Chemistry and Biological Activities of 1,3-Benzothiazoles

Benzothiazoles and its derivatives are an important group of heterocyclic compounds that exhibit a wide range of pharmacological properties such as antitumor, antimicrobial, antidiabetic, anticonvulsant and anti-inflammatory. These different biological applications for benzothiazole compounds have motivated new efforts in search for novel derivatives with improved biological activity and diverse applications in pharmaceutical industry. Owing to the importance of this system, the aim of this review is to highlight aspects reported on the chemistry and biological activity of benzothiazoles during the past few years (2000-2010).

Summary Benzothiazole, a heterocyclic ring consisting of N and S at symmetrical position have been studied extensively owing to their interesting pharmacological activities. This review article covers the most active Benzothiazoles derivatives that have shown considerable biological actions such as antimicrobial, anti-inflammatory, anticancer, antilipidemic, antiviral, antihypertensive, and Antitubercular. This review also discusses the structure-activity relationship of the most potent compounds. It can act as an important tool for medicinal chemists to develop newer compounds possessing Benzothiazoles moiety that could be better agents in terms of efficacy and safety.

Benzothiazole analogues and their biological aspects: A Review

1659-1669Heterocyclic compounds analogues have attracted strong interest in medicinal chemistry due to their pharmacological properties. Benzothiazole belongs to the heterocyclic class of bicyclic compounds. It is a combination of two rings six membered and five membered and both the rings are responsible for the therapeutic activity. Different methods are used to synthesize benzothiazole compounds and have been found to have numerous biological activities like – anticancer, antimicrobial, anti-inflammatory, anti-leishmanial, antidiabetic activity. Benzothiazole is a class of heterocyclic compound having two hetero atoms namely sulphur and nitrogen. It is a two membered ring heterocyclic moiety, weak base. The analogues of benzothiazole and its derivatives have a significant role in research area especially in synthetic, medicinal and pharmaceutical chemistry because of its biological and pharmacological properties. So, in present the researchers are interested to work in this field.

Synthesis of some biologically active benzothiazole derivatives

N-(4,5-dihydro-1H-imidazol-2-yl)-6-substituted-1,3-benzothiazol-2-amines and N-(1Hbenzimidazol-2-yl)-6-substituted-1,3-benzothiazol-2-amines were synthesized by the reaction of 6-substituted-2-aminobenzothiazoles with carbon disulphide and methyl iodide. It was followed by the reaction with o-phenylene diamine/ ethylene diamine. All the synthesized compounds were characterized by elemental analysis, IR spectra, 1H NMR and MASS spectral studies. They were screened for their anti-inflammatory, antiulcer, antitumor, entomological (antifeedant, acaricidal, contact toxicity and stomach toxicity) and antibacterial activities.

Synthesis, Characterization and Biological Activity of Various Substituted Benzothiazole Derivatives

A series of 2-(benzo[d]thiazol-2-ylthio)-N-(2-oxoindolin-3-ylidene)acetohydrazide (3a-3g), 2-(benzo [d]thiazol-2-ylthio)-N-(2,4'-dioxospiro[indolin-3,2'-thiazolidin)-3'yl)acetamide (4a-4g) and 2'-((benzo[d]thiazol-2-ylthio)methyl)spiro[indoline-3,5'thiazolo[4,3-b][1,3,4]oxadiazol]-2-ones (5a-5g) have been synthesized and screened for their anti-inflammatory, analgesic and antibacterial activities. The most potent antiinflammatory and antibacterial compound of this series was compound 5d and most potent analgesic compound was compound 5e. Structures of all the compounds were established by elemental and spectral (IR and 1 H NMR) analysis.

Different heterocycle compound are made to synthesize by large number of efforts and their derivatives were found to posses anti-microbial, anti-anthelmintic anti-convulsant, anti-diabetic, anti-tumor activities. Benzothiazole moiety is very small but it possesses different biological activities but not only benzothiazoles but its different substituted derivatives also give different biological activities. Benzthiazoles are remarkably effective compounds, extensive structural modification and pharmacological studies have confirmed that these molecules are effective against various strains of microorganisms. This review was focus on the benzothiazole moiety and its different derivatives that posses different biological activities.

In recent year several methods have been developed for the synthesis of Benzothiazole derivatives due to their importance in bioorganic and medicinal chemistry. The present review focuses on systematic literature report of various synthetic procedures of Benzothiazoles derivatives using different strategies from the period 2010 to 2021.
