A series of 5-(pyridin-4-yl)-3-thioacetamide-1,2,4-triazoles (4a-o), was synthesized, characterized and evaluated for screening antibacterial and antifungal activities on microorganisms, respectively on five bacteria and and candida tropicalis. The structures of original compounds were confirmed by analytical and spectroscopic (FT-IR, 1 H NMR and 13 C NMR) methods and elemental analysis. Both the antibacterial and antifungal activities and MIC values of compounds were reported. Among the tested compounds, the most effective compound providing a MIC values of 64 μg mL-1 are 4a,b aganist B. subtilis and S. typhimurium and 4o against E. coli only.

1-Naphthyl-2-cyanoacetamide 1 reacts with arylidene malononitrile to afford a novel 2-oxo-1,2-dihydropyridine-3,5-dicarbonitrile derivative 6. Heating of 1 under reflux with the 1,3-diketones acetylacetone and benzoylacetone furnished the corresponding 3-cyano-2-pyridones derivatives 7 and 8, respectively. Fusion of 1 with 2 mol malononitrile afforded pyridinylacetamide 9. Condensation of 1 with nitrosonaphthols or salicylaldehyde afforded naphthoxazines 11 and 13, or chromene 17, respectively. Coupling of 1 with 4,6-dimethyl-1H-pyrazolo[3,4-b]pyridin-3-diazonium chloride gave the hydrazone 15, which cyclized with acetic acid to afford the corresponding pentaaza derivative 16. Treatment of 1 with DMF-DMA gave acrylamide 19, which when reacted with hydrazine hydrate, o-phenylenediamine, thiourea, or guanidine hydrochloride to afford the corresponding 3-aminopyrazole, diazepine, and pyrimidine derivatives 20, 21, 22, and 23 respectively. The newly synthesized compounds were characterized by elemental analysis and use of spectral data (IR, 1 H NMR, 13 C NMR, and MS).The newly synthesized compounds were tested for their in-vitro antibacterial activity against Gram-positive (Staphylococcus aureus and Bacillus subtilis) and Gram-negative (Pseudomonas aeruginosa and Escherichia coli) bacteria. The results clearly showed that most of the synthesized compounds had mild to moderate activity against the bacteria.

Utility of succinic acid dihydrazide for the synthesis of some new heterocyclic compounds, containing bis-1,3,4-oxadiazole, bis-4-thiazolidinone, bis-5-amino-3-pyrazolone and bis-1,3-dioxo-isoindole. The chemical structures of the prepared compounds have been confirmed by their elemental analysis, FT-IR, 1H NMR and Mass spectra. Investigation of the antimicrobial activity of the compounds was done by the paper disc technique. Some of the tested compounds showed the most favorable antimicrobial activity.
Hybrid Approach for Synthesis and Antimicrobial Activity of Heterocyclic Compounds

We have synthesized novel series of N-(1-(2-(1-phenyl-3-(p-tolyl)-1H-pyrazol-4-yl)-5-(pyridin-4-yl)-1,3,4-oxadiazol-3(2H)-yl)ethylidene)arylaniline and their derivatives. The structures of synthesized compounds were well characterized by spectroscopic techniques. Antimicrobial activity of the newly synthesized derivatives was evaluated against gram positive (S. aureus and S. pyogenes), gram negative bacteria (E. coli and P. aeruginosa), and strains of fungi (C. albicans, A. niger and A. clavatus). Among the screened derivatives 5c, 5f, 5i, 5l and 5t demonstrated superior antimicrobial activity against microbial strains.

5-Hydroxy-3-methyl-1-(4,6-dimethylpyrimidin-2-yl)pyrazol-4-yl-1,3-butanedione 4, obtained by the rearrangement of 2-(4,6-dimethylpyrimidin-2-yl)-1-[1-(4-hydroxy-6-methylpyran-2-one-3-yl)ethylidene]hydrazine 3, afforded 1-(4,6-dimethylpyrimidin-2-yl)-1′-aryl/heteroaryl-3,3′-dimethyl-(4,5′-bipyrazol)-5-ols (6) in excellent yields after treatment with various aryl/ heteroaryl hydrazines. The structures of the compounds were established by nuclear magnetic resonance (1 H and 13 C), infrared, and mass spectral data, and by elemental analysis. All the compounds synthesized were screened for their antibacterial activity against two Grampositive bacteria (Staphylococcus aureus and Bacillus subtilis) and two Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa), and their antifungal activity against two yeasts (Candida albicans and Saccharomyces cerevisiae).

Heterocyclic compounds play an important role in our life due to their biological importance in the struggle of microorganisms. Herein, a series of novel hybrid compounds of thienopyrimidine with triazine and pyrimidine scaffolds were synthesized starting from difunctionalized compound 5-amino-4-phenyl-2-(p-tolylamino)thieno[2,3d]pyrimidine-6-carbonitrile (1). Moreover, the diazotization of compound 1 with sodium nitrite in an acidic medium gave the chloro-triazine compound 2 which was subjected to the nucleophilic substitution of chlorine atom with different nucleophiles delivered compounds 3a-5c. Furthermore, the reaction of compound 1 with carbon disulfide led to the formation of dithione derivative 6 which was alkylated with ethyl chloroacetate to give compound 7, on the other hand, the reaction of compound 1 with phenyl isothiocyanate produced 4-imino-3,9-diphenyl-7-(p-tolylamino)-3,4dihydropyrimido[4',5':4,5]thieno [2,3-d]pyrimidine-2(1H)-thione (8), while acylation of the amino group in compound 1 with acetic anhydride gave compound 9. All synthesized compounds were characterized by elemental and spectral analysis techniques (IR, 1 H NMR, 13 C NMR, Mass spectroscopy). Furthermore, the synthesized compounds were tested for their antimicrobial activity against different strains of bacteria and fungi, and the results obtained showed good to moderate activity with almost all the strains.
SYNTHESIS AND ANTIBACTERIAL EVALUATION OF NOVEL HETEROCYCLIC COMPOUNDS
The work describes QSAR and SAR studies on the reserve transcriptase inhibitor by using the 2D-topological, physicochemical and hydrophobic parameters along with the indicator parameters. For the synthesis of new heterocyclic compounds containing a sulfonamido moiety suitable for use as antibacterial agents, the precursor ethyl {[4-N-(4,6-dimethylpyrimidin-2-yl)sulfamoyl] phenylazo}cyanoacetate was reacted with a variety of active methylene compounds producing pyran, pyridine and pyridazine derivatives. Also, the reactivity of the precursor hydrazone towards hydrazine derivatives to give pyrazole and oxazole derivatives was studied. On the other hand, treatment of the same precursor with urea, thiourea and/or guanidine hydrochloride furnished pyrimidine and thiazine derivatives, respectively. Treatment of 2,3-diaryloxirane-2,3-dicarbonitriles 1a–c with different nitrogen nucleophiles, e.g., hydrazine, methyl hydrazine, phenyl hydrazine, hydroxylamine, thiosemicarbazide, and/or 2-amino-5-phenyl-1,3,4-thiadiazole, afforded pyrazole, isoxazole, pyrrolotriazine, imidazolothiadiazole derivatives 2–5, respectively. Reacting pyrazoles 2a–c with aromatic aldehydes and/or methyl glycinate produced Schiff’s bases 7a–d and pyrazolo[3,4-b]-pyrazinone derivative 8, respectively. Treating 7 with ammonium acetate and/or hydrazine hydrate, furnished the imidazolopyrazole and pyrazolotriazine derivatives 9 and 10, respectively. Reaction of 8 with chloroacetic acid and/or diethyl malonate gave tricyclic compound 11 and triketone 12, respectively. On the other hand, compound 1 was reacted with active methylene precursors, e.g., acetylacetone and/or cyclopentanone producing adducts 14a,b which upon fusion with ammonium acetate furnished the 3-pyridone derivatives 15a,b, respectively. Some of newly synthesized compounds were screened for activity against bacterial and fungal strains and most of the newly synthesized compounds showed high antimicrobial activities. The structures of the new compounds were elucidated using IR, 1H-NMR, 13C-NMR and mass spectroscopy.
