Abstract

The use of reliability of Random Amplified Polymorphic DNA (RAPD) method in analysis of genetic diversity has been employed in six potato cultivars and its wild species. A total of 30 random primers were used to produce 230 polymorphic bands. The results of research showed that there were a number of primers that could come up with specific bands on potato cultivars BF-15, Nicola, Cardinal, Aminca, SPV-10, Katahdin, and the wild species of S. phureja and S. stenotomum , but it was not found in other wild species (S. chacoense, S. sparsipilum, S. microdontum and S. verney). It also showed that there were a number of primers that could come up with specific bands in each of wild potatoes species but it was not found in the potato cultived. Clustering analysis using Unweighted Pair-Group Method arithmetic (UPGMA) showed that cultived potatoes clustering made group itself in the degree of similarity around 70 percent, and significantly similar with S. stenotomum and S. phureja than other wild species.
