A root-knot nematode parasitising kiwi fruit (Actinidia deliciosa) in Rio Grande do Sul State, Brazil and grape-vine (Vitis vinifera) in Chile, is identified as Meloidogyne ethiopica Whitehead, 1968. The species is redescribed from this new material and compared with the type description of M. ethiopica and another population of the species from Kenya. The female has an oval to squarish perineal pattern with a moderately high to high dorsal arch, smooth, widely spaced striae, and no 'shoulders'. The head region is set off with an elevated squarish labial disc, usually marked by annulations, and the stylet is 12-15 μm long, the cone usually being slightly curved dorsally and the shaft gradually widening posteriorly to near the junction with the rounded stylet knobs. The DGO is 3.0-5.0 μm behind the knobs. Meloidogyne luci n. sp. (Nematoda: Meloidogynidae), a root-knot nematode parasitising different crops in Brazil, Chile and Iran

A new root-knot nematode parasitising vegetables, flowers and fruits in Brazil, Iran and Chile, is described as Meloidogyne luci n. sp. The female has an oval to squarish perineal pattern with a low to moderately high dorsal arc and without shoulders, similar to M. ethiopica. The female stylet is robust and 15-16 μm long; the distance from the dorsal pharyngeal gland orifice to the stylet base (DGO) is 3-4 μm. Males have a high, rounded head cap continuous with the body contour. The labial disc is fused with the medial lips to form an elongated lip structure. The head region is not marked by incomplete annulations. Male stylet robust, 20.8-23.0 μm long with rounded knobs; the DGO is 2.5-4.5 μm. Meloidogyne polycephannulata n. sp. (Nematoda: Meloidogynidae), a root-knot nematode parasitizing carrot in Brazil

Meloidogyne polycephannulata n. sp. is described from specimens collected from an area cultivated with carrot cv. Brasilia, in the city of Rio Paranaíba, in the region of Alto Paranaíba, Minas Gerais State, Brazil. The perineal pattern of the female is circular to ovoid with a high dorsal arch that has widely spaced, coarse annulations. The lateral field may have a deep furrow separating the dorsal and ventral arches. The medial lips are short and wide, whereas the lateral lips are large and triangular. First report of, and additional information on, Meloidogyne konaensis (Nematoda: Meloidogyninae) parasitising various crops in Brazil

In a survey for Meloidogyne spp. in different crops from 11 regions in Ceará State, Brazil, using esterase isozyme electrophoresis as a specific identification method, four atypical populations were characterised from cabbage, papaya, noni and canapum plants, all of which showed an esterase profile different from those previously detected in Brazil. Morphological studies showed typical characteristics of Meloidogyne konaensis. Perineal patterns of females were variable, similar to M. arenaria and M. incognita, stylet length 14-20 μm. In females, the knobs gradually merged with the shaft and the dorsal pharyngeal gland orifice (DGO) ranged from 4 to 7 μm. Although males are not frequently found, the stylet morphology provides the most useful source of diagnostic character for the species, having 6-12 large projections protruding from the shaft. Genetic diversity of the root-knot nematode Meloidogyne enterolobii and development of a SCAR marker for this guava-damaging species

The objectives of this work were to evaluate the genetic variability of Meloidogyne enterolobii by molecular markers, and develop species-specific molecular markers for application in detection. Sixteen M. enterolobii isolates from different geographical regions (Brazil and other countries) and hosts were used in this study. The identification and purification of the populations were carried out based on isoenzyme phenotype. The DNA amplification of the intergenic region (IGS) of the rDNA and of the region between the cytochrome oxidase subunit II (COII) and 16S rRNA genes (mtDNA) produced specific fragments of the expected size for this nematode, i.e. 780 and 705 bp, respectively. Intraspecific variability among the isolates was evaluated with three different neutral molecular markers: AFLP, ISSR and RAPD. The results showed a low level of diversity among the isolates tested, indicating that M. enterolobii is a genetically homogeneous root-knot nematode species. The RAPD method allowed the identification of a species-specific RAPD fragment for M. enterolobii. This fragment was cloned and sequenced, and from the sequence obtained, a set of primers was designed and tested. The amplification of a 520-bp-long fragment occurred only for the 16 isolates of M. enterolobii and not for the 10 other Meloidogyne species tested. In addition, positive detection was achieved in a single individual female, egg-mass and second stage juvenile of this nematode. This SCAR species-specific marker for M. enterolobii represents a new molecular tool to be used in the detection of this nematode from field samples and as a routine diagnostic test for quarantine devices.

The cosmopolitan root-knot nematodes of the genus Meloidogyne are considered as the most important plant-parasitic nematodes damaging almost all higher plant species. This paper provides an updated checklist of 105 valid species of Meloidogyne, along with their synonyms and non-valid members. Furthermore, species are grouped based upon their diagnostic characteristics of second-stage juveniles (tail shape, tail length, stylet length and dorsal gland orifice (DGO) distance from stylet base) and males (number of lateral incisures, stylet length, DGO and spicules length).

A root-knot nematode parasitising yakon (Polymia sonchifolia) in São Paulo State, Brazil, is identified as Meloidogyne inornata. The species is redescribed from this material and compared with the original description of M. inornata. The female perineal patterns have a distinct, high, dorsal arch composed of smooth to wavy striae, similar to Meloidogyne incognita. The female stylet is 15.0-17.0 μm long with the cone generally slightly curved dorsally and with well developed knobs. DGO is 3.5-4.5 μm. Males have a high, rounded, head cap that is continuous with the body contour and has a large, round, centrally concave, labial disc raised above the medial lips. First report of rice root-knot nematode, Meloidogyne graminicola, infecting Juncus microcephalus in Brazil

Juncus microcephalus plants showing symptoms of root-knot nematode infestation were observed in the municipality of Agudo, Rio Grande do Sul state, Brazil. Based on morphological observation, esterase phenotypes, and molecular analyses of rDNA-ITS and D2-D3 regions of 28S rDNA, the causal agent of the observed symptoms was identified as Meloidogyne graminicola. Pathogenicity of M. graminicola was confirmed by fulfilling modified Koch’s postulates. To our knowledge, this is the first report of M. graminicola in J. microcephalus in Rio Grande do Sul State, Brazil.
