and its governing factors. Both REOF and SVD results confirm that Indonesia is the most significant land region that affects the interannual variation of CO in the tropical UT, and El Niño-Southern Oscillation (ENSO) is the dominant climate condition that affects the relationships between surface CO emission, convection and UT CO. In addition, our results also show that the impact of El Niño on the anomalous CO pattern in the tropical UT varies strongly, primarily due to different anomalous emission and convection patterns associated with different El Niño events. In contrast, the anomalous CO pattern in the tropical UT during La Niña period appears to be less variable among different events. Transport pathway analysis suggests that the average CO transported by the "local convection" pathway ( CO local ) accounts for the differences of UT CO between different ENSO phases over the tropical continents during biomass burning season. CO local is generally higher over Indonesia-Australia and lower over South America during El Niño years than during La Niña years. The other pathway ("advection within the lower troposphere followed by convective vertical transport") occurs more frequently over the west-central Pacific during El Niño years than during La Niña years, which may account for the UT CO differences over this region between different ENSO phases.
ABSTRACT The new Level 3 aerosol profile data derived from the Cloud-Aerosol Lidar and Infrared P... more ABSTRACT The new Level 3 aerosol profile data derived from the Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observations (CALIPSO) provide a multiyear global aerosol distribution with high vertical resolution. We analyzed seasonal and diurnal variations of the vertical distributions of aerosol properties represented by 5-year CALIPSO data. Results show that dust, smoke, and polluted dust are the most frequently detected aerosol types during all seasons. Dust is the dominant type, especially in the middle to upper troposphere, over most areas during boreal spring and summer, while smoke and polluted dust tend to dominate during biomass burning seasons. The seasonal variations of dust layer top height and dust contribution to all-aerosol extinction are positively correlated with the seasonal variation of the dust occurrence frequency. The seasonal cycle of aerosol properties over west Australia is similar to that over biomass burning regime areas, despite its desert regime. In general, smoke is detected more frequently from the lower to middle troposphere; clean marine and polluted continental aerosols are detected more frequently, while polluted dust is detected less frequently, in the lower troposphere during nighttime than daytime. The all-aerosol extinction is generally larger, and the aerosol layer top is detected at high altitudes more frequently during nighttime than daytime. The diurnal changes of aerosol properties are similar within the same aerosol regime. Dust extinction shows little diurnal variation except when dust is the dominant aerosol type. The results contribute to an initial global 3-D aerosol climatology which will likely be extended and improved in the future.
Comparison of the upper tropospheric relative humidity (UTH) of CCM2 forced by observed sea surfa... more Comparison of the upper tropospheric relative humidity (UTH) of CCM2 forced by observed sea surface temperatures (SSTs) with data from TOVS, MSU, and the GEOS-1 assimilation shows that the model reproduces the observed large-scale pattern of interannual changes of UTH in the tropics, but gives too much drying during the 1987 El Niilo. How UTH responds to the changes of precipitation is well-simulated in the areas of anomalous subsidence and drying in the subtropical Pacific. However, the exaggerated increase of precipitation in the model in the equatorial central Pacific leads to excessive amplitudes of vertical motion, which dries the subtropical upper troposphere. Since the uplift does not result in excessive moistening in the equatorial Pacific, the effect of the too vigorous hydrological cycle is an unreali•c•y large reduction of UTH in the tropics during an E1 Nilto in CCM2.

Quantities of whole B lymphocytes and CD5+ B lymphocytes as well as the expressions of Fas and Bcl-2 in B lymphocytes in 35 patients with untreated IRP, 15 IRP patients in complete remission (CR), and 10 normal controls were assayed by flow cytometry. The percentages of B lymphocyte and CD5+ B lymphocyte were significantly higher in untreated IRP patients than in CR IRP patients and normal controls (P < 0.05), and there was no significant difference between the latter two groups (P > 0.05). Inspired by the excellent adhesive property of mussel adhesive protein, we added polydopamine (PD... more Inspired by the excellent adhesive property of mussel adhesive protein, we added polydopamine (PDA) to calcium phosphate cement (PDA-CPC) to enhance its compressive strength previously. The mineralization and mechanism on PDA-CPC were investigated by soaking it in simulated body fluid in this study. The results indicated that PDA promoted the conversion of dicalcium phosphate dihydrate and α-tricalcium phosphate to hydroxyapatite (HA) in the early stage but inhibited this conversion subsequently. PDA promoted the rapid mineralization on PDA-CPC to form a layer of nanoscale calcium phosphate (CaP) whereas there was no CaP formation on the control-CPC after 1d of soaking. This layer of nanoscale CaP was similar to that of natural bone, which was always observed during soaking. Prior studies have suggested a strong correlation between residual hypoxanthine recycling activity and disease severity. However, the relevance of guanine recycling and compensatory changes in the de novo synthesis of purines has received little attention. In the current studies, fibroblast cultures were established for 21 healthy controls and 36 patients with a broad spectrum of disease severity related to HGprt deficiency. We assessed hypoxanthine recycling, guanine recycling, steady-state purine pools, and de novo purine synthesis. To identify the incidence of telomere attrition in SAA patients and investigate the relationship of telomere length with clinical parameters, SAA patients (n=27) and healthy controls (n=15) were enrolled in this study. Telomere length of PWBCs was significantly shorter in SAA patients than in controls. Analysis of gene expression of Shelterin complex revealed markedly low levels of POT1 expression in SAA groups relative to controls. No differences in the gene expression of the other Shelterin components-TRF1, TRF2, TIN2, TPP1, and RAP1-were identified. Addition of IFN-γ to culture media induced a similar fall in POT1 expression in bone marrow cells to that observed in cells cultured in the presence of SAA serum, suggesting IFN-γ is the agent responsible for this effect of SAA serum. Twenty-one newly diagnosed ITP patients, twenty ITP patients in recovery stage and eighteen normal controls were enrolled in this study. The percentages of Tfh cells, Tfh-related molecules ICOS, CD40L, IL-21 in BM were detected by flow cytometry (FCM), and the mRNA expression of BCL-6 in BMMNC was determined by semi-quantitive RT-PCR. Correlation of Tfh cell level with the disease severity of ITP patients was analysed. The bone marrows of IRP patients have remarkably increased erythroblastic islands (EIs). We determined the immunoglobulin G (IgG) autoantibodies in some parts of EIs of IRP patients using immunofluorescence to investigate the biological function of EIs with IgG in the pathophysiology of IRP. The dominant class of autoantibodies detected in mononuclear cells was IgG (CD34 IgG, CD15 IgG, and GlycoA IgG), specifically IgG on GlycoA-positive cells (GlycoA IgG). Results show that extravascular hemolysis occurred in IRP through IgG autoantibodies in the EIs. These data included a high percentage of reticulocytes in the peripheral blood, hypererythrocytosis in the bone marrow, and high serum bilirubin. This study investigates the impacts of fire emission, convection, various climate conditions and ... more This study investigates the impacts of fire emission, convection, various climate conditions and transport pathways on the interannual variation of carbon monoxide (CO) in the tropical upper troposphere (UT), by evaluating the field correlation between these fields using multisatellite observations and principle component analysis, and the transport pathway auto-identification method developed in our previous study. The rotated empirical orthogonal function (REOF) and singular value decomposition (SVD) methods are used to identify the dominant modes of CO interannual variation in the tropical UT and to study the coupled relationship between UT CO and its governing factors. Both REOF and SVD results confirm that Indonesia is the most significant land region that affects the interannual variation of CO in the tropical UT, and El Niño-Southern Oscillation (ENSO) is the dominant climate condition that affects the relationships between surface CO emission, convection and UT CO. In addition, our results also show that the impact of El Niño on the anomalous CO pattern in the tropical UT varies strongly, primarily due to different anomalous emission and convection patterns associated with different El Niño events. In contrast, the anomalous CO pattern in the tropical UT during La Niña period appears to be less variable among different events. Transport pathway analysis suggests that the average CO transported by the "local convection" pathway ( CO local ) accounts for the differences of UT CO between different ENSO phases over the tropical continents during biomass burning season. CO local is generally higher over Indonesia-Australia and lower over South America during El Niño years than during La Niña years. The other pathway ("advection within the lower troposphere followed by convective vertical transport") occurs more frequently over the west-central Pacific during El Niño years than during La Niña years, which may account for the UT CO differences over this region between different ENSO phases.
ABSTRACT The new Level 3 aerosol profile data derived from the Cloud-Aerosol Lidar and Infrared P... more ABSTRACT The new Level 3 aerosol profile data derived from the Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observations (CALIPSO) provide a multiyear global aerosol distribution with high vertical resolution. We analyzed seasonal and diurnal variations of the vertical distributions of aerosol properties represented by 5-year CALIPSO data. Results show that dust, smoke, and polluted dust are the most frequently detected aerosol types during all seasons. Dust is the dominant type, especially in the middle to upper troposphere, over most areas during boreal spring and summer, while smoke and polluted dust tend to dominate during biomass burning seasons. The seasonal variations of dust layer top height and dust contribution to all-aerosol extinction are positively correlated with the seasonal variation of the dust occurrence frequency. The seasonal cycle of aerosol properties over west Australia is similar to that over biomass burning regime areas, despite its desert regime. In general, smoke is detected more frequently from the lower to middle troposphere; clean marine and polluted continental aerosols are detected more frequently, while polluted dust is detected less frequently, in the lower troposphere during nighttime than daytime. The all-aerosol extinction is generally larger, and the aerosol layer top is detected at high altitudes more frequently during nighttime than daytime. The diurnal changes of aerosol properties are similar within the same aerosol regime. Dust extinction shows little diurnal variation except when dust is the dominant aerosol type. The results contribute to an initial global 3-D aerosol climatology which will likely be extended and improved in the future.
Comparison of the upper tropospheric relative humidity (UTH) of CCM2 forced by observed sea surfa... more Comparison of the upper tropospheric relative humidity (UTH) of CCM2 forced by observed sea surface temperatures (SSTs) with data from TOVS, MSU, and the GEOS-1 assimilation shows that the model reproduces the observed large-scale pattern of interannual changes of UTH in the tropics, but gives too much drying during the 1987 El Niilo. How UTH responds to the changes of precipitation is well-simulated in the areas of anomalous subsidence and drying in the subtropical Pacific. However, the exaggerated increase of precipitation in the model in the equatorial central Pacific leads to excessive amplitudes of vertical motion, which dries the subtropical upper troposphere. Since the uplift does not result in excessive moistening in the equatorial Pacific, the effect of the too vigorous hydrological cycle is an unreali•c•y large reduction of UTH in the tropics during an E1 Nilto in CCM2.
