Asymmetric Synthesis of 2-Aryl-5-oxotetrahydrofuran-2-carboxylic Acids
Synthesis, 2006
3-Aryl-2-hydroxycyclopent-2-en-1-ones, when subjected to asymmetric oxidation, result in enantiomerically enriched 2aryl-5-oxotetrahydrofuran-2-carboxylic acids. Electron-donating substituents in the para position of the phenyl ring increase the yield and decrease the enantioselectivity of the process.
Synthesis of New Ethyl 3-Amino-4-arylfuran-2-carboxylates
Synthesis, 2002
An efficient method for the preparation of new ethyl 3amino-4-arylfuran-2-carboxylates is described. The synthesis proceeds via the corresponding hydroxyacrylonitrile sodium salt and an intermediate malonate vinyl ether. The last step of ring closure afforded the ethyl 3-amino-4-arylfuran-2-carboxylates in 15% to 40% yields.
Synthesis of 1Methyl3-oxo-7-oxabicyclo[2.2.1]hept-5-ene-2-carboxylic Acid Methyl Ester
Molecules, 2005
A simple and efficient method for the preparation of 1-methyl-3-oxo-7oxabicyclo[2.2.1]hept-5-en-2-carboxylic acid methyl ester (1) is described. The first step is a highly regioselective Diels-Alder reaction between 2-methylfuran and methyl-3-bromopropiolate. A remarkably difficult ketal hydrolysis reaction was effected by treatment with HCl, a simple reagent that was shown to be more efficient, in this case, than commonly used more elaborate methods.
Synthesis of 4-aminomethyl-tetrahydrofuran-2-carboxylates with 2,4-cis and 2,4-trans relationships
Tetrahedron, 2006
Templated tetrahydrofuran-based g-azido esters were prepared with the C-2 and C-4 functionalities in cis and trans relative configurations. This was achieved by ring contraction of the suitably protected 2-O-triflates of pentono-1,5-lactones (D-ribose and L-arabinose) with subsequent introduction of the azide via the 4-O-triflate. Access to a corresponding b-azido ester was achieved in good yield. Little elimination product was observed by introduction of the azide via the 3-O-triflate. These azido esters are scaffolds, which may be predisposed to adopt secondary structural motifs, for example, for use as peptidomimetics; they may also be utilised for the preparation of stereodiverse compound libraries. q Figure 1. Highly Stereoselective Synthesis oftrans-3-Aryl-4-carbethoxy-2,3-dihydro-2-fur-2′-oyl-5-methylfurans
Chinese J Chem, 2007
Multi-substituted dihydrofurans are valuable intermediates for the synthesis of natural products and pharmaceuticals. Considerable attention has been focused on the development of efficient and regioselective methods for their preparation. Using K 2 CO 3 as a base, with the reaction of fur-2-oylmethyltriphenylarsonium bromide 1 and ethyl 2-acetyl-3-arylacrylate 2 in tetrahydrofuran at room temperature, we found an efficient protocol was achieved to synthesize trans-3-aryl-4-carbethoxy-2,3-dihydro-2-fur-2'-oyl-5-methylfurans 3 in good yield with high stereoselectivity. The structure of compound 3 was confirmed by IR, 1 H NMR, MS and HRMS. The mechanism for the formation of 3 was proposed.

A series of 2-alkynyl2-diazo-3-oxobutanoates, when treated with a catalytic quantity of rhodium(I1) acetate, were found to produce furo[3,4-clfurans in good yield. The reaction proceeds by addition of a rhodium-stabilized carbenoid onto the acetylenic a-bond to give a vinyl carbenoid which subsequently cyclizes onto the neighboring carbonyl group to produce the furan ring. In a similar manner, indeno[ l,2-clfurans were produced from 2-butynyl 2-diazoarylacetates. Compounds substituted with methyl groups on the propargylic carbon atom cyclized at a slower rate. This rate diminution is attributed to the fact that the unsubstituted propargyl group can more easily achieve the required conformation for cyclization. C-H insertion was found to compete with furan formation when an alkyl group was attached to the keto functionality of the starting diazo acetoacetate. Diazo alkynyl sulfones undergo a novel oxygen transfer reaction when treated with rhodium(I1) acetate at 80 O C . The cyclized product is formed by sulfone oxygen attack onto the vinyl carbenoid producing a dipolar species which subsequently collapses to give the butenolide sulfoxide.
