Review: Solid Dispersion Technique for Enhancement of Solubility of Poorly Soluble Drug

Nearly 40% of novel drugs comes in pharmaceutical industries are showing poor capability of solubilization in water. Therefore enhancing the solubilization of such drugs in water to enhance their bioavailability be the major challenge to formulation scientists. So the preparation of solid dispersion from the drug which shows poor solubility in water with carriers having good water solubility has decrease the occurrence of such problems and increase dissolution. Hence solid dispersion found to be attention-grabbing method for solubility enhancing of drugs which showing poor solubility in water. This review, shows an overview on the different solid dispersion types, rationale, their advantages, limitations, manufacturing processes as well as its characterization methods.

The solid dispersion has become a momentous solubilization technology for poorly water soluble drugs.It is usually two component system of drug polymer, which interacts between the drug design and its performance. Solid dispersion in water-soluble carriers have interacted a means of developing the dissolution rate and bioavailability of the hydrophobic drugs. The aim of this present study is to compare the solubility of different drugs with the increased polymer concentration. The solubility behavior of drugs remains one of the most exigent aspects in formulation development. With the advent of combinatorial chemistry and high throughput screening, the number of poorly water soluble compounds has dramatically increased. Solid Dispersions: an Approach to Enhance Solubility of poorly Water Soluble Drug

Solid dispersions have attracted considerable interest as an efficient means of improving the dissolution rate and hence the bioavailability of a range of poorly water-soluble drugs. Solid dispersions of poorly water-soluble drugs with water-soluble carriers has reduced the incidence of these problems and enhanced dissolution. The focus of this review article is on the advantages, limitations, various methods of preparation and characterization of the solid dispersion. The different types of solid dispersions based on their molecular arrangement have been highlighted. Some of the practical aspects to be considered for the preparation of solid dispersions, such as selection of carrier and methods of physicochemical characterization have also been discussed. In this review, it is intended to discuss the future prospects related to the area of solid dispersion manufacturing.

Solid dispersion is used for enhancing dissolution rate of a therapeutically active substance and in turns its absorption and in vivo efficacy. Solid dispersion is generally prepared with drug which is having poor aqueous solubility and hydrophilic carrier. Generally Polyethylene Glycol, Polyvinyl Pyrrolidone, Mannitol, Urea, Gums, Eudragit are used as hydrophilic carriers. Certain Hydrophilic Swellable Polymers Sodium Carboxy Methyl Cellulose, Pregelatinized Starch, Sodium Starch Glycolate are also used. Sometimes surfactant is also added to further improve wetting property of solid dispersion. In solid dispersion particle size of drug is reduced or a crystalline pure drug is converted into amorphous form and hence the solubility of drug is increased. Solid dispersion is not only used in improving dissolution rate of poorly water soluble drug but also in masking the taste of the drug substance, preparing rapid disintegration oral tablets and in producing sustained release microspheres. Various methods are available to prepare solid dispersion generally solvent evaporation method, melting method, melt solvent method, kneading method, cogrinding method, co-precipitation method, modified solvent evaporation method, spray drying, gel entrapment technique, co-precipitation with supercritical fluid. Evaluations of solid dispersion are done by Fourier Transform infra-red spectroscopy, X-Ray diffractometry, scanning electron microscopy, differential scanning calorimetry, solubility and dissolution experiments.

The dissolution rate of a drug directly depends upon its solubility. Most of the new drugs have poor water solubility; thereby pose a difficulty in formulating into drug delivery systems. Therefore, solubility enhancement of poorly water soluble drugs is one of the necessary preformulation steps in the pharmaceutical product development research. Solid dispersion is a unique and promising approach for enhancing the dissolution characteristics and oral bioavailability of poorly water-soluble drugs. The present review highlights portrayal of solid dispersions, its types including eutectic mixtures and solid solutions, method of preparation and also the useful carriers for the preparation of solid dispersions. Furthermore, the wide research conducted hitherto on solid dispersions for enhancing solubility of different efficacious drugs, challenges encountered in the development of solid dispersions and recent advancements to overcome its pitfalls have also been elaborated.

A review on solid dispersion and latest approaches used to improve solubility of poorly water soluble drugs

In the present day solubility of drugs in GIT fluids becomes necessary for the preparation of solid unit dosage forms of drugs. Otherwise if the drug has poor solubility then the drug does not show its effect properly in the body. So, solubility is a main aspect in the formulation of unit doses. Various methods like Lyophilization (Freeze drying process), Super critical fluid technology, Nanotechnology, Liquisolid compact technology, Self-emulsifying agents (SMEDDS) etc. are used to improve the solubility and dissolution rate of poorly water soluble drugs.

A CRITICAL REVIEW ON DIFFERENT PHARMACEUTICAL ASPECTS OF SOLID DISPERSION TECHNIQUE FOR SOLUBILITY ENHANCEMENT

Solid dispersion is the prerequisite technique for increasing the dissolution and bioavailability of the hydrophobic drugs. The technique generally provides more significant enhancement in solubility or dissolution rate as compared to other techniques which mainly relies only on the reduction of particle size. The factor of re-crystallization associated with reduction of particle size has great negative impact of further reduction of solubility but in more generalized view along with reduction of particle size the factor which increases the wettability by reducing the contact angle between particle and water is mainly important and solid dispersed particles are formulated with this approach only as they composed of matrix dispersion of hydrophobic drug and hydrophilic polymer which increases the wettability of drug molecules and also reduces the size to form molecular dispersion. The recent research in the field of polymerization has also made it possible to make available those polymers which allows intermolecular hydrogen bonding with drug molecules in the solid dispersed particles, these bondings have significant effect over the prevention of precipitation during solubilization of particles. Thorough knowledge regarding the physicochemical properties of the solid dispersed particles are very essential for understanding the relation or mechanism of wetting of particle and the release mechanism of drug. However, in this review the current researches are highlighted with their modifications over the conventional methods.

Solid dispersions: A feasible technique to improve the aqueous solubility of poorly soluble drugs

Solid dispersion technology deals with dispersing one or more active ingredients in an inert matrix in the solid state in order to achieve enhanced dissolution rate and stability. Upon increasing the dissolution rate in the gastro intestinal tract, the rate of absorption is increased as long as the dissolution rate is still the rate-limiting step. Various carriers have been used in the formation of solid dispersion, which can facilitate in improving the dissolution rate of poorly soluble drugs to improve better bioavailability.
