The tautomerism of heteroaromatic compounds with five-membered rings—VIII
Tetrahedron, 1965
Ahsm-The constitution is discussed of the various compounds described in conflicting literature claims as phenyl-hydroxy-oxadiazoles, and authentic specimens of several of the isomers are described. Spectroscopic measurements and pK values show that 3-hydroxy-1,2,4-and-1,2,5_oxadiazoles exist predominately in the hydroxy-form, whereas Zhydroxy-1,3,4-and 5-hydroxy-1,2&oxidiazolcs occur mainly as the corresponding oxadiazolinones. These results are related to the general pattern of tautomeric behaviour in the hydroxy-azole series.
A 13C NMR study of tautomerism in phenoxymethylisoxazol‐5‐ones
Organic Magnetic …, 1979
A 'C NMR study of new isoxazal-5-ones has been carried out in different solvents. Mathematical analysis of the tautomeric ring proton exchange in pyridine solvent indicates that the relative contributions of the NH, CH and OH tautomers are respectively 1Ok5, 15k3, 1Ok8%, while the anionic form represents the remaining 65 * 8%. Recently we have shown by I3C NMR that in methanol or dimethyl sulphoxide (DMSO) solution, the most stable form (with respect to the heterocycle) of 3-[(o-tert-butylphenoxy)methyl]pyrazol-5-ones is the 5-hydroxypyrazole. This result was already accessible by more routine spectroscopic methods which have demonstrated that the latter tautomer predominates.' In the case of isoxazol-5-ones the following three tautomeric structures are possible: R3 3; N< 1 R3x; "N\o R3x:H "0 (A)
Ab initio study of the tautomerism of 2,5-substituted diazoles
Structural Chemistry, 2010
Detailed investigation of the tautomerism of oxadiazoles, thiadiazoles, and selenadiazoles containing hydroxy, mercapto, or selenyl groups in position 2 and 5 of the five-membered ring was carried out at MP2, MP4, and CCSD(T) levels of theory. The relative stabilities of the tautomers of the heterocycles were investigated in gas phase. It is found that the dioxo form is the preferred tautomer if the exocyclic group is hydroxy. If the exocyclic group is mercapto or selenyl group the most stable tautomer is the mixed mercapto/thione or selenyl/selone form. The barriers of intramolecular proton transfer are too large and tautomerization should not occur.
Theoretical studies on tautomerism of 1H-pyrazole-5-thiol
Structural Chemistry, 2013
The chemical properties of the 1H-pyrazole-5thiol have been extensively studied. In the present study, the relative stability of all tautomers in the gas phase and solution (PCM model) are evaluated at DFT and MP2 levels using the 6-311G??(d, p) basis set. The results suggest that 1H-pyrazole-3-thiol (B2 form) and 1H-pyrazole-5-thiol (A form) are the most stable forms in the gas phase and 1,2dihydropyrazole-3-thione (C form) and 1H-pyrazole-3-thiol (B2 form) are the predominant tautomers in solution at the DFT and MP2 methods, respectively. In addition, variation of dipole moments in the gas phase and solution, the specific solvent effects with addition of one molecule of water near the electrophilic centers of tautomers, the transition state of proton transfer assisted by a water molecule and the NBO charges of atoms were investigated. In addition, it was found that aggregation with water molecule does not change the order of stability of isomers, but proton-transfer reaction assisted by a water molecule needs less energy than the intramolecular process. The heats of formation, dipole moments, polarizabilities and ionization potentials of 96 compounds, eight theoretically possible tautomeric forms of N-unsubstituted pyrazolones (hydroxypyrazoles) and 11 of their C-substituted derivatives, are calculated by means of the MNDO method, with and without configuration interaction (CI). The MNDO+ CI results for the relative stabilities are in agreement with the available experimental data. They predict that in all cases the most stable in the vapour phase are hydroxypyrazole forms, excepting 3-and 4-phenyl substituted compounds, among which the most stable are the 2-pyrazoline-5one and 1-pyrazoline-3-one forms, respectively.
A theoretical study of substituent effects on tautomerism of 2-hydroxybenzimidazoles
Journal of Molecular Modeling, 2003
The geometries, relative stabilities of some 4(7) and 5(6) substituted 2-hydroxybenzimidazole derivatives were calculated with full geometry optimization using AM1 and PM3 in aqueous phase. With the exception of molecules 4, 6 and 7 for all the 4(7) and 5(6) substituted 2-hydroxybenzimidazole derivatives the 3H and keto forms were found to be favored.
1,3,4-oxadiazoles: evaluation of aromaticity and atomic charge distribution
Molecular Physics, 2008
The aromaticity and atomic charge distribution were investigated for the mono-and disubstituted 1,3,4oxadiazoles. Using an electron density-based approach (B3LYP), the structures of the molecules were estimated, then the changes of -electron delocalization of the heterocyclic ring were estimated based on geometric (HOMA) as well as magnetic properties (NICS). Aromaticity of the oxadiazole ring varies to some extend depending on the electron character of the substituent, however, the two methods do not correlate well. The HOMA approach suggests a non-aromatic character for the oxadiazole ring, while NICS predicts a relatively high aromaticity. Additionally, two computational methodologies were used for quantitative measures of atomic charges, i.e. AIM and GAPT. Results of these computations reveal accumulation of electron density inside the heterocyclic ring due to substitution by the aryl groups and its significant removal in the presence of the imino group. However, both procedures suggest different explanation of this fact.

The compound has been synthesized and characterized by IR, NMR and X-ray diffraction. Quantum chemical calculations at B3LYP/6À311++G(d,p) level were performed to study the molecular and spectroscopic properties, conformational equilibrium, thione M thiol tautomerism and intermolecular double proton transfer reaction of the compound. The obtained structural and spectroscopic results are well in agreement with the experimental data. The solvent effect on the proton transfer reaction was investigated in three solvents using the polarizable continuum model approximation and solvent-assisted mechanism. The anti-thione tautomer is the most stable isomer among the four possible structural forms both in the gas phase and in solution phase. A high tautomeric energy barrier is found for the tautomerism between the anti and syn forms of the compound, indicating a quite disfavored process. Although the presence of one methanol or water solvent molecule significantly lowers the energy barrier, it is not adequate for the reaction to occur.
Annular Tautomerism of 3(5)-Disubstituted-1H-pyrazoles with Ester and Amide Groups
Molecules, 2019
A series of disubstituted 1H-pyrazoles with methyl (1), amino (2), and nitro (3) groups, as well as ester (a) or amide (b) groups in positions 3 and 5 was synthesized, and annular tautomerism was investigated using X-ray, theoretical calculations, NMR, and FT-IR methods. The X-ray experiment in the crystal state showed for the compounds with methyl (1a, 1b) and amino (2b) groups the tautomer with ester or amide groups at position 3 (tautomer 3), but for those with a nitro group (3b, 4), tautomer 5. Similar results were obtained in solution by NMR NOE experiments in CDCl3, DMSO-d6, and CD3OD solvents. However, tautomer equilibrium was observed for 2b in DMSO. The FT-IR spectra in chloroform and acetonitrile showed equilibria, which can be ascribed to conformational changes of the cis/trans arrangement of the ester/amide group and pyrazole ring. Theoretical analysis using the M06-2X/6-311++G(d,p) method (in vacuo, chloroform, acetonitrile, and water) and measurement of aromaticity (NI...
