Glyoxylic acid (5 mol%) performs as a novel highly water-soluble catalyst for the synthesis of 2-aryl-1-arylmethyl-1Hbenzimidazoles from a wide range of substituted o-phenylenediamines and various substituted aldehydes in good to excellent isolated yields (85-95%) using water as solvent at ambient temperature. The remarkable advantages offered by this method are easily and inexpensive available catalyst, simple procedure, mild conditions, much faster (20-40 min) reactions and excellent yields of products.

Chemistry and Applications of Benzimidazole and its Derivatives

This chapter describes contemporary strategies for selective catalytic intermolecular functionalization of the benzimidazole scaffold. Functionalization at nitrogen and position C-2 is well developed employing copper, palladium, rhodium, nickel, and cobalt catalysis. Direct CH activation is the predominant approach to C-2 functionalization. Nickel-based catalysts can activate CO bonds in conjunction with C-H activation at benzimidazole which grants access to a very broad range of phenols and enols as convenient functionalization precursors in this chemistry. The remaining carbon positions of benzimidazoles are typically functionalized via a sequential halogenation/coupling strategy to ensure selectivity. A key success factor in enabling these chemistries has been the fine-tuning of catalystligand combinations.

Benzimidazole is the heterocyclic compound formed from benzene and imidazole ring containing nitrogen, oxygen sulphur and its derivatives are of wide interest because of their diverse biological activity. A new method for synthesis of 2-(substituted phenyl)-1Hbenzo[d]imidazole (3a-k) are developed by using simple method, green approach in good yields. We have synthesized 33 molecules in gram scale. This method is extremely useful for the synthesis of benzimidazole derivatives in excellent yields.

From the group of heterocyclic compounds, benzimidazoles remain the central focus, which owns the important features as six-five member attached rings having two nitrogen atoms at 1,3 position in six-member benzene and five-member imidazole fused ring system. Molecules having benzimidazole derivatives plays significant role as therapeutic agent & also showed excellent application in clinical and biological studies. These classes of scaffolds are those which hold several pharmacological properties like analgesics, antimicrobial, antiviral, antidiabetic, antihistamine, antiparasitic, anticancer, and also they are used for neurology, endocrinology, ophthalmology, cardiovascular illness and in many other therapeutic areas. Benzimidazoles got an increased attention due to their outstanding bioavailability, stability and important biological efficiency. This article reveals and discuss about typical synthetic methods established for benzimidazole class of scaffolds &their biological and therapeutic applications illustrated on variety of benzimidazole products.
