二酸化炭素を固定し、有用な物質を生産するために、光独立栄 養生物であるシアノバクテリアに異種生物由来の酵素遺伝子群からなる合成代 謝経路を導入する研究が広く行われている。これらの合成代謝経路における出 発物質は、解糖系下流に位置する Pyruvate, Acetyl-CoA が用いられることが多 い。当研究室でも Synechococcus elongatus PCC 7942 を用いて Acetyl-CoA を出発 物質とした isopropanol 生産系の構築に成功している。しかし、一般的にシアノ バクテリアにおいて解糖系下流への代謝流束は、炭酸固定を行うカルビンベン ソン回路と比較して小さいとされており、isopropanol の生産量は低い。そこで、 本研究では、糖異化遺伝子群の転写調節因子に注目し、これを過剰発現するこ とで物質生産の向上を目指した代謝流束の改変を行うことを目的とした。 【方法】 相同性解析から、S. elongatus PCC 7942 で糖異化遺伝子群の転写調節因 子として機能していると考えられた crr37 を、相同組み換えにより導入し、過 剰発現させた。crr37 発現効果を評価するために Glycogen の蓄積量および、大 腸菌由来の phosphate acetyltransferase(pta)を導入した際の Acetate 生産量を比較 した。 【結果】 crr37 導入株の、Glycogen 量は、野生株に比べ 29.8%減少した。また pta-crr37 導入株と pta 導入株の Acetate 生産量は培養 21 日時点で 0.7 mM、1.4 mM となり、約 2 倍に増加した。このことから、crr37 は S. elongatus PCC 7942 において糖異化遺伝子群の転写調節因子として機能しており、過剰発現により 細胞の代謝流束改変が可能であることが示された。 Overexpression of a transcriptional regulator crr37 changes the metabolic flux in Synechococcus elongatus PCC 7942

Three kinds of extrusion-type friction tests were performed to evaluate friction shear factors at different kinds of extrusion dies, or at the backward extrusion punch. The first kind is a forward rod-backward can extrusion type friction test for evaluating friction at a taper die. The second kind is a new friction test based on forward rod-backward can extrusion with a conical die providing projections. The third kind is a friction test based on combined forwardbackward straight can extrusion. The third kind was modified to evaluate friction at the bearing of a backward extrusion punch quantitatively. The lubricity of conversion coating lubricant for low-carbon steel, the lubricities of conversion coating lubricant and various viscosity mineral oils for aluminum alloy were evaluated. The lubricities of the different lubricants were compared for each extrusion type. Surface expansion ratio does not always affect friction shear factor directly. The lubricity of conversion coating is almost the same for each extrusion type. On the other hand, the lubricity of mineral oils is markedly affected by the extrusion type. In the case of extrusion in which oil is difficult to retain, the lubricity decrease, and adhesion occurred.

Neostigmine in the hippocampus increases thermogenesis and T4-to-T3 conversion

The firing rate of the nerves innervating interscapular brown adipose tissue (IBAT), IBAT and colonic temperatures (TIBAT and TC), and O2 consumption were monitored in urethan-anesthetized male Sprague-Dawley rats. These variables were measured for 40 min before (baseline values) and 40 min after a neostigmine (5 x 10(-7) mol) or saline injection in the hippocampus. The blood level of 3,5,3'-triiodothyronine and L-thyroxine (T3 and T4) and the 5'-deiodinating activity of IBAT, liver, and kidneys were determined in other rats with neostigmine or saline injection. The results showed that neostigmine injection increased firing rate, TIBAT, TC, O2 consumption, blood level of T3, and 5'-deiodinating activity of IBAT. No change was found in the T4 level and in 5'-deiodinating activity of the liver and kidneys. Activation Mechanism of Receptors on the Biomembrane

The biological importance of membrane protein is emphasized in statistical analyses of the genomes that indicate about 30% of all open reading frames encode membrane proteins. For the purpose of performing structural biology on membrane proteins, we have been developing a methodology for chemical synthesis of membrane proteins. Now, accumulated knowledge and experience has enabled us to work on the structure and function of transmembrane-juxtamembrane region of membrane protein with solid state NMR and other spectroscopic techniques. Since little attention has been paid to this portion of protein as such, obtaining the structural information that provides a clue to fully understand how membrane protein, especially, receptor on the membrane, passes the structural change at the outside to the inside. In this review, we introduce our approach from peptide chemistry to work on structural biology of membrane protein.
