Comparison of Mucoadhesive Buccal Patches of Ondansetron HCl with Conventional Marketed Tablets

The purpose of this study was to design and evaluate sustained release mucoadhesive buccal patches of Ondansetron hydrochloride to bypass first pass metabolism thereby enhancing its bioavailability. The patches were fabricated using mucoadhesive polymers PVA, tamarind gum and chitosan along with a hydrophilic polymer PVP K30 by solvent casting method. A comparative in-vitro drug release study carried out with conventional marketed tablet showed that the prepared patches sustained the release upto 8hrs. The patches showed good flexibility and prolonged release upto 8hrs thereby achieving therapeutic efficacy and improved patient compliance. The sustained release patches are useful in reducing dose, dosing frequency and direct delivery of drug into the systemic circulation.
Taste Masking and Formulation of Ondansetron Hydrochloride Mouth Dissolving Tablets

This study was done to mask the bitter taste of ondansetron HCl using complexing agent, a polacrilex resin: Tulsion 335 and subsequently forming mouth dissolving tablet using superdisintegrants: Croscarmellose sodium and sodium starch glycollate. A preliminary screening was done. Batch process, a most preferential method for drug loading with ion exchange resins was selected. The process was optimized for drug: resin ratio to get maximum drug loading. A ratio of drug: resin at 1:3 was selected. Taste evaluation was carried out by selecting volunteers. Drug resin complex (DRC) was evaluated for drug release. The resultant DRC was formulated by direct compression into mouth dissolving tablet using microcrystalline cellulose PH 102, as diluent and croscarmalose sodium and sodium starch glycolate as superdisintegrants and aspartame was used as sweetening agent to enhance palatability. Objective: The aim of the present study was to prepare the ondansetron hydrochloride Mouth Dissolving Tablets (MDTs) followed by its comparison with ethical and non-ethical (generic) marketed tablets. Methods: Prior to the formulation, drug excipient compatibility study was carried out by FTIR spectroscopy. The λmax was determined by UV spectroscopy. The ondansetron hydrochloride MDTs were prepared by direct compression method using Sodium Starch Glycolate (SSG) as super disintegrant and camphor as a sublimating agent. Then the prepared MDTs were subjected to evaluation of post compression parameters such as thickness and diameter, weight variation, wetting time, hardness, friability, disintegration and dissolution. Medicated Chewing Gum: A Novel Drug Delivery System

In the recent years scientific and technological advancements have been made in the research and development of oral drug delivery systems. The reasons that the oral route achieved such popularity may be primarily due to its ease of administration. Chewing gum is one of the very popular oral confectionary products. It is a potentially useful means of administering drugs either locally or systematically via, the oral cavity. The medicated chewing gum has through the recent years gained increasing acceptance as a drug delivery system. Chewing gum known as gum base (insoluble gum base resin) contains elastomers, emulsifiers, fillers, waxes, antioxidants, softners, sweeteners, food colorings, flavoring agents, and in case of medical chewing gum, active substances. It offers various advantages over conventional drug delivery systems. Unlike chewable tablets, medicated chewing gums are not supposed to be swallowed and may be removed from the site of application without resorting to invasive means. Moreover medicated chewing gums require the active and continuous masticatory activities for activation and continuation of drug release. An In-vitro apparatus was specially designed and constructed for release testing of medicated chewing gums. Medicated chewing gums are excellent mobile drug delivery systems for self-medication as it is convenient and can be administered discretely without water.

The potential of chewing gum as a drug delivery system together with different formulation principles and methods of assessment are discussed in this article. The release of a drug from chewing gum is dependent upon its water solubility. Water soluble substances are released rapidly and completely from chewing gum and methods are available which retard their release from chewing gum to provide an extended release profile. Slightly water-soluble drugs are released slowly and incompletely from chewing gum and require special formulation techniques to produce a satisfactory release profile. Studies evaluating the potential application of medicated and non-medicated chewing gum in the treatment of local diseases in the oral cavity are described. Specific examples of the use of chewing gum as a delivery system for dental health, smoking cessation and antifungal therapy are cited. Few drugs are suitable candidates for incorporation into chewing gum formulations for the intention of their systemic delivery. Know-how derived from the development and manufacture of already existing medicated and non-medicated chewing gum, supplemented with today's knowledge of the principles of pharmaceutical formulation, constitute the basis for the development of the medicinal chewing gum of tomorrow.
Mouth Disintegrating Tablets of Taste-Masked Ondansetron HCl
Ondansetron HCl is used in the management of nausea and vomiting induced by cytotoxic chemotherapy and radio-therapy and is bitter drug. The purpose of research work was focused on taste masking of the Ondansetron HCl and further development of the drug in mouth disintegrating tablets. Ion exchange resins, Indion 204 and Indion 234 were used with a view to mask the taste of the drug. Tablet formulations were prepared using wet granulation and direct compression tech-niques. All formulations were subjected to post compression parameters like uniformity of thickness, hardness, friability test, weight variation and drug content uniformity, all these parameters were within pharmacopoeial limits. Medicated chewing gum: An unconventional drug delivery system INTRODUCTION
Medicated chewing gum has a history for about a century. Now-a-days it is considered to be a potential and convenient modified release drug delivery system which can be used in pain relief medication, smoking cessation, travel illness, freshening of breath, prevention of dental caries, alleviation of xerostomia, vitamin or mineral supplementation etc. Medicated chewing gums are prepared by using a water insoluble gum base with water soluble bulk portion. This formulation offers both local and systemic effects and has a range of advantages over conventional oral solid dosage forms. USP currently has no in vitro release testing apparatus for the evaluation and determination of drug release from the prepared chewing gums. But European Pharmacopoeia adopted a compendial apparatus to do so. Medicated chewing has drawn attention to the researchers as potential drug delivery system and it could be a commercial success in near future.
