Boysenberry ingestion supports fibrolytic macrophages with the capacity to ameliorate chronic lung remodeling

Lung fibrosis negatively impacts on lung function in chronic asthma and is linked to the development of pro-fibrotic macrophage phenotypes. Epidemiological studies have found that lung function benefits from increased consumption of fruit high in polyphenols. We investigated the effect of Boysenberry consumption, in both therapeutic and prophylactic treatment strategies in a mouse model of chronic antigen-induced airways inflammation. Boysenberry consumption reduced collagen deposition and ameliorated tissue remodeling alongside an increase in the presence of CD68+CD206+arginase+ alternatively activated macrophages in the lung tissue. The decrease in tissue remodeling was associated with increased expression of pro-fibrolytic matrix metalloproteinase-9 protein in total lung tissue. Effects of the Flavanone combination Hesperetin‐Naringenin, and Orange and Grapefruit Juices, on Airway inflammation and Remodeling in a murine asthma model

We investigated whether flavanones, hesperetin–naringenin, orange, and grapefruit juices reduce airway inflammation and remodeling in murine chronic asthma model. To establish chronic asthma, mice received house dust mite (HDM) for 3 days in 2 weeks, followed by twice per week for 4 weeks. Concurrently, during the last 4 weeks, mice received hesperetin plus naringenin (HN), orange plus grapefruit juice (OGJ), orange juice (OJ), or grapefruit juice (GJ); whereas the asthmatic control (AC) group and non‐asthmatic control (NC) group consumed water ad libitum. In histopathological examination, no goblet cells metaplasia was observed in the HN, OJ, and GJ groups; also, intra‐alveolar macrophages decreased compared with those of the AC group. Effects of raspberry fruit extracts and ellagic acid on respiratory burst in murine macrophages

The mechanism of action of polyphenolic compounds is attributed to their antioxidant, anti-inflammatory, and anti-proliferative properties and their effects on subcellular signal transduction, cell cycle impairment and apoptosis. A raspberry (Rubus idaeus L.) fruit extract contains various antioxidant active compounds, particularly ellagic acid (EA); however the exact intracellular mechanism of their action is not fully understood. The aim of the study was to evaluate the antioxidant effect of raspberry extracts, and that of ellagic acid by assessment of the production of the reactive oxygen species (ROS) by murine macrophage J774 cells. Raspberry extracts and their active compound EA did not affect or had very minor effects on cell viability. No significant difference in the ROS generation in arachidonic acid stimulated macrophages was determined for raspberry extracts and EA whereas in the phorbol-12 myristate-13 acetate model ROS generation was significantly (p < 0.05) reduced. Our observation that raspberry pomace extracts in vitro reduce ROS production in a J774 macrophage culture suggests that raspberry extract and ellagic acid mediated antioxidant effects may be due to the regulation of NADPH oxidase activity.

Blackcurrant proanthocyanidins augment IFN-γ-induced suppression of IL-4 stimulated CCL26 secretion in alveolar epithelial cells

Epidemiological studies reveal that fruit consumption reduces the prevalence of airway inflammation and childhood asthma. In particular, blackcurrant polyphenolic extracts have been shown to alleviate lung inflammation. Since IL-4-stimulated eotaxin-3 (CCL26) secretion is a major factor in the continuous eosinophil recruitment observed in atopic asthma, our focus was to evaluate the effectiveness of blackcurrant polyphenolic compounds on CCL26 secretion in human alveolar epithelial cells. Our results indicate that a proanthocyanin-enriched blackcurrant extract (BC-P), but not anthocyanin-enriched blackcurrant extract suppressed both IL-4-and IL-13-stimulated CCL26 secretion in a dose-dependent manner. Furthermore pre-incubation of cells with BC-P caused a time-dependent suppression of IL-4-stimulated CCL26 secretion. Moreover, epigallocatechin (EGC), and to a lesser extent epicatechin, metabolites identified in the proanthocyanidin extract, suppressed IL-4-stimulated CCL26 secretion. EGC was also effective at reducing the cellular phosphorylated STAT-6/STAT-6 ratio. Furthermore, both BC-P and purified EGC potentiated the ability of IFN-g to suppress IL-4-stimulated CCL26 secretion. The progression of an allergic immune response is complex, identifying plant compounds that target specific cellular events and complement the body's own immune actions is important for the development of functional foods. Our findings support the potential for blackcurrant polyphenolic compounds to reduce eosinophil recruitment and alleviate eosinophilic-driven airway inflammation.

The effects of proresolution of ellagic acid in an experimental model of allergic airway inflammation

Asthma is a disease of airway inflammation characterized by airway hyperresponsiveness, eosinophilic inflammation, and hypersecretion of mucus. Ellagic acid, a compound derived from medicinal plants and fruits, has shown anti-inflammatory activity in several experimental disease models. We used the classical experimental model, in BALB/c mice, of sensibilization with ovalbumin to determine the effect of ellagic acid (10 mg/kg; oral route) in the resolution of allergic airways response. Dexamethasone (1 mg/kg; subcutaneous route) was used as a positive control. The control group consisted of nonimmunized mice that received challenge with ovalbumin. Anthocyanins have several beneficial effects, especially on inflammatory and oxidative conditions. Fruit polyphenols, immunity and inflammation

Flavonoids are a large class of naturally occurring compounds widely present in fruits, vegetables and beverages derived from plants. These molecules have been reported to possess a wide range of activities in the prevention of common diseases, including CHD, cancer, neurodegenerative diseases, gastrointestinal disorders and others. The effects appear to be related to the various biological/pharmacological activities of flavonoids. A large number of publications suggest immunomodulatory and anti-inflammatory properties of these compounds. However, almost all studies are in vitro studies with limited research on animal models and scarce data from human studies. The majority of in vitro research has been carried out with single flavonoids, generally aglycones, at rather supraphysiological concentrations. Few studies have investigated the anti-inflammatory effects of physiologically attainable flavonoid concentrations in healthy subjects, and more epidemiological studies and prospective randomised trials are still required. This review summarises evidence for the effects of fruit and tea flavonoids and their metabolites in inflammation and immunity. Mechanisms of effect are discussed, including those on enzyme function and regulation of gene and protein expression. Animal work is included, and evidence from epidemiological studies and human intervention trials is reviewed. In Vitro and in Vivo Antioxidant and Anti-inflammatory Capacities of an Antioxidant-Rich Fruit and Berry Juice Blend. Results of a Pilot and Randomized, Double-Blinded, Placebo-Controlled, Crossover Study

The objective of this study was to compare three tests frequently used for evaluation of antioxidant potential in natural products: (1) oxygen radical absorbance assay (ORAC), (2) cell-based antioxidant protection in an erythrocyte model (CAP-e), and (3) reactive oxygen species formation in polymorphonuclear cells (ROS PMN). The methods were applied to four natural products, all containing antioxidants capable of entering and protecting cells in the CAP-e assay. The magnitude of this effect was not directly correlated to the ORAC value of each product. Furthermore, the products showed different effects in the ROS PMN assay. Açai provided strong inhibition of ROS formation, indicating anti-inflammatory properties. In contrast, Immunel and EpiCor mildly enhanced ROS formation, suggesting activation of the innate immune response. HA Joint Formula showed a complex, nonlinear dose-response in the ROS PMN assay. This illustrates that complex natural products may have similar antioxidant properties but different effects on human cells. Cell-based antioxidant protection is addressed best in the CAP-e assay, since some natural products contain compounds that may provoke cellular signaling in other cell types. The PMN cell type is a useful model for assessment of overall anti-inflammatory versus immune supportive properties of a product. The sequential use of the three methods serves to bridge analytical and biological testing methods.

In vitro assessment of the cytotoxicity and anti‑inflammatory properties of a novel dietary supplement

Studies on the immunopharmacological activities of various plant species have provided evidence for the high therapeutic potential of different extracts. These represent a promising alternative to reduce the inflammatory processes and, thus, diseases related to inflammation. Numerous scientific studies strongly suggest that diet plays an essential role in inflammation, and that certain dietary factors can act as preventive or treatment methods to lower inflammation. In the present study, a novel lingonberry-based dietary supplement was investigated for the ability to suppress the inflammatory response in activated monocytes/macrophages. Based on cell viability/proliferation and cytotoxicity tests, concentrations between 40 and 130 µg/ml of the extracts showed a high viability/proliferation effect and no cytotoxic activity in monocyte/macrophage cells. To further investigate the anti-inflammatory potential of our novel lingonberry-based dietary supplement, we studied the effect of the extract on the inflammatory response in lipopolysaccharide (LPS)-stimulated macrophages. We found that the extract exhibited a strong anti-inflammatory potential by inhibiting the expression of major inflammatory cytokines [interleukin (IL)-6, IL-8 and tumor necrosis factor (TNF)α] in activated monocyte/macrophage cells. The expression of IL-6 and IL-8 was subsequently validated by enzyme-linked immunosorbent assay (ELISA). In conclusion, we demonstrated that our product exhibits no cytotoxicity and suppresses inflammation, and thus can be considered a natural important tool for inflammation control.

In Vivo Anti-Inflammatory Potential of Viscozyme®-Treated Jujube Fruit

The fruit of Ziziphus jujuba, commonly called jujube, has long been consumed for its health benefits. The aim of this study was to examine the protective effect of dietary supplementation of enzymatically hydrolyzed jujube against lung inflammation in mice. The macerated flesh of jujube was extracted with aqueous ethanol before and after Viscozyme treatment. The extract of enzyme-treated jujube, called herein hydrolyzed jujube extract (HJE), contained higher levels of quercetin, total phenolics, and flavonoids, and exhibited more effective radical-scavenging abilities in comparison to non-hydrolyzed jujube extract (NHJE).
