Effects of the methanolic extracts of Zizyphus spina christi, Olea europaea and Morus alba leaves in Streptozotocin- induced diabetic rats

Background:The present study aims to investigate the hypoglycemic, hypolipidimic and antioxidant effect of the methanolic crude extracts of Zizyphus spina christi, Morus alba and Olea europaea leaves, individually or in combination against diabetes induced rats by Streptozotocin (STZ). Results:Hyperglycemia and hyperlipidaemia except in high density lipoproteins (HDL) were observed in serum after 5 weeks of STZ administration. This was associated with a depression in hepatic glutathione (GSH) concentration as well as hepatic catalase (CAT), glutathione-stransferase (GST) and superoxide dismutase (SOD) activates. In addition hepatic thiobarbituric acid-reactive substance (TBARS) and protein carbonyl (PC) were significantly elevated, indicating increased lipid and protein oxidation and oxidative stress. Depression in blood hemoglobin (Hb) content, serum insulin levels, total antioxidant capacity (TAOC) and nitric oxide (NO) levels as well as body weight gain were also observed in diabetic rats. Administration of 100mg/kg alcoholic extracts of Zizyphus spina christi, Morus alba and Olea europaea leaves 3 days before and after STZ injection daily for 5 weeks significantly ameliorated the oxidative stress evidenced by lowering TBARS & PC as well as increasing hepatic GSH concentration and CAT, GST and SOD activates as compared with STZ treated rats. These effects were paralleled with marked protection against STZ induced hyperglycemia and disturbance of lipid profile. They also caused a great improvement in insulin levels, TAOC, NO, Hb content and body weight gain. Conclussion:Thus, these results showed that the administration of the crude extracts of either Zizyphus spina christi, Morus alba or Olea europaea leaves individually or in combination might improve the clinical manifestation of diabetes and decrease the oxidative stress, this study supports the beneficial effects of these extracts especially Zizyphus spina christi, which showed marked amelioration and this may be attributed to the presence of saponin glycosides which have an inhibitory effect of serum glucose level in addition to enhance the cellular antioxidant defense. This activity contributes to the protection against oxidative damage in STZ induced diabetes.

A Review on Zizyphus as Antidiabetic

The present review paper aims to highlight the effects of extracts of genus zizyphus for the treatment of Diabetes clinically throughout the world. The basic aim is to study the hyperglycemic effect of this genus and its use. So as the effect of herbal medicine is more friendly then synthetic medicines. The effects of three species Ziziphus spinacristi, Zizyphus jojoba, and Zizyphus vulgaris have been studied clinically and have great effect on insulin secretion. While work on Zizyphus maritiana have also been done. It is concluded that genus zizyphus have potential of antidiabetic activity. Many species of Zizyphus grow widely. As natural drugs are with no or less side effects and safe for human health so they can greatly replace synthetic drugs in the forth coming future.

Antidiabetic activity and stability study of the formulated leaf extract of Zizyphus spina-christi (L.) Willd with the influence of seasonal variation

Aim of the study: The present study aimed to evaluate the anti-diabetic activity of Zizyphus spina-christi leaf extract (200 mg/kg b.w.), plain and formulated in STZ-diabetic rats. Percentage yield of extracts, marker yield (christinin-A) and antihyperglycemic potencies, depending on seasonal variation were investigated. The chemical stability, study of storage conditions, shelf life T90 prediction of both plain extract and formulated soft gelatin capsules by accelerated studies were studied. Material and methods: Changes in all studied parameters after oral administration of Z. spina-christi extract for 28 days were reported. Seasonal variation affecting yield and activities was studied. Flavonoid contents were HPLC evaluated. The capsules were stored at 30, 40 and 50 • C [75% relative humidity] and their residual christinin-A content was assayed for 24 weeks. Christinin-A chemical degradation was monitored by HPLC stability indicating method previously validated. Possible physical examination was checked by dissolution test of the content of the capsules using dissolution tester USP XXIV. Result: Oral administration of Z. spina-christi leaf extract, plain and formulated for 28 days reduced blood glucose level with significant increase in serum insulin and C-peptide levels. Marked elevation in total antioxidant capacity with normalization of percentage of glycated hemoglobin (HbA1C%) was reported. Moreover, they succeeded to reduce the elevated blood lactate level and to elevate the reduced blood pyruvate content of diabetic rats. In line with amelioration of the diabetic state, Zizyphus extract, plain and formulated restored liver and muscle glycogen content together with significant decrease of hepatic glucose-6-phosphatase and increase in glucose-6-phosphate dehydrogenase activities. In vitro experiments showed a dose-dependent inhibitory activity of Zizyphus extract against ␣-amylase enzyme with (IC 50 ) at 0.3 mg/ml. Such finding has been supported by the in vivo suppression of starch digestion and absorption by Zizyphus extract in normal rats.

Antidiabetic and Antioxidant Activities of Zizyphus lotus L Aqueous Extracts in Wistar Rats

Objectives: Zizyphus lotus L. is a medicinal plant, used in traditional medicine for its multiple therapeutic properties. The aim of the present study was to determine the antidiabetic and antioxidant effects of aqueous extracts of different parts of Zizyphus lotus in diabetic Wistar rats. Research methods and procedures: The male Wistar rats were rendered diabetic by intraperitoneal (i.p.) injection of streptozotocin (40 mg/kg body weight). Aqueous extracts from different parts, i.e, root, leaf, and seed of Zizyphus lotus L. were prepared and administrated orally to the animals at the dose of 300 mg/kg for 21 days. Blood glucose level was measured, and antioxidant status was assessed by determining oxygen radical absorbance capacity (ORAC) and the activities of enzymes like catalase, glutathione reductase and peroxidase in pancreas, liver, and erythrocytes. Vitamin C levels were determined by precipitating with 10% trichloroacetic acid. Vitamins A and E concentrations were measured by HPLC. Results: The leaf and root, but not seed, extracts exerted the glucose lowering effect on 21 st day of postadministration. The leaf and root extracts corrected antioxidant status of diabetic animals in pancreas, liver and erythrocytes. The concentrations of different vitamins (vitamin A, C and E) in diabetic rats were also modulated by leaf and root, but not seed, extracts. Conclusion(s): Our study shows that oral administration of Zizyphus lotus L. extracts from roots and leaves exerted antidiabetic and antioxidant effects in diabetic rats. Z. lotus L. seems to be a good candidate to lower, in addition to conventional antidiabetic drugs, the hyperglycaemia in diabetic subjects.

Antioxidant, hypoglycemic and anti-diabetic activities of Ziziphus spina-christi (L) Willd (Rhamnacae) leaf extract

Purpose: To investigate the antioxidant, hypoglycemic and antidiabetic activities as well as the phenolic composition of Ziziphus spina-christi (ZSC) Methods: Eighty percent methanol extract (ZSC-1), as well as ethyl acetate (ZSC-2), n-butanol (ZSC-3) and aqueous (ZSC-4) fractions were administered orally to mice at doses of 250 and 500 mg/kg of body weight. The hypoglycemic and anti-diabetic activities of these fractions in the treated mice were evaluated after 7 and 15 days, using glibenclamide as a standard. Antioxidant activities in vitro were determined using stable free radical DPPH and ABTS radical scavenging techniques. Results: The strongest (p < 0.001) anti-diabetic activity (25.59 and 39.48 % after 7 and 15 days, respectively) was found following treatment with 500 mg/kg ZSC-3 fraction. Similarly, the highest (p > 0.001) hypoglycemic effect was achieved with 500 mg/kg ZSC-3 treatment (29.07 and 35.56 % after 7 and 15 days, respectively). ZSC-1 possessed the highest content of total flavonoids (36.2 mg/g) and total polyphenol (82.3 mg/g). Conclusion: Fraction ZSC-3 displayed potential hypoglycemic activity while ZSC-1, ZSC-2 and ZSC-3 possess remarkable DPPH scavenging ability equivalent to 89, 96 and 80.3 % of the activity of the standard drug respectively.

EVALUATION OF HYPOGLYCAEMIC, HYPOLIPIDAEMIC AND NON TOXIC EFFECT OF HYDRO-METHANOLIC EXTRACTS OF ZIZIPHUS MAURITIANA, ZIZIPHUS SPINA CHRISTI FRUIT AND GLIBENCLAMIDE ON ALLOXAN INDUCED DIABETIC RATS

The use of plant products in the management of diabetes has gained ground in pharmacotherapy. It becomes imperative to evaluate the antidiabetic effects of fruit extracts of Ziziphus mauritiana (HMZM), Ziziphus spina christi (HMZS) and Glibenclamide on blood glucose, total protein, albumin and lipid profile in alloxan induced diabetic rats. 68 albino rats weighing 70-130g were used in this study. 26 rats were used for acute toxicities of Ziziphus mauritiana and Ziziphus spina christi. 42 rats of 7 groups of 6 rats each were used to test antidiabetic effects of Ziziphus mauritiana and Ziziphus spina christi plant extracts. Group 1 served as the negative control, groups 2-7 were intraperitoneally administered 360mg/kg of alloxan in normal saline respectively. Group 2 served as positive control, groups 3 and 4; 5 and 6 were respectively administered daily dose of 200 and 400mg/kg of HMZM and HMZS, group 7 was administered 0.21mg/kg of glibenclamide. Results showed Ziziphus mauritiana and Ziziphus spina christi non toxic at dose of 5000mg/kg. 48 hours after alloxan administration, blood glucose levels were found to be significantly higher (P<0.05) in groups 2-7 compared to group 1, thus confirming induction of diabetes. In groups 3-7, on 7 th and 14 th days of administration of extracts and Glibenclamide, there was a significantly lower (p<0.05) serum glucose, triacylglycerol, High density Lipoprotein, Low density lipoprotein and significantly higher (P > 0.05) serum level of total protein and albumin compared to group 2. The results obtained showed a significantly lower (P < 0.05) serum level of glucose. The effects of HMZM and HMZS fruit extracts on blood glucose, total protein, albumin and lipid profile was dose dependent. Conclusively, this study has demonstrated the antidiabetic effects of HMZM and HMZS with HMZS having a more pronounced effect on Blood glucose and lipid profile. Anti-hyperglycaemic and selected organ protective effects of Ziziphus spina-christi hydroethanol leaf extract in alloxan induced diabetic rats

Ziziphus spina-christi has been traditionally used for the treatment of stomach pain, wounds, and hair loss, skin infections, diarrhoea, scorpion stings, and malaria. This study aims to investigate the anti-hyperglycaemic, anti-dyslipidaemic, liver and kidney protective effects of Ziziphus spina-christi ethanol leaf extract on diabetic rats. The experimental design comprises of five groups of albinos Wistar rats, with three rats per group. Hypoglycemic and Hypolipidemic Effects of Zizyphus jujuba Lam. In Streptozotocin-Induced Diabetic Rats

Zizyphus jujuba Lam. Is also called as Baer tree, belongs to the family Rhamnaceae. The dried bark was powdered and extracted with various solvents by successive soxhlet hot extraction process with increasing order of polarity. Antihyperglycemic and antihyperlipidemic effects of Ziziphus vulgaris L. onreptozocin-induced diabetic adult male Wistar rats

Herbal medicine and medical plants such as Ziziphus vulgaris L. are widely used for the treatment of diseases such as diabetes mellitus. We investigated the effects of water extracts of Ziziphus vulgaris L. fruit on serum glucose, triglycerides, LDL-cholesterol, HDLcholesterol and activities of aminotransferase enzymes inreptozocin-induced diabetic adult male rats. Continuous supplementation of this water extract by gavage at doses of 0.25, 0.5, 1, 1.5 and 2 g/kg in 0.5 ml distilled water in diabetic rats resulted in a significant decrease of fasting blood glucose and LDL-cholesterol and triglyceride levels after 14 days. The levels of HDL-cholesterol and insulin, and activities of serum aminotransaminase enzymes, alanine aminotransferase and aspartate aminotransferase were not changed significantly in the extract-supplemented group compared to the control group.

Phytochemical Constituents and In Vitro Anti-Diabetic Properties of Ziziphus jujubа (Rhamnaceae) Fruits

In the current study the phytochemical profile of 70% ethanol extract of Ziziphus jujubа (Rhamnaceae), cultivated in Bulgaria has been investigated. The fruit extract contained numerous phytochemicals, such as triterpenes, phenolic acids and flavonoids. Five triterpenes, 11 phenolic acids and 5 flavonoids have been identified. The fruit extract had a total phenolic content of 21.62 ± 0.0265 mg/g and total flavonoid content 1.34 ± 0.017 mg/g dried extract. Among the triterpenes with the highest concentration was the betulinic acid 20943.17±527.06 µg/g dried extract, rosmarinic acid (1174.26±29.55 µg/g) among the phenolic acids, followed by myricetin (214.61±5.40 µg/g) as a representative of flavonoids and rutin 3 046.89±76.68 among the quercetin glycosides.
