The aim of the study was to investigate the relationship between plasma levels of malondialdehyde (MDA), a routinely used marker of oxidative stress, and squamous cell carcinoma of the oral cavity and oropharynx (OSCC). The prospective cohort study comprised a total of 67 patients who underwent surgery for OSCC. MDA was assessed using high performance liquid chromatography. The MDA levels in the pooled T1-2 patients were lower than in the patients with T3-4 tumors. A negative correlation of MDA and tumor grade was shown. Seventeen patients who manifested recurrence during the 49.6 months follow-up had significantly increased MDA compared to those staying in complete remission. Kaplan-Meier analysis revealed that the median disease-free interval and overall survival in the group with MDA > median was 19.3 and 22.5 months respectively, in contrast to 31.5 and 31.6 months respectively, in patients with MDA < or = median. Background: Tobacco causes the generation of free radicals and reactive oxygen species (ROS) which are responsible for the high rate of lipid peroxidation. Malondialdehyde (MDA) is the most widely used agent to estimate the extent of lipid peroxidation. Timely diagnosis of the condition followed by supplementation with antioxidants like beta-carotene, pro-vitamin A, vitamin A, vitamin C, vitamin E, lipoic acid, zinc, selenium, and spirulina can prevent potentially malignant disorders. Materials and Methods: In this study, serum MDA was measured according to the method of Buege, in 15 normal samples and 15 patients who were histopathologically diagnosed with potentially malignant disordered and they were prescribed with antioxidants for a period of 4 week-time following which potentially malignant patients serum MDA was analyzed again to determine the extent of peroxidation reactions. Results: The mean serum MDA level in Group C1 was 0.7900 ± 0.2336 µM/L were as the mean serum MDA level of Group S1 was 2.478 ± 0.50756 µM/L and the values between them were highly significant. The values between C1 and S2 were found to be statistically significant. The mean serum MDA of S2 was 2.160 ± 0.41252 µM/L and the values were significant when compared to S1. Conclusion: Serum MDA estimation in oral pre-cancer would serve in determining the extent of lipid peroxidation. Diagnosis of patients and administration of antioxidants has proven to be effective in declining the ROS and thus reducing the extent of damage on the cells. MDA may serve as a diagnostic tool in the estimation of oral pre-cancer and in evaluation of post-treated cases.

Many researchers have shown that lipid peroxides (Malondialdehyde) are the by-products of the chemical damage done by-oxygen free radicals (ROS), to the lipid components of cell membrane, which showed changes in its serum values in radiation treated malignant cases and antioxidants mediate anticancer activity, by modifying the cellular metabolic balance. Present study was conducted with the aim to estimate and compare the serum malondialdehyde (MDA) level in normal individuals, oral cancer patients before and after radiotherapy undergoing antioxidant therapy.

OBJECTIVES The malondialdehyde (MDA) level and TA count represent the progression of oral potentially malignant disorders (OPMD) to malignancy and thus may be used as an indicator of oral epithelial dysplasia (OED). This study aimed to determine the MDA level and tissue apoptosis (TA) count in oropharyngeal tissue of Wistar rats exposed to sidestream cigarette smoke. Objective. The present systematic review and meta-analysis aimed to assess the oxidative stress-mediated lipid peroxidation end product malondialdehyde (MDA) in periodontitis using the available literature. Materials and Methods. An electronic literature search was performed for the published articles from 2000 to 2022 in PubMed (MeSH), Science Direct, Wiley Online library, and cross-reference using specific keywords. Results. The literature search identified 1,166 articles. After analyzing the abstracts of the obtained articles, the articles were excluded for the following reasons: duplicate studies (n = 395) and not relevant to the research question (n = 726). The remaining 45 articles were chosen for full-text evaluation. Finally, the present qualitative synthesis selected 34 articles that met the inclusion criteria for evaluation and removed the articles which did not meet the required criteria (n = 11). Malondialdehyde levels in serum of patients with skin cancer

The relationship of free radicals with some types of carcinomas is known, these studies have been performed on samples taken from the tumor tissue. In this study, the malondialdehyde levels in the serum of patients with skin cancer were studied and compared with a control group. The differences between malondialdehyde levels in these two groups were evaluated using Mann Whitney-U. There was a very statistically significant increase in malondialdehyde levels in serum of the skin cancer patients compared with the control group (p=0.0095, z=2.59). Thus, increased serum malondialdehyde levels may be a sign of carcinoma.
Status of salivary lipid peroxidation in oral cancer and precancer

Lipid oxidation gives rise to number of secondary by-products. Malondialdehyde (MDA) is the principal and most widely studied product of polyunsaturated fatty acid peroxidation. This aldehyde is a highly toxic molecule and should be considered as more than just a marker of lipid peroxidation in oral carcinogenesis. Salivary malondiadldehyde was evaluated in 65 healthy controls (HC), 115 subjects with oral, potentially malignant disorders (PMD) and 50 subjects with oral squamous cell carcinoma (OSCC) using the Thiobarbituric-Trichloroacetitic acid (TBA-TCA) method. A consistent elevation in the levels of salivary MDA was observed in HC with tobacco related habits, subjects with PMD and subjects with OSCC. The elevation in the salivary MDA was significant (P = 0.001) in the groups PMD and OSCC and group OSCC when compared to HC. Abstract: Background and Objective: Oral cancer is one of the 10 most common cancers in the world and its global outbreak is increasing. Saliva assessment is one of the non-invasive diagnostic methods for this cancer. The aim of the study was to achieve a comprehensive conclusion about changes in salivary antioxidants in patients with squamous cell carcinoma of the mouth (OSCC). Materials and Methods: In the present study, as a systematic review and meta-analysis study, information extracted by searching in external databases (PubMed, Science Direct, Scopus, Web of Science, google scholar) and internal databases (IranMedex, Scientific Information Database, Magiran) between the years 1980 to 2020. Comprehensive meta-analysis version 2 software was used to analyze the data (P <0.05). Results: Of 497 articles, 271 duplicate articles were deleted, and then, after reviewing the titles and abstracts of the articles, another 197 articles were removed. The full text of 26 articles were reviewed and evaluated. Finally, based on the STROB checklist, the data of 10 articles were entered into the meta-analysis. Salivary malondialdehyde (MDA) index levels were much higher in patients with OSCC than in healthy people (P=0.0001). All salivary antioxidant indexes (TAC) (P=0.003) and GSH glutathione (P=0.008) saliva was significantly lower in patients than in the healthy group and regarding the salivary superoxide dismutase (SOD) index was no significant. Conclusion: In general, antioxidant supplements may be able to help prevent, treat, and improve the prognosis of this disease, which requires further research in this area. Malondialdehyde Levels in Patients with Chronic Periodontitis

Periodontitis is a chronic condition leading to the destruction of the periodontium. A case control study was carried out in 30 subjects with chronic periodontitis aged 30-65 years (group II) and age matched with 30 control subjects (group I). Salivary and serum malondialdehyde, which is a marker of lipid peroxidation, was estimated in the cases and controls. Salivary MDA was elevated (p<0.001) in patients with chronic periodontitis there was no change in serum MDA levels when compared with normal controls. Increased levels in MDA may be closely associated with periodontal disease and salivary estimation may provide advantage in pathogenesis of the periodontal disease.
