Tocotrienols belong to the vitamin E family of chemicals known to have potent anti-proliferative and apoptotic activities against a variety of cancer cells with little to no comparable influence on the normal cells. Whether tocotrienols control the expression of phase II antioxidant enzymes in the context of their anti-carcinogenic mechanisms has not been investigated. The present studies were performed to test whether the differential growth inhibition resulting from exposure to α-, γ- and δ-tocotrienols in estrogen receptor-positive human MCF-7 and estrogen receptor-negative MDA-MB-231 breast cancer cells might be accompanied by changes in phase II antioxidant enzymes. Cell proliferation and clonogenicity in both cell lines were significantly inhibited by γ- and δ-tocotrienols with little affect when cells were similarly exposed to α-tocotrienol, at doses up to 10 μM. Vitamin E, a fat-soluble compound, possesses both antioxidant and non-antioxidant properties. In this study we evaluated, in intestinal HT29 cells, the role of natural tocopherols, α-Toc and δ-Toc, and two semi-synthetic derivatives, namely bis-δ-Toc sulfide (δ-Toc)2S and bis-δ-Toc disulfide (δ-Toc)2S2, on TNFα-induced oxidative stress, and intercellular adhesion molecule-1 (ICAM-1) and claudin-2 (Cl-2) expression. The role of tocopherols was compared to that of N-acetylcysteine (NAC), an antioxidant precursor of glutathione synthesis. The results show that all tocopherol containing derivatives used, prevented TNFα-induced oxidative stress and the increase of ICAM-1 and Cl-2 expression, and that (δ-Toc)2S and (δ-Toc)2S2 are more effective than δ-Toc and α-Toc. Tocotrienol (Tocomin) are naturally occurring analogues of vitamin E family and has been reported to possess a potent free radical scavenging activity. In the present study we have initially investigated protective role of tocotrienol against infection and inflammation induced alterations in tissues antioxidant defense system, as well as speculated, via in silico docking studies, that tocotrienol can act by directly binding to antioxidant enzymes. Syrian hamsters were injected with bacterial lipopolysaccharide (LPS, 200 microg), zymosan (20 mg), or turpentine (0.5 ml) to mimic acute infection, acute systemic inflammation, and acute localized inflammation, respectively, which are responsible for the generation of plenty of free radicals that causes oxidative stress. Antioxidative activities of tocotrienols on phospholipid liposomes

Antioxidative activity of tocotrienol (Toc3) was studied in the oxidation of dilinoleoylphosphatidylcholine (DLPC) liposomes. The objective was to measure the differences in the antioxidative activities betweenα-Toc3 andα-tocopherol (α-Toc), and betweenγ-Toc3 andγ-Toc. When each antioxidant was added to the already prepared DLPC liposome solution, the antioxidative activity of Toc3 was larger than that of Toc. However, when incorporated into the liposomal membrane, the antioxidative activities of Toc3 and Toc were the same and were intermediate between those of the added Toc3 and Toc. When added to the liposome solution, the consumption of Toc3 during the induction period was larger than that of Toc. When incorporated into the liposomal membrane, the consumptions of Toc3 and Toc were the same and were intermediate between those of the added Toc3 and Toc. These results suggest that the reactions of Toc3 and Toc with phospholipid peroxide within the membrane are inhibited to a different degree depending on the dosing manner of Toc3 and Toc. Namely, the degree of inhibition decreases in the following order: Toc(added)> Toc(incorporated)= Toc3(incorporated)> Toc3(added).
