Characterization and validation of point mutation in Exon 19 of Calcium Channel, voltage-dependent, Alpha-2/Delta subunit 1(CACNA2D1)gene and its relationship with mastitis traits in Sahiwal

Calcium channel, voltage-dependent, alpha-2/ delta subunit 1 (CACNA2D1) gene plays an important role in excitation-contraction coupling in muscle-cells, glial cells and neurons. The CACNA2D1 gene may be one of the candidate genes related with some phenotypic traits due to its location in QTLs associated with Somatic cell score (SCS ) and mastitis. A total of 120 Sahiwal cattle were selected to characterize exon 19 of CACNA2D1 gene to identify polymorphism and its association with mastitis susceptibility/resistance. A 249 bp PCR fragment of CACNA2D1 gene encompassing the exon 19 and partial intronic region was amplified and digested with Hae III to screen the SNP which was significantly associated with SCS. The genetic background of clinical mastitis in Holstein-Friesian cattle

Mastitis is an inflammatory disease of the mammary gland, which has a significant economic impact and is an animal welfare concern. This work examined the association between single nucleotide polymorphisms (SNPs) and copy number variations (CNVs) with the incidence of clinical mastitis (CM). Using information from 16 half-sib pairs of Holstein-Friesian cows (32 animals in total) we searched for genomic regions that differed between a healthy (no incidence of CM) and a mastitis-prone (multiple incidences of CM) half-sib. Three cows with average sequence depth of coverage below 10 were excluded, which left 13 half-sib pairs available for comparisons. In total, 191 CNV regions were identified, which were deleted in a mastitis-prone cow, but present in its healthy half-sib and overlapped in at least nine half-sib pairs. Characterization of the bovine C α gene

The complete genomic sequence of a bovine Ca gene is reported here. The genomic sequence was obtained from a Ca phage clone that had been cloned from a genomic EMBL4 phage vector library. The Ca sequence had previously been expressed as a chimeric antibody and identified as IgA using IgA-specific antibodies. Intron/exon boundaries were determined by comparison of the genomic sequence with an expressed bovine Ca sequence obtained from spleen by reverse transcription-polymerase chain reaction (RT-PCR). Analysis of 50 Swedish bovine genomic DNA samples using genomic blots and five diÂerent restriction enzymes failed to detect evidence of polymorphism. However, PstI digests of Brown Swiss DNA showed a restriction fragment length polymorphism (RFLP), suggesting that at least two allelic variants of bovine IgA exist. Comparison of the deduced amino acid sequence of bovine IgA with sequences available for other species indicated that the highest homology was with that of swine, another artiodactyl. This was the highest homology observed for all mammalian IgA compared except for that between IgA1 and IgA2 in humans. Bovine IgA shares with rabbit IgA3 and IgA4, an additional N-linked glycosylation site at position 282. However, the collective data indicate that cattle are like swine and rodents and unlike rabbits in having a single locus of the gene encoding IgA of this species.

Calpastatin (CAST) is a specific inhibitor of the ubiquitous calcium-dependent proteases -l-calpain and m-calpain. This proteolytic system plays a key role in the tenderization process that occurs during postmortem storage of meat under refrigerated conditions. In the present study using comparative sequencing seven novel polymorphisms located within P3 promoter region for exon 1u of the bovine CAST gene: À357 (C/G), À556 (G/T), À557 (A/G), À580 (G/C), À750 (T/C), and two InDel at position À890 (A/À) and (GTT/À) at position À353/À351 were found. This region directs the expression of type III calpastatin mRNA, encoding the prototypical calpastatin. The genotype frequencies and haplotypes distribution were studied in 191 bulls belonging to six cattle breeds. All genotypes were distributed according to the HWE test and two major combined haplotypes were identified. The frequency of the haplotype1 varied from 0.45 in Aberdeen Angus to 0.82 in Simmental.

Genetic variations of the bovine MX1 and their association with mastitis

The primary agent of mastitis is a wide spectrum of bacterial strains; however, viral-related mastitis has also been reported. The MX dynamin-like GTPase 1 (MX1) gene has been demonstrated to confer positive antiviral responses to many viruses, and may be a suitable candidate gene for the study of disease resistance in dairy cattle. The present study was conducted to investigate the genetic diversity of the MX1 gene in Chinese cattle breeds and its effects on mastitis in Holstein cows. First, polymorphisms were identified in the complete coding region of the bovine MX1 gene in 14 Chinese cattle breeds. An association study was then carried out, utilizing polymorphisms detected in Holstein cows to determine the associations of these single nucleotide polymorphisms (SNPs) with mastitis. Database of cattle candidate genes and genetic markers for milk production and mastitis

A cattle database of candidate genes and genetic markers for milk production and mastitis has been developed to provide an integrated research tool incorporating different types of information supporting a genomic approach to study lactation, udder development and health. The database contains 943 genes and genetic markers involved in mammary gland development and function, representing candidates for further functional studies. The candidate loci were drawn on a genetic map to reveal positional overlaps. For identification of candidate loci, data from seven different research approaches were exploited: (i) gene knockouts or transgenes in mice that result in specific phenotypes associated with mammary gland (143 loci); (ii) cattle QTL for milk production (344) and mastitis related traits (71); (iii) loci with sequence variations that show specific allele-phenotype interactions associated with milk production (24) or mastitis (10) in cattle; (iv) genes with expression profiles associated with milk production (207) or mastitis (107) in cattle or mouse; (v) cattle milk protein genes that exist in different genetic variants (9); (vi) miRNAs expressed in bovine mammary gland (32) and (vii) epigenetically regulated cattle genes associated with mammary gland function (1). Fourty-four genes found by multiple independent analyses were suggested as the most promising candidates and were further in silico analysed for expression levels in lactating mammary gland, genetic variability and top biological functions in functional networks. A miRNA target search for mammary gland expressed miRNAs identified 359 putative binding sites in 3¢UTRs of candidate genes.

Novel polymorphisms of the KCNB1 gene and their association with production traits in Indian Sahiwal cattle

Mastitis in cattle is a prevalent mammary gland disease that contributes significantly to the increase in veterinary expenditures in the dairy sector. KCNB1 (Potassium voltage-gated channel, subfamily B member 1) gene is involved in regulating apoptosis, cell proliferation and differentiation, udder epithelial tissue maintenance and repair, mammary gland development and recommended as a candidate gene for production related traits in cattle. The purpose of this research was to detect the genetic variants of KCNB1 gene in Sahiwal cattle and to analyze the association between polymorphisms with milk production traits, udder traits, and teat traits in Sahiwal cattle.
