Assessment of Genetic Variability for Some Hyoscymus Species Using Biochemical and Molecular Markers (original) (raw)
Abstract
Hyocyamus sp. is an important medicinal plant belonging to the Solanaceae family. It contains tropane alkaloids, mainly hyoscyamine and scopolamine, which have medical usage. The primary objective of this study was to compare between proteins, isozymes as biochemical markers and RAPD-PCR molecular markers to assess the genetic diversity and relationships among five Hyoscyasmus species. SDS-PAGE for total soluble protein and PPO-isozyme analysis revealed low degree of polymorphism. In addition, some species from Hyocyamus sp. did not show any markers. A high degree of polymorphism was detected among the five Hyoscyasmus species through five random primers used for RAPD-PCR marker analysis. Fifteen monomorphic bands and 37 polymorphic distinct fragments (71.15% polymorphism) were revealed in the five Hyocyamus sp. with five primers.
