Objective: The present work presents determination of physicochemical parameters of the leaves and the antioxidant properties of methanolic extract of leaves, fruits and bark of Rhus javanica L. Methods: The leaves of Rhus javanica L. was used for the determination of physicochemical parameters using standard protocol. The leaves, fruits and bark of the plant were extracted successively with petroleum ether, chloroform and methanol. The methanolic extracts were used to determine the antioxidant properties. The radical scavenging activity was determined with different methods like DPPH, hydrogen peroxide and hydroxyl radical scavenging assay. The reducing power was determined by reduction of ferric cyanide complex (Fe 3+) to the ferrous cyanide form (Fe 2+ Results: The moisture content, extractive value (alcohol and water), total ash, acid insoluble ash and water-soluble ash were 0.29%, 24.502%, 19.314%, 8.2%, 0.45% and 4.75% respectively. The fruit extract exhibits the highest scavenging activity presented as percentage inhibition, 75.00±0.89% (100 μg/ml) for DPPH free radical and 74.7±0.68% (100 μg/ml) for hydrogen peroxide radical. The standard BHT (100 μg/ml) shows 78.5±0.43% inhibition for DPPH free radical and 82.5±0.50% inhibition for hydrogen peroxide free radical. The leaves show the highest hydroxyl radical scavenging activity, 65.00±0.40% (100 μg/ml) while the standard Ascorbic acid exhibit 73.81±0.61% (100 μg/ml) inhibition. The highest reducing power was shown by the bark extract but lower than the standard Ascorbic acid. The bark extract shows the highest phenolic and flavonoid content 49.86 mg GAE/g and 19.38 mg QE/g respectively of the dry weight of the extract.). Total phenols and flavonoids concentrations were analysed using Folin-Ciocalteu's and aluminium chloride methods. Conclusion: The results of quantitative pharmacognostic parameters in this study is expected to be useful for setting standards for correct identification of the plant. The radical scavenging properties, total phenolic and flavonoid content also suggest that the extracts provide substantial antioxidant activities.
Phytochemical Study and Antioxidant Activities of Leaves Extracts from Rhamnus alaternus L
ABSTRACT : This study was designed to examine the chemical composition and in vitro antioxidant activities of leaves extracts from Rhamnus alaternus L.; we submitted two extracts (methanolic and aqueous) of different polarity to a deep compositional analysis through the use of an advanced hyphenated technique like LC/Uv-vis-DAD/MS, to our knowledge no metabolic fi ngerprint studies have been done on this species so far. So, we report for the fi st time the complete secondary metabolic fi ngerprint of R. alaternus polar extract, the chromatographic pattern from aqueous extract has several similarities with the methanolic one, two triglycoside fl avonoids were identifi ed in both extracts (kaempferol 3- O -[α-L-rhamnopyranosyl(1→3)- O-α-L-rhamnopyranosyl(1→6)]-β-D-galactopyranoside, rhamnetin 3- O -[α-L-rhamnopyranosyl(1→3)- O -α-L-rhamnopyranosyl(1→6)]-β-D-galactopyranoside), we note the presence of quercetin, kaempferol and rhamnetin derivatives. The samples were also subjected to a screening for their possible antioxidant activities by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and β-carotene-linoleic acid assays, in the first case the IC 50 value was of 0.082 ± 0.0006 mg/ml for the methanolic extract and 0.398 ± 0.007 mg/ml for the aqueous one, in the β-carotene-linoleic acid system, the inhibition values of linoleic acid oxidation were estimated as 89.007 ± 1.914% and 59.639 ± 3.824% for the methanolic and aqueous extract respectively, in both tests the methanolic extract was the most active. On the other hand, total phenolics determination in the test solutions was carried out according to the spectrophotometric Prussian blue assay and the determination of fl avonoids was performed using the method of aluminum trichloride, (AlCl 3 ) all the results indicated that the methanolic extract has higher total phenolics and fl avonoids being of: 33.655 ± 2.503 mg GAE/g for the fi rst method and 61.127 ± 1.217mg EQ/g; 129.681 ± 1.546 mg ER/g, for the second one. According to all these results, we conclude that there is a clear relationship between chemical composition of R. alaternus and its antioxidant activities. Rhus species are known in traditional medicine for their therapeutic virtue and their extracts showed numerous important properties including antimalarial, antimicrobial, antiviral, and hypoglycemic and anticonvulsant activities. Rhus tripartitum (Ucria) is a medicinal plant widely used in Tunisia folk medicine against chronic diarrhea and gastric ulcer. This study was designed to examine in vitro and ex vivo antioxidant, anti-inflammatory and anticancer activities of four extracts of Rhus tripartitum root cortex with increasing solvent polarity (hexane, dichloromethane, methanol and water). HPLC was used to identify and quantify phenolic compounds in Rhus extract. Water extract showed the highest antioxidant activity using oxygen radical absorbance capacity (ORAC method) with 8.95 ± 0.47 µmol Trolox/mg and a cell based-assay with 0.28 ± 0.12 µmol Trolox/mg as compared to the other fractions. In the present research work was to examine the possible antioxidant activities of the aqueous and ethanol leaf extract of Rhinacanthus nasutus (Linn.) Kurz. DPPH, Hydroxyl radical scavenging and reducing power assays were employed. The results showed that the DPPH activity of aqueous and ethanol leaf extract at the dose of 50μg/ml has exhibited in 63.81±0.013 and 79.36±0.028 inhibition with an IC50 value of 21.39 and 29.41μg/ml. The highest Hydroxyl radical scavenging activity showed aqueous and ethanol leaf extract at the dose of 50μg/ml has exhibited in 96.18±0.029 and 121.23±0.081 inhibition with an IC50 values of 30.19and 41.39μg/ml, the reducing power assay aqueous and ethanol leaf extract showed the 0.59 and 0.71 absorption at 700 nm extract at the dose of 50μg/ml suggested that promising antioxidant activity of crude aqueous and ethanol extract could be used as a source of natural antioxidants of R. nasutus and needs further studies to bring new natural products into pharmac...
