Bioactive Molecules from Indian Medicinal Plants as Possible Candidates for the Management of Neurodegenerative Disorders

Natural Products - From Bioactive Molecules to Human Health [Working Title]

The present review gives an account of various bioactive molecules obtained from Indian medicinal plants for neurological degenerative disorders. Emphasis is laid on their correlation with the plants used in traditional system of medicine in India. The methodology involved in present review was enlisting of medicinal plants used for neurodegenerative disorders followed by their chemistry. A correlation with the chemical constituents and their recent findings has been done. Many medicinal plants such as Aloe vera and Bacopa monnieri have documented correlations and also need to be explored more. Molecules like garcinol (34), which was originally an anticancer compound, have good correlation as neuroprotective agent. Likewise many plants that have not been explored but are used in traditional system of medicine have also been listed. Jaggery and honey, which are used in traditional formulations in large quantity, also have natural products that are used as neuroprotective agents. In conclusion, a lot more study is required to correlate the medicinal plants and herbal formulations to have much more natural products for neurodegenerative disorders.

Ayurvedic medicine is a personalized system of traditional medicine native to India and the Indian subcontinent. Screening of antioxidant activity of three Indian medicinal plants, traditionally used for the management of neurodegenerative diseases

A number of Indian medicinal plants have been used for thousands of years in the traditional system of medicine (Ayurveda). Amongst these are plants used for the management of neurodegenerative diseases such as Parkinson's, Alzheimer's, loss of memory, degeneration of nerves and other neuronal disorders by the Ayurvedic practitioners. Though the etiology of neurodegenerative diseases remains enigmatic, there is evidence, which indicates that defective energy metabolism, excitotoxicity and oxidative damage may be crucial factors (Ann. Neurol. 38 (3) (1995) 357). The part of the Ayurvedic system that provides an approach to prevention and treatment of degenerative diseases is known as Rasayana , and plants used for this purpose are classed as rejuvenators. This group of plants generally possesses strong antioxidant activity (Pharmacol. Biochem. Behav. 43 (1992) 1175), but only a few have been investigated in detail. In the present study, three such rasayana plants were tested for the first time for their toxicity and free radical scavenging activity both in vitro and ex vivo. All the three plant infusions (up to 1 mg/ml) showed no toxic effects on the viability of PC12 cell line as judged by MTT-test. Both ethanolic extracts and water infusions of the plants were tested for their antioxidant activity in the 2,2?-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS + ') radical cation decolorization assay; inhibition of lipid peroxidation by plant infusions was carried out using spontaneous lipid peroxidation of rat brain homogenate, and IC50 values were determined. The results from the ABTS assay showed that the ethanolic extract of Sida cordifolia was found to be most potent (IC50 16.07 mg/ml), followed by Evolvulus alsinoides (IC50 33.39 mg/ml) and Cynodon dactylon (IC50 78.62 mg/ml). The relative antioxidant capacity for the water infusions was observed in the following order: E. alsinoides (IC50 172.25 mg/ml) /C. dactylon (IC50 273.64 mg/ml) /S. cordifolia (IC50 342.82 mg/ml). The results of water infusions of the plants on lipid peroxidation were as follows: E. alsinoides (IC50 89.23 mg/ml) /S. cordifolia) (IC50 126.78 mg/ml) /C. dactylon (IC50 608.31 mg/ml).

A changing lifestyle in food consumption due to the frequent taking of junk drinks based on the fastfood and ready-to-eat concept has brought about overloaded oxidative stress, thus weakening the body's defence mechanism to scavenge free oxygen radicals by destroying antioxidants. Oxidative stress occurs as a result of an imbalance between pro and antioxidant levels in favour of pro-oxidants resulting in the pathogenesis of neurodegenerative diseases. Though researches on the probable cure for neurodegenerative diseases are ongoing, fruits have been found to play an important dietetic role in preventing neurodegenerative diseases and other physiological disorders. Most fruits have their own valuable, unique active ingredients, having tremendous nutraceutical potentials and therapeutic implications. Therefore, people are required to make rational choices on the correct type of food for consumption to engender proper growth and development. Cognitive impairment, associated with ageing, stress, hypertension and various neurodegenerative disorders including Parkinson’s disease and epilepsy, is a major health issue. The present review focuses on Alzheimer’s disease (AD), since it is the most important cause of cognitive impairment. It is characterized by progressive memory loss, language deficits, depression, agitation, mood disturbances and psychosis. Pharmacological exploration of traditional plants for the treatment of neurodegenerative disorders

Alzheimer's disease (AD) is clinically characterized as memory deficits, altered behavior and impaired cognitive functions. The most important risk factor for AD is aging and mounting. Evidences suggested in different studies that traditionally used plants in Asia, China, and Europe significantly affect aging and AD involved neurodegeneration pathways. Research into ethnobotanicals for impaired memory and cognition has been burgeoned in last decades. The inclusion and exclusion criteria for the plant selection were based on reputed herbs recommended for treatment of neurological disorders and their scientific validation to cure neurodegenerative disorders. A range of traditional plants imparts effects via acetylcholinesterase activity, β-amyloid peptide formation in plaques, neurotrophic factors and through antioxidant activity. On one side preclinical investigations identified promising drug candidates for AD, on the other side, clinical evidences are still pending. Presently, according to WHO, around more than 80% world population relay on natural remedies to cure their health related issues. Plants contain rich source of primary and secondary metabolites for improving health problems. Pharmaceutical industry is facing intriguing challenges like elevated cost and unendurable risk management due to the high burden of neurodegenerative disorders. A significant shift of drug discovery is being witnessed from synthetic moieties to herbal formulation.

A study on less explored plants with potential neuroprotective activity against neurodegenerative diseases

The gradual deterioration of the structure and function of neurons, including neural death, is known as neurodegeneration. The most known diseases of neurodegeneration are Alzheimer’s, Parkinson's, Huntington's, Amyotrophic sclerosis lateral disease. These diseases are seen throughout the world including Bangladesh and for this reason the demand of neuroprotective agents are high. Now, there are some rare plants such as Ginkgo biloba, Withania somnifera Magnolia officinalis and Hypericum calycimum L. are used not only as folk medicine but also their extracts have shown some crucial effects in lab experiments. Some of the plants such as Centella asiatica, Withania somnifera and Hypericum perforatum L. decreases neural degeneration and also reverses amnesia. Here, this research is going to be discussing on the potential benefits of these plants as neuroprotective agents that are currently unclear or under research.
