Phytotaxa, 2015
Curvularia is an important genus whose species are widely distributed phytopathogens as well as opportunistic pathogens on human and animals. The purpose of this study is to re-evaluate the phylogenetic relationships of the species in the genus Curvularia using ITS (nuclear ribosomal internal transcribed spacer), GPDH (glyceraldehyde-3-phosphate dehydrogenase) and TEF (translation elongation factor) gene regions and to provide modern descriptions and illustrations of Curvularia australis, Curvularia buchloës, C. cymbopogonis, C. hawaiiensis, C. neoindica, C. neergaardii, C. nicotiae, C. nodulosa, C. ryleyi, and C. subpapendorfii which lack recent descriptions with details of host and distribution. A multi-gene phylogenetic tree based on ITS, GPDH and TEF gene regions is used to define species of a fresh collections obtained from various hosts and geographic locations in the world. Both human and plant associated species of Curvularia are included in the phylogenetic analysis. Some s...
Curvularia mosaddeghii sp. nov., a novel species from the family Pleosporaceae
Mycosphere, 2018
The new species C. mosaddeghii sp. nov. isolated from plants of Syzygium cumini and Vigna unguiculata is described and illustrated. Three-locus DNA sequence based phylogeny, in combination with morphology of the asexual morph, were used to characterize this species. Phylogenetic analysis used combined sequences of internal transcribed spacer regions 1 & 2 and 5.8S nrDNA (ITS), partial glyceraldehyde-3-phosphate dehydrogenase (GPDH) and part of the translation elongation factor 1-α (EF1α). In the phylogenetic trees, both isolates of C. mosaddeghii clustered together as a monophyletic clade with strong support, distinct from other previously known species of Curvularia. Morphologically, this species is distinguished from closely related species by having narrower conidia and hila.
Three new Curvularia species from clinical and environmental sources
MycoKeys, 2020
Curvularia is a Pleosporalean monophyletic genus with a great diversity of species, including relevant phytopathogenic, animal and human pathogenic fungi. However, their microscopic identification is difficult due to overlapping morphological features amongst species. In recent years, multi-locus sequence analysis using the ITS region of the rDNA and fragments of the genes gapdh and tef1 revealed numerous cryptic species, especially in isolates that commonly produced 3-septate conidia. Therefore, based on sequence analysis of the above-mentioned DNA barcodes recommended for species delineation in Curvularia, we propose three novel species, C. paraverruculosa, C. suttoniae and C. vietnamensis, isolated from soil, human clinical specimens and plant material, respectively, collected in different countries. These new species are morphologically characterised and illustrated in the present study. Curvularia paraverruculosa differs from its counterparts, C. americana and C. verruculosa, m...
Novel Curvularia species from clinical specimens
Persoonia, 2014
The fungal genus Curvularia includes numerous plant pathogens and some emerging opportunistic pathogens of humans. In a previous study we used morphology and sequences of the nuclear ribosomal internal transcribed spacer region (ITS) and the glyceraldehyde-3-phosphate dehydrogenase (gpd) gene to identify species within a set of 99 clinical Curvularia isolates from the USA. Seventy-two isolates could be identified while the remaining 27 isolates belonged in three unclassified clades that were tentatively labelled Curvularia sp. I, II and III. In the present study, we further assess the taxonomic placement of these isolates using sequences of ITS, gpd, the large subunit rDNA, and the second largest subunit of RNA polymerase II. DNA sequence comparisons with a set of 87 isolates representing 33 Curvularia spp. and members of the closely-related genera Bipolaris and Exserohilum revealed that Curvularia sp. I, II and III represent novel lineages in Curvularia. These lineages are morpholo...
Curvularia ahvazensis and C. rouhanii spp. nov. from Iran
Mycosphere, 2018
Three isolates of Curvularia were obtained from diseased Eucalyptus sp., Syngonium vellozianum and Zinnia elegans. Phylogenetic trees inferred from combined dataset of the internal transcribed spacer ribosomal DNA (ITS), partial glyceraldehyde-3-phosphate dehydrogenase (GPDH), and partial translation elongation factor 1-α (EF1α) along with morphological characterization, showed they are two novel species. Curvularia ahvazensis sp. nov. and C. rouhanii sp. nov. are described and illustrated and shown to be morphologically distinct from previously known species of Curvularia.
Phylogenetic Study of Curvularia on Sorghum from Indonesia Based on ITS rDNA Sequence
Jurnal Mikologi Indonesia
Seven species of Curvularia were recorded from Indonesia, including species that pathogen to sorghum (Sorghum bicolor). The present study aims to determine four isolates of Curvularia isolated from sorghum in Indonesia using a combination of molecular phylogenetic analysis based on internal transcribed spacer (ITS) rDNA sequence and morphological examination. The results showed that the four isolates were genetically closed to C. lunata and C. dactyloctenicola. The morphological examination also insufficient to determine the identity of the four Curvularia isolates from sorghum until species level. Therefore, additional sequences from the partial fragments of the glyceraldehyde-3-phosphate dehydrogenase and the translation elongation factor 1-a genes are necessary to determine the identity of the Curvularia sequences isolated from sorghum in Indonesia until species level.
Pathogens, 2019
Members of the genus Curvularia are melanin-producing dematiaceous fungi of increasing clinical importance as causal agents of both local and invasive infections. This study contributes to the taxonomical and clinical knowledge of this genus by describing two new Curvularia species based on isolates from corneal scrapings of South Indian fungal keratitis patients. The phylogeny of the genus was updated based on three phylogenetic markers: the internal transcribed spacer (ITS) region of the ribosomal RNA gene cluster as well as fragments of the glyceraldehyde-3-phosphate dehydrogenase (gpdh) and translation elongation factor 1-α (tef1α) genes. The maximum likelihood phylogenetic tree constructed from the alignment of the three concatenated loci revealed that the examined isolates are representing two new, yet undescribed, Curvularia species. Examination of colony and microscopic morphology revealed differences between the two species as well as between the new species and their close relatives. The new species were formally described as Curvularia tamilnaduensis N. Kiss & S. Kocsubé sp. nov. and Curvularia coimbatorensis N. Kiss & S. Kocsubé sp. nov. Antifungal susceptibility testing by the broth microdilution method of CLSI (Clinical & Laboratory Standards Institute) revealed that the type strain of C. coimbatorensis is less susceptible to a series of antifungals than the C. tamilnaduensis strains.
Morphological and molecular characterization of Curvularia lunata pathogenic to andropogon grass
Bragantia
The fungal genus Curvularia is associated with a number of diseases in plants, commonly producing foliar spots in forage grasses. The objective of this study was to characterize the morphological and molecular diversity of the isolates of Curvularia sp. associated with Andropogon seeds, and to assess both their capacity to transmit disease and the pathogenicity of this fungus to crop. Ten isolates of Curvularia sp. were sourced from Andropogon seeds from agricultural producing regions in the Brazilian states Tocantins and Pará. Morphological characterization was achieved by observing fungus colonies and conidia and molecular characterization by DNA extraction and amplification with sequence-specific primers.
