An Assessment of Antibacterial Activity of Four Endodontic Sealers on Enterococcus faecalis by a Direct Contact Test: An In Vitro Study

Aim. To evaluate the antibacterial activity of four endodontic sealers on Enterococcus faecalis by a direct contact test. Material and Methods. Enterococcus faecalis was used as a test organism. Direct contact test which is based on measuring the effect of close contact between test bacteria and tested material on the kinetics of bacterial growth was performed to overcome the disadvantages of agar diffusion test. The sealers tested were zinc oxide eugenol-based sealer, glass-ionomer-based sealer, polydimethyl-siloxane-based sealer, and urethane dimethacrylate resin-based sealer. Data was collected by recording the optical density with the help of a spectrophotometer. Results. The sealers exhibited different inhibitory effects. The results obtained were subjected to statistical analysis by Kruskal Wallis analysis of variance and Dunn's multiple comparison test. In vitro antibacterial activity of endodontic sealers

Objectives. To evaluate the antibacterial activity of four endodontic sealers: one epoxy resin sealer (AH Plus), two zinc oxide eugenol (ZOE)-based sealers (Endomèthasone, Pulp Canal Sealer), and one sealer containing both ZOE and orthophenilphenol (Vcanalare). Methods. A direct contact test (DCT) was performed. A 10 ml suspension of Enterococcus faecalis was placed on the test material 20 min, 24 h and 7 days after mixing. Bacteria were allowed to directly contact the sealers for 1 h at 37 8C. Bacterial growth was then spectrophotometrically measured every 30 min for 7 h, and again after 24 h as well.

Antibacterial Properties of Four Endodontic Sealers

The purpose of this study was to evaluate the antimicrobial effects of root canal sealers. The direct contact test (DCT) was used to assess the antibacterial properties of AH plus, Apexit Plus, Epiphany SE, and Roe-koSeal when in contact with Enterococcus faecalis. The materials were examined immediately after setting and 1, 2, 7, and 14 days after aging in phosphate-bufferedsaline. Statistical analysis with two-way analysis of variance, one-way analysis of variance, and Tukey multiple comparison was applied to the data. Apexit Plus had a short-term antibacterial effect of 1 day on E. faecalis, whereas Epiphany SE enhanced bacterial growth for at least 7 days. AH plus and RoekoSeal were ineffective. Antibacterial activity of three endodontic sealers with various bases

The antibacterial activity of three endodontic sealers, a Ca(OH)2 based sealer (apexit) a Zoe based sealer (Drofill) and a resin based sealer (AH26) was assessed on the growth of an anaerobic bacteria (peprostreptococcus spp.) using the Agar diffusion test (ADT). Materials and Method: Thirty brain heart diffusion agar plates were incubated with peptostreptococcus anaerobics. Each plate was divided into two separate areas. In one area, 0.1ml droplet of a given fresh sealer and in the other area a dry material of the same sealer were placed, such that each plate had two areas with the same sealer in two forms of fresh and dry (setform). There were ten plates for each of the three sealers. The plates were placed into anaerobic jars and incubated at 37°C. After varying periods, zone of inhibition of bacterial growth were observed, measured and compared by t and Paired T-test. Results: There was no statistically significant differences between the activity of AH26 and drofill in the fresh form(P>0.05). However there were significant differences between (the two sealers in set form) (P<0.05) Drofill was more antibacterials in the set form than AH26. All sealers were more active when fresh than ages. Conclusion: It is likely that the eugenol in the Dorifill and the formaldehyde in the AH26 are responsible for their greater antimicrobial activity.

In Vitro Evaluation of Antimicrobial Activity of Sealers and Pastes Used in Endodontics

The antimicrobial activity of four root canal sealers (AH Plus, Sealapex, Ketac Endo, and Fill Canal), two calcium hydroxide pastes (Calen and Calasept), and a zinc oxide paste was evaluated. Seven bacterial strains were used, six of them standard; Micmcoccus luteus ATCC 9341, Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, Staphylococcus epidemidis ATCC 12228, Escherichia coli ATCC 25922, and Entemcoccus faecalis ATCC 10541. There was a wild strain of Streptococcus mutans isolated from saliva obtained in an adult dental clinic. Activity was evaluated using the agar diffusion method with Brain Heart Infusion agar and Muller Hinton medium seeded by pour plate. Calcium hydroxidebased sealers and pastes were either placed directly into 4.0 x 4.0 mm wells or by using absorbent paper points. The plates were kept at room temperature for 2 hr for diffusion. After incubation at 37°C for 24 hr, the medium was optimized with 0.05 g% l T C gel and inhibition haloes were measured. All bacterial strains were inhibited by all materials using the well method. However, when the materials were applied with absorbent paper points, Enterncoccus faecalis was not inhibited by zinc oxide, and Pseudomonas aeruginosa was not inhibited by AH Plus, Fill Canal, and the zinc oxidebased paste. We conclude that sealers and pastes presented antimicrobial activity in vitro and culture medium optimization with 0.05 g% l T C gel facilitated observation of the inhibition haloes.

An Antimicrobial Activity Assessment of Three Endodontic Sealers on Enterococcus faecalis, Candida albicans, and Staphylococcus aureus by a Direct Contact Test: An In Vitro Study

Introduction: For a successful endodontic treatment, antimicrobial property of endodontic filing is mandatory. Hence, the aim of this in vitro study was to assess the antibacterial efficacy of three endodontic sealers on Enterococcus faecalis, Candida albicans, and Staphylococcus aureus by direct contact test (DCT). Materials and Methods: The antimicrobial efficacy of three different sealers, AH Plus, mineral trioxide aggregate (MTA) Fillapex, and Endosequence BC was tested against E. faecalis ATCC 29212, C. albicans ATCC 10231 and S. aureus ATCC 25923 by DCT. Freshly mixed sealers were placed in flat bottom test tubes incubated at 37°C. Thereafter, 10 μL of suspension was drawn and spread over cultural plates to determine the colony count using a digital colony counter. An in vitro Comparison of Antimicrobial Activity of Three Endodontic Sealers with Different Composition

Aims: The aim of the study was to compare the antimicrobial property of newly introduced EndoSequence BC sealer with commonly used zinc oxide-eugenol-based sealer (Zical) and epoxy resin-based sealer (MM-Seal) against Candida albicans, Lactobacillus, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. Materials and methods: The agar diffusion test was done to measure the antimicrobial activity of sealers. The sealers were put in the 4 mm wells prepared in the inoculated agar plates. The agar plates were incubated at 37°C for 24 hours and the zones of inhibition that appeared was measured. Chi-square test was done to evaluate intraobserver bias for all study samples. Intergroup comparison was done for all five parameters using Pearson correlation statistical analysis. Results: EndoSequence BC sealer showed maximum mean of diameter of zones of inhibition against all the microorganisms but with no statistically significant difference with other sealers. Zical did not show any zone of inhibition against the P. aeruginosa. MM-Seal did not show any inhibitory activity against the P. aeruginosa and C. albicans. Conclusion: EndoSequence BC sealer showed antimicrobial activity against all the microorganisms and proved to be a better choice when compared with other two sealers. Clinical significance: Antimicrobial properties of endodontic sealers are important to prevent the persistent infection of the complex root canals. EndoSequence BC sealer has been proved to be a better sealer in this aspect.

In vitro comparison of antibacterial properties of bioceramic-based sealer, resin-based sealer and zinc oxide eugenol based sealer and two mineral trioxide aggregates

Our goal of the study was to evaluate the antibacterial properties of endodontic sealers against the E. faecalis. Six millimeters wells were made for each material in all the preinoculated petri plates. Then, the petri plates were incubated for 24 h. The zones of inhibition appeared were measured, and the measurements were put to statistical analysis. EndoSequence BC Sealer, MM-mineral trioxide aggregate (MTA), and ProRoot MTA showed maximum means of diameter of zones of inhibition, whereas MM-seal and Endoseal did not show any zones of inhibition. EndoSequence BC Sealer was found to be a better endodontic sealer as compared to resin-based and zinc oxide-eugenol-based sealer.
