Inflamasi memiliki peran penting pada perkembangan penyakit dengan pravalensi tinggi. Penelitian ini bertujuan untuk mengetahui potensi aktivitas anti-inflamasi dari berbagai family tanaman herbal. Model yang digunakan untuk mengetahui aktivitas inflamasi secara in vivo adalah induksi edema pada hewan uji. Sedangkan model in vitro yang digunakan adalah makrofag terstimulasi lipooksigenase. Ekstrak yang diuji berasal dari tanaman Aloe barbadensis, Curcuma longa, Mimosa tenuiflora, Lavandula angustifolia, Psidium guajava, Punica granatum, Pseudoptergorgia elisabethae, Solanum lycopersicum, Capsicum frutescens, dan Zingiber zerumbet . ABSTRAK Kalkon merupakan golongan flavonoid yang mempunyai aktivitas biologis seperti antioksidan, antiinflamasi, dan antibakteri. Penelitian ini bertujuan untuk memprediksi profil farmakokinetik (absorpsi dan distribusi) serta toksisitas senyawa turunan analog kalkon dan pirimidin sebagai obat antiinflamasi. Prediksi parameter absorpsi dan distribusi menggunakan perangkat lunak PreADMET® dan sifat toksisitas menggunakan perangkat lunak Toxtree®. Penapisan senyawa berdasarkan kaidah Lipinski menunjukkan bahwa 24 senyawa hasil modifikasi memenuhi kaidah Lipinski, kecuali senyawa 22. Studi prediksi absorpsi dengan parameter HIA (Human Intestinal Absorption) menunjukkan semua senyawa terabsorpsi baik pada usus (70-100%), kecuali senyawa 4, 10, dan 11 moderate absorption (20-70%) dan senyawa 22 kurang terabsorpsi (0-20%). Probiotics have already proven beneficial effects in the treatment of several intestinal infections, but the underlying mechanisms how the probiotics can affect responses of porcine IPEC-J2 enterocytes to oxidative stress remained to be elucidated. The immunmodulatory effect of five bacterial strains (Lactobacillus plantarum 2142, Lactobacillus casei Shirota, Bifidobacterium animalis subsp. lactis BB-12, Bacillus amyloliquefaciens CECT 5940 and Enterococcus faecium CE-CT 4515) on 1 mM peroxide-triggered upregulation of interleukin (IL)-8 and tumor necrosis factor alpha (TNF-α) level was screened by q RT-PCR. Our data revealed that spent culture supernatant (SCS) of L. plantarum 2142 had significant lowering effect on IL-8 and TNF-α level with concomitant promoting activity on protective Hsp70 gene expression. According to our results, lactic acid (racemic, D-and L-lactic acid) and acetic acid produced by lactobacilli had no protective effect in quenching upregulation of proinflammatory cytokines. Furthermore, L. plantarum 2142-specific supernatant peptides were detected by gel electrophoresis and capillary zone electrophoresis.

Chronic inflammatory conditions such as obesity, type-2 diabetes, and cardiovascular diseases are the leading causes of mortality worldwide. Hence, much research interest in bioactive peptides has been stimulated due to lack of potent pharmacological interventions. Although many such peptides have been identified from food proteins, insufficient information is available on their structurefunction relationship. Presence of hydrophobic and positively charged amino acids is a common occurrence for the peptides with anti-inflammatory properties. However, inconsistent findings have also been reported. Most of the food-derived peptides exhibited their anti-inflammatory activities primarily by inhibiting signaling components of either NF-jB or MAPK pathway, which are the two major pathways involved in chronic inflammation following uncontrolled signal activation. This review highlighted the structural requirements of the peptides to exhibit antiinflammatory activity based on the current knowledge about food-derived anti-inflammatory peptides and their underlying molecular mechanisms of action. Practical applications While research in the food-derived bioactive peptide is gaining momentum, but the ability to translate these new findings into the commercial product such as nutraceuticals and functional foods, remains delayed. The most prominent reasons for this delay are the lack of detailed research on, (i) the structure-function relationship of the peptide and the underlying molecular mechanisms of these bioactive peptides, and (ii) the interaction of these peptides with different cellular elements in the disease pathophysiology. This review gives an insight into the structure-activity relationship of bioactive peptides involved in anti-inflammatory responses. The information provided here would be highly beneficial to describe the possible anti-inflammatory activity of any newly identified peptides from different food sources.
Isolation and characterization of anti-inflammatory peptides derived from whey protein

The present study was conducted to isolate and characterize anti-inflammatory peptides from whey protein hydrolysates using alcalase. Nine subfractions were obtained after sequential purification by ultrafiltration, Sephadex G-25 gel (GE Healthcare (Uppsala, Sweden) filtration chromatography, and preparative HPLC. Among them, subfraction F4e showed the strongest inhibitory activity on interleukin-1β (IL-1β), cyclooxygenase-2, and tumor necrosis factor-α (TNF-α) mRNA expression in lipopolysaccharide-induced RAW 264.7 mouse macrophages. Eight peptides, including 2 new peptides Asp-Tyr-Lys-Lys-Tyr (DYKKY) and Asp-Gln-Trp-Leu (DQWL), were identified from subfractions F4c and F4e using ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry, respectively. Our aim was to investigate anti-inflammatory effect of sevelamer in experimentally induced renal failure (RF) and whether this effect was related to a decrease in Escherichia coli colony counts in feces. Group 1 was the control group. Group 2 rats were those with induced RF receiving a high phosphate diet. Group 3 contained rats with induced RF receiving a high phosphate diet + sevelamer. E.coli count in feces, plasma creatinine, high-sensitive C-reactive protein (hsCRP), interleukin 6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha) concentrations were determined at the beginning and at the end of 1 st , 3 rd and 6 th weeks in all rats. R Re es su ul lt ts s: : In Group 2 and 3, creatinine concentrations were found to be increased (p=0.03, p=0.02) in the 1 st week. Asam salisilat memiliki aktivitas anti-inflamasi dan antioksidan, namun dapat menimbulkan efek samping pada saluran cerna. Modifikasi gugus karboksilat senyawa tersebut menjadi turunan amida dapat menjadi solusi untuk mengatasi kekurangannya. Pada penelitian ini dilakukan sintesis senyawa analog salisilamida,2‐hidroksi‐N‐(piridin‐2‐il)benzamida (1) dan turunan basa Mannich-nya (2a-b). Berdasarkan uji aktivitas penghambatan denaturasi protein senyawa sintesis menunjukkan aktivitas antiinflamasi, dengan rentang nilai IC50 = 0.121-0.145 mM. Aktivitas tersebut lebih rendah dibandingkan piroksikam yang digunakan sebagai senyawa standar (IC50 = 0,0073 mM). Hasil penambatan molekuler menunjukkan bahwa rasio afinitas ikatan COX-2/COX-1 dan interaksi ligan semua senyawa hasil sintesis tidak selektif terhadap COX-2.
Milk Bioactive Peptides: Antioxidant, Antimicrobial and Anti-Diabetic Activities

The main milk proteins are caseins and whey proteins, some other minor proteins and peptides, are also found in milk. Proteins are vital ingredients for human because they provide all the essential amino acids needed for body and human health. Milk proteins are very important sources of bioactive peptides. The bioactive peptides are inactive within the sequence of the parent protein and can be released by proteolytic enzymes, during gastrointestinal digestion or during milk processing, for example the adding coagulation enzymes and starter culture. Once bioactive peptides are present in the body, these peptides may act as regulatory compounds with hormone-like activity. Furthermore, Bioactive peptides from milk proteins have many biological activities such as antimicrobial, antihypertensive, antithrombotic, antioxidant, mineral binding, and anti-diabetic. Bioactive peptides have potential health and have pharmaceutical applications. Antimicrobial peptides are recognized as an important component of innate immunity, particularly at mucosal surfaces such as the lungs and small intestine that are constantly exposed to a range of potential pathogens. The ability of protein hydrolysates to inhibit deleterious changes caused by lipid oxidation appears to be related to the nature and composition of the different peptide fractions. Milk protein hydrolysate possesses free-radical-scavenging and anti-inflammatory activities have many beneficial effects on the increase of the glucose-induced insulin secretion and reduction in postprandial glycemia. This article is tried through exposure in some detail to review characteristics of some milk protein peptides and its positive effects on human health.

Increasing evidence on goat milk and the health benefits of its derived products beyond its nutritional value show its potential as a functional food. In this study, goat milk fractions were tested for their total antioxidant capacity using different methods (ORAC, ABTS, DPPH and FRAP), as well as their angiotensin-I-converting-enzyme inhibitory and antimicrobial (against Escherichia coli and Micrococcus luteus) activities. Different whey fractions (whey, cation exchange membrane permeate P and retentate R) of two fermented skimmed goat milks (ultrafiltered goat milk fermented with the classical starter bacteria or with the classical starter plus the Lactobacillus plantarum C4 probiotic strain) were assessed.
